    Extraosseous (extramedullary) plasmacytomas: a clinicopathologic and immunophenotypic study of 32 Chinese cases

    <p>Abstract</p> <p>Background</p> <p>Extraosseous plasmacytoma, so called extramedullary plasmacytoma (EMP) is relatively rare in China. The aim was investigate the clinicopathologic features of EMP and the role of Immunophenotype and genotype detection in diagnosis of EMP.</p> <p>Methods</p> <p>Thirty-two cases of EMP were investigated retrospectively by histopathology, immunophenotype, genotype and survival analysis.</p> <p>Results</p> <p>Clinically, the mean age of the patients was 53.4. Most of the patients received no treatment after the diagnosis was established, and the prognosis was relatively poor. Histologically, in 40% of the cases, the neoplastic cells were grade II or III. The neoplastic cells expressed one or more PC associated antigens. The immunophenotype of EMP and inflammation of sinonasal regions with numerous PC infiltrations were compared and showed some difference in expression of CD45, CD27, CD44v6 and Bcl-2 as well. Ig light chain restriction was detected in 87.5% of the cases.</p> <p>Conclusions</p> <p>we described 32 Chinese cases of EMP, compare with that reported in the literature, some differences are presented, including higher percentage of grade II and III cases, clinically inconsistent treatment and management as well as poor outcome of the disease.</p

    Survivin is a prognostic marker and therapeutic target for extranodal, nasal-type natural killer/T cell lymphoma.

    Background: The relationship between survivin and extranodal, nasal-type natural killer/T cell lymphoma (ENKTCL) was unclearly established yet. We here studied the potential prognostic roles of survivin and its implication as a target in ENKTCL therapy. Methods: ENKTCL patients\u27 peripheral blood were collected and tested by ELISA. ENKTCL cell lines were cultured with or without survivin inhibitor and tested by MTT and Flow cytometry. According to the gene expression profiles from the ArrayExpress Archive under E-TABM-702, survivin co-regulated cluster was established by Coupled Two-way Clustering Algorithm. Results: Seventeen point six percent of total 17 ENKTCL patients were serum survivin-positive. These patients had poorer outcome than that of negative cases (P Conclusions: We concluded that survivin was a potential prognostic marker and a critical regulatory molecule in the pathological process of ENKTCL. It would be a promising target in drugs discovery for ENKTCL therapy

    Different yellowing degrees and the industrial utilization of flue-cured tobacco leaves

    Yellowing is a key stage in the curing of flue-cured tobacco (Nicotiana tobacum L.) as much of the chemical transformation occurs during this period. This study examined the effect of different yellowing degrees on the value of flue-cured tobacco leaves at the farm level for both processing and manufacturing. The study was conducted in the counties of Chuxiong, Dali, and Yuxi in Yunnan, China over two years. Yellowing treatments have been designed to have either a mild or a regular yellowing degree. Yield, value, appearance, suction property, smoking characteristics, and physical resistance to further processing were investigated to evaluate the effect of degree of yellowing on the industrial utilization of flue-cured tobacco leaves. The regular yellowing degree enhanced yield, value, and appearance compared to the mild yellowing degree, regardless of cultivar or location; however, physical resistance to further processing and the suction property of the mild yellowing degree treatment were better than with the regular yellowing degree regardless of cultivar or location. Furthermore, although the regular yellowing degree recorded higher smoking characteristic scores than the mild yellowing degree immediately after flue-curing, the scores of mild yellowing degree leaves could be further augmented by increasing intensity in the re-drying stage. The smoking characteristic score in the regular yellowing degree can only be increased by low intensity re-drying, and significantly decreased by mild and high intensity re-drying. Therefore, in terms of industrial utilization, mild yellowing is the better choice for flue-curing tobacco. This study also suggested that the current regular yellowing stage in Yunnan should be shortened to meet the demands of the traditional tobacco industry

    A novel baseline-correction method for standard addition based derivative spectra and its application to quantitative analysis of benzo(a)pyrene in vegetable oil samples

    In the present work, a baseline-correction method based on peak-to-derivative baseline measurement was proposed for the elimination of complex matrix interference that was mainly caused by unknown components and/or background in the analysis of derivative spectra. This novel method was applicable particularly when the matrix interfering components showed a broad spectral band, which was common in practical analysis. The derivative baseline was established by connecting two crossing points of the spectral curves obtained with a standard addition method (SAM). The applicability and reliability of the proposed method was demonstrated through both theoretical simulation and practical application. Firstly, Gaussian bands were used to simulate 'interfering' and 'analyte' bands to investigate the effect of different parameters of interfering band on the derivative baseline. This simulation analysis verified that the accuracy of the proposed method was remarkably better than other conventional methods such as peak-to-zero, tangent, and peak-to-peak measurements. Then the above proposed baseline-correction method was applied to the determination of benzo(a) pyrene (BaP) in vegetable oil samples by second-derivative synchronous fluorescence spectroscopy. The satisfactory results were obtained by using this new method to analyze a certified reference material (coconut oil, BCR (R)-458) with a relative error of -3.2% from the certified BaP concentration.     NOTCH-induced aldehyde dehydrogenase 1A1 deacetylation promotes breast cancer stem cells

    High aldehyde dehydrogenase (ALDH) activity is a marker commonly used to isolate stem cells, particularly breast cancer stem cells (CSCs). Here, we determined that ALDH1A1 activity is inhibited by acetylation of lysine 353 (K353) and that acetyltransferase P300/CBP–associated factor (PCAF) and deacetylase sirtuin 2 (SIRT2) are responsible for regulating the acetylation state of ALDH1A1 K353. Evaluation of breast carcinoma tissues from patients revealed that cells with high ALDH1 activity have low ALDH1A1 acetylation and are capable of self-renewal. Acetylation of ALDH1A1 inhibited both the stem cell population and self-renewal properties in breast cancer. Moreover, NOTCH signaling activated ALDH1A1 through the induction of SIRT2, leading to ALDH1A1 deacetylation and enzymatic activation to promote breast CSCs. In breast cancer xenograft models, replacement of endogenous ALDH1A1 with an acetylation mimetic mutant inhibited tumorigenesis and tumor growth. Together, the results from our study reveal a function and mechanism of ALDH1A1 acetylation in regulating breast CSCs
