    ASSETS AND CONSTRAINTS RELATING TO THE LOCATION DECISIONS OF SMALL MANUFACTURING BUSINESSES IN VERMONT

    The goal of this research is to identify the assets and constraints that exist specific to small business manufacturers in Vermont. To satisfy this goal, the study examines factors that influence location decisions as well as identifying what obstacles business owners have experienced. The idea for this project originated in response to the troubled economic condition of several Vermont communities, where unemployment rates are unusually high and income unusually low. Understanding what obstacles business faces might enable future ideas on how to solve these problems. Once assets are identified, they can be capitalized on, leading to more successful business operations. Preliminary data was collected through a telephone interview survey with Vermont small business owners. The data was analyzed in order to discover incentives and obstacles that existed for Vermont manufacturers as a whole, as well as in specific industries. Results from the survey suggested that Vermont's largest asset is that it offers an excellent quality of life. However, results alluded to several difficulties that owners are faced with, particularly complying with certain rules and regulations, obtaining adequate finances, a low-skilled workforce, and a weak communication network for small businesses.community development, small business, Community/Rural/Urban Development,

    The Dynamical Distinction between Elliptical and Lenticular Galaxies in Distant Clusters: Further Evidence for the Recent Origin of S0 Galaxies

    We examine resolved spectroscopic data obtained with the Keck II telescope for 44 spheroidal galaxies in the fields of two rich clusters, Cl0024+16 (z=0.40) and MS0451-03 (z=0.54), and contrast this with similar data for 23 galaxies within the redshift interval 0.3<z<0.65 in the GOODS northern field. For each galaxy we examine the case for systemic rotation, derive central stellar velocity dispersions sigma and photometric ellipticities, epsilon. Using morphological classifications obtained via Hubble Space Telescope imaging as the basis, we explore the utility of our kinematic quantities in distinguishing between pressure-supported ellipticals and rotationally-supported lenticulars (S0s). We demonstrate the reliability of using the v/(1-epsilon) vs sigma and v/sigma vs epsilon distributions as discriminators, finding that the two criteria correctly identify 63%+-3% and 80%+-2% of S0s at z~0.5, respectively, along with 76%+8-3% and 79%+-2% of ellipticals. We test these diagnostics using equivalent local data in the Coma cluster, and find that the diagnostics are similarly accurate at z=0. Our measured accuracies are comparable to the accuracy of visual classification of morphologies, but avoid the band-shifting and surface brightness effects that hinder visual classification at high redshifts. As an example application of our kinematic discriminators, we then examine the morphology-density relation for elliptical and S0 galaxies separately at z~0.5. We confirm, from kinematic data alone, the recent growth of rotationally-supported spheroidals. We discuss the feasibility of extending the method to a more comprehensive study of cluster and field galaxies to z~1, in order to verify in detail the recent density-dependent growth of S0 galaxies.Comment: 7 pages, 4 figures, updated with version accepted to Ap

    Neutral, water-soluble poly(ester amide) hydrogels for cell encapsulation

    Ā© 2020 Elsevier Ltd Hydrogels are of significant interest for cell encapsulation and delivery in regenerative medicine. Poly(ester amide)s (PEAs) are a class of biodegradable polymers that exhibit promise for biomedical applications due to the degradability of the ester and amide linkages in their backbones, their preparation from biomolecules such as amino acids, and the ability to readily tune their properties through a modular synthesis approach. Water-soluble PEAs containing cationic arginine moieties have previously been developed, but to the best of our knowledge, neutral water-soluble PEAs based on non-charged amino acids have not been reported. Using a poly(ethylene glycol) (PEG)-based macromonomer, we describe here the syntheses of water-soluble amino acid-containing PEAs containing crosslinkable alkenes in their backbones. These PEAs were converted into hydrogels through photoinitiated crosslinking and their properties were compared, including gel content, water content, swelling, and Young\u27s moduli. Subsequent cell culture studies on a subset of hydrogels confirmed that human adipose-derived stromal cells (ASCs) showed \u3e 75% viability at 24 h post-encapsulation. To explore the potential of the hydrogels as cell delivery systems for applications in soft tissue regeneration, adipogenic differentiation of the encapsulated ASCs was probed in vitro at 7 days. Analysis of glycerol-3-phosphate dehydrogenase (GPDH) enzyme activity and intracellular lipid accumulation indicated that the hydrogels provided a supportive environment for ASC adipogenesis. Overall, these PEAs provide a new platform that warrants further development for regenerative medicine applications

    Zonal Soil Type Determines Soil Microbial Responses to Maize Cropping and Fertilization.

    Soil types heavily influence ecological dynamics. It remains controversial to what extent soil types shape microbial responses to land management changes, largely due to lack of in-depth comparison across various soil types. Here, we collected samples from three major zonal soil types spanning from cold temperate to subtropical climate zones. We examined bacterial and fungal community structures, as well as microbial functional genes. Different soil types had distinct microbial biomass levels and community compositions. FiveĀ years of maize cropping (growing corn or maize) changed the bacterial community composition of the Ultisol soil type and the fungal composition of the Mollisol soil type but had little effect on the microbial composition of the Inceptisol soil type. Meanwhile, 5Ā years of fertilization resulted in soil acidification. Microbial compositions of the Mollisol and Ultisol, but not the Inceptisol, were changed and correlated (P &lt; 0.05) with soil pH. These results demonstrated the critical role of soil type in determining microbial responses to land management changes. We also found that soil nitrification potentials correlated with the total abundance of nitrifiers and that soil heterotrophic respiration correlated with the total abundance of carbon degradation genes, suggesting that changes in microbial community structure had altered ecosystem processes. IMPORTANCE Microbial communities are essential drivers of soil functional processes such as nitrification and heterotrophic respiration. Although there is initial evidence revealing the importance of soil type in shaping microbial communities, there has been no in-depth, comprehensive survey to robustly establish it as a major determinant of microbial community composition, functional gene structure, or ecosystem functioning. We examined bacterial and fungal community structures using Illumina sequencing, microbial functional genes using GeoChip, microbial biomass using phospholipid fatty acid analysis, as well as functional processes of soil nitrification potential and CO2 efflux. We demonstrated the critical role of soil type in determining microbial responses to land use changes at the continental level. Our findings underscore the inherent difficulty in generalizing ecosystem responses across landscapes and suggest that assessments of community feedback must take soil types into consideration.     Circadian rhythms in the pineal organ persist in zebrafish larvae that lack ventral brain

    <p>Abstract</p> <p>Background</p> <p>The mammalian suprachiasmatic nucleus (SCN), located in the ventral hypothalamus, is a major regulator of circadian rhythms in mammals and birds. However, the role of the SCN in lower vertebrates remains poorly understood. Zebrafish <it>cyclops </it>(<it>cyc</it>) mutants lack ventral brain, including the region that gives rise to the SCN. We have used <it>cyc </it>embryos to define the function of the zebrafish SCN in regulating circadian rhythms in the developing pineal organ. The pineal organ is the major source of the circadian hormone melatonin, which regulates rhythms such as daily rest/activity cycles. Mammalian pineal rhythms are controlled almost exclusively by the SCN. In zebrafish and many other lower vertebrates, the pineal has an endogenous clock that is responsible in part for cyclic melatonin biosynthesis and gene expression.</p> <p>Results</p> <p>We find that pineal rhythms are present in <it>cyc </it>mutants despite the absence of an SCN. The arginine vasopressin-like protein (Avpl, formerly called Vasotocin) is a peptide hormone expressed in and around the SCN. We find <it>avpl </it>mRNA is absent in <it>cyc </it>mutants, supporting previous work suggesting the SCN is missing. In contrast, expression of the putative circadian clock genes, <it>cryptochrome 1b (cry1b) </it>and <it>cryptochrome 3 (cry3)</it>, in the brain of the developing fish is unaltered. Expression of two pineal rhythmic genes, <it>exo-rhodopsin </it>(<it>exorh) </it>and <it>serotonin-N-acetyltransferase </it>(<it>aanat2</it>), involved in photoreception and melatonin synthesis, respectively, is also similar between <it>cyc </it>embryos and their wildtype (WT) siblings. The timing of the peaks and troughs of expression are the same, although the amplitude of expression is slightly decreased in the mutants. Cyclic gene expression persists for two days in <it>cyc </it>embryos transferred to constant light or constant dark, suggesting a circadian clock is driving the rhythms. However, the amplitude of rhythms in <it>cyc </it>mutants kept in constant conditions decreased more quickly than in their WT siblings.</p> <p>Conclusion</p> <p>Our data suggests that circadian rhythms can be initiated and maintained in the absence of SCN and other tissues in the ventral brain. However, the SCN may have a role in regulating the amplitude of rhythms when environmental cues are absent. This provides some of the first evidence that the SCN of teleosts is not essential for establishing circadian rhythms during development. Several SCN-independent circadian rhythms have also been found in mammalian species. Thus, zebrafish may serve as a model system for understanding how vertebrate embryos coordinate rhythms that are controlled by different circadian clocks.</p

    Treatment with an Angiopoietin-1 mimetic peptide promotes neurological recovery after stroke in diabetic rats

    AIM: Vasculotide (VT), an angiopoietin-1 mimetic peptide, exerts neuroprotective effects in type one diabetic (T1DM) rats subjected to ischemic stroke. In this study, we investigated whether delayed VT treatment improves long-term neurological outcome after stroke in T1DM rats. METHODS: Male Wistar rats were induced with T1DM, subjected to middle cerebral artery occlusion (MCAo) model of stroke, and treated with PBS (control), 2 Āµg/kg VT, 3 Āµg/kg VT, or 5.5 Āµg/kg VT. VT treatment was initiated at 24 h after stroke and administered daily (i.p) for 14 days. We evaluated neurological function, lesion volume, vascular and white matter remodeling, and inflammation in the ischemic brain. In vitro, we evaluated the effects of VT on endothelial cell capillary tube formation and inflammatory responses of primary cortical neurons (PCN) and macrophages. RESULTS: Treatment of T1DM-stroke with 3 Āµg/kg VT but not 2 Āµg/kg or 5.5 Āµg/kg significantly improves neurological function and decreases infarct volume and cell death compared to control T1DM-stroke rats. Thus, 3 Āµg/kg VT dose was employed in all subsequent in vivo analysis. VT treatment significantly increases axon and myelin density, decreases demyelination, decreases white matter injury, increases number of oligodendrocytes, and increases vascular density in the ischemic border zone of T1DM stroke rats. VT treatment significantly decreases MMP9 expression and decreases the number of M1 macrophages in the ischemic brain of T1DM-stroke rats. In vitro, VT treatment significantly decreases endothelial cell death and decreases MCP-1, endothelin-1, and VEGF expression under high glucose (HG) and ischemic conditions and significantly increases capillary tube formation under HG conditions when compared to non-treated control group. VT treatment significantly decreases inflammatory factor expression such as MMP9 and MCP-1 in macrophages subjected to LPS activation and significantly decreases IL-1Ī² and MMP9 expression in PCN subjected to ischemia under HG conditions. CONCLUSION: Delayed VT treatment (24 h after stroke) significantly improves neurological function, promotes vascular and white matter remodeling, and decreases inflammation in the ischemic brain after stroke in T1DM rats

    Metrics of the Gynecologic Oncology Literature Focused on Cited Utilization and Costs

    OBJECTIVE: The newest findings on literature utilization relevant to gynecologic oncology were published by Thomson Reuters during June 2013 as determinants of journal standing. Our objective was to assess the different metrics reported for relative impact and cost for journals relevant to gynecologic oncology. METHODS: 55 journals were evaluated for Impact Factor (IF), 5Year IF, Immediacy Index, Cited Half Life, Eigenfactor (EF) Score, Article Influence (AI) scores and subscription costs obtained from publisher information. RESULTS: CA-A Cancer Journal for Clinicians had the highest IF (101.78) & AI (24.502). The top EF cancer-specific journals were the Journal of Clinical Oncology, Cancer Research, Clinical Cancer Research and Oncogene. Rankings for Gynecologic Oncology (409 articles, 18,243 citations) were IF=3.929, 43/55, EF=0.038, 28/55, and AI=1.099, 44/55, all higher than the previous year. The IF improved from the 5year IF in 31 journals, including Gynecologic Oncology, 29/31. Subscription costs for Gynecologic Oncology compared favorably to other journals. CONCLUSIONS: The high utilization of review information in CA-A Cancer Journal for Clinicians and Nature Review Cancer illustrated by the IF coupled with a relatively low number of articles and short cited half life indicates that they serve as a leading source of quoted cancer statistics (CA-A Cancer Journal for Clinicians). Rankings for Gynecologic Oncology and the International Journal of Gynecologic Cancer have improved. Regardless of specialty size, the Impact Factor for Gynecologic Oncology is respectably strong. The decreased IF in 44% of the journals may reflect the international economy\u27s effect on cancer research

    Contemporary Childrenā€™s Literature in Education Courses: Diverse, Complex, and Critical While research has established the importance of childrenā€™s literature in classrooms, the inclusion of childrenā€™s literature courses in teacher preparation programs increasingly faces resistance. A team of childrenā€™s literature and literacy education scholars conducted a national survey of childrenā€™s literature courses in P-8 teacher certification programs to better understand the role these courses currently serve. Survey questions centered on course types, formats, descriptions, goals/objectives, readings, and assignments. Initial analysis focused on a comparison of undergraduate and dual-degree programs, and further analysis of a subset of undergraduate courses involved cross-data comparison between course descriptions, course objectives/goals, and course assignment descriptions. Cross-program analyses largely revealed many commonalities between undergraduate and dual-degree programs. In-depth cross-data comparison resulted in two key findings: (1) significant attention to diversity in courses, and (2) striking complexity and volume of assignments requiring students to understand, analyze, and integrate quality childrenā€™s literature into classrooms. Overall, the survey results offer a compelling argument for the centrality of childrenā€™s literature courses, particularly those that focus on cultural diversity, in teacher education programs
