    Numerical Analysis of the Effect of the Pre-Existing Crack Opening on the Direction of the Secondary Crack Propagation

    This study involves an analysis of the development and direction of secondary crack propagation starting from openings of different shapes located in brittle prismatic samples subjected to either uniaxial compression or direct shear stress conditions. The findings from the study were then used to understand how notches (cracks) located at the toe of slopes made of brittle materials influence their failure. For the first part of the study, a theoretical analysis that uses closed form solutions, linear elastic fracture mechanics (LEFM) and a numerical analysis that uses a finite element software (ABAQUS) were conducted to understand how the degree of curvature of circular and elliptical openings develops critical stresses that were the locations of the failure of the openings. The theoretical and numerical analysis established that the degree of curvature of the openings had a tremendous influence on where the secondary cracks developed. For the case of the elliptical samples, the attitude of the ellipse also had an influence on how the secondary cracks extend. For the case of the rock slopes with a notch (crack) at their toe, an extended finite element method (XFEM) based on linear elastic fracture mechanics (LEFM) theory was utilized to model the secondary crack propagation that takes place from the tip of the notch subjected to gravity induced compression and gravity induced lateral pressure. The analysis that used XFEM established that the ratio between the shear stress acting parallel to the axis of the toe crack, and the vertical overburden compression normal to the axis of the notch had a marked influence on how the secondary cracks developed from the notch. If the ratio was less than one, the secondary crack developed at angles greater than 90° with respect to the crack axis. If the ratio was equal to one, the secondary crack developed exactly at 90°. If ratio was greater than one, the secondary crack was inclined at angles less than 90°. XEFM study also indicated that the opening of the notch has also an influence on the angle of secondary crack propagation from the tip of the notch

    Describing Strong Correlation with Block-Correlated Coupled Cluster Theory

    A block-correlated coupled cluster (BCCC) method based on the generalized valence bond (GVB) wave function (GVB-BCCC in short) is proposed and implemented at the ab initio level, which represents an attractive multireference electronic structure method for strongly correlated systems. The GVB-BCCC method is demonstrated to provide accurate descriptions for multiple bond breaking in small molecules, although the GVB reference function is qualitatively wrong for the studied processes. For a challenging prototype of strongly correlated systems, tridecane with all 12 single C-C bonds at various distances, our calculations have shown that the GVB-BCCC2b method can provide highly comparable results as the density matrix renormalization group method for potential energy surfaces along simultaneous dissociation of all C-C bonds

    Development of Eighteen Microsatellite Markers in Anemone amurensis (Ranunculaceae) and Cross-Amplification in Congeneric Species

    Polyploidy plays an important role in the evolution of plant genomes. To enable the investigation of the polyploidy events within the genus Anemone, we developed eighteen microsatellite markers from the hexaploid species A. amurensis (Ranunculaceae), and tested their transferability in five closely related species. The number of total alleles (NA) for each resulting locus varied from one to eight. The polymorphism information content (PIC) and Nei’s genetic diversity (NGD) for these microsatellites ranged from 0.00 to 0.71 and 0.00 to 0.91, respectively. For each population, the NA was one to seven, and the values of PIC and NGD varied from 0.00 to 0.84 and 0.00 to 0.95, respectively. In addition, most of these microsatellites can be amplified successfully in the congeneric species. These microsatellite primers provide us an opportunity to study the polyploid evolution in the genus Anemone

    Relationship between the morphology of A-1 segment of anterior cerebral artery and anterior communicating artery aneurysms

    Background: The anterior communicating artery (ACoA) is one of the most frequent sites for cerebral aneurysm. The peculiar directions of projection of aneurysms offer great challenges to clinical treatment. Objetives: To establish the relationship between morphology of A-1 segment of anterior cerebral artery (ACA) and aneurismal projection. Methods: Randomly selected digital subtraction angiography data of 264 anterior communicating artery aneurysms (ACoAA) cases and 296 cases of other cerebral vascular diseases in the same period were retrospectively analyzed. Results: Among 264 ACoAA patients, the morphology of A-1 segment showed type Ⅰa in 158 sides, type Ⅰb in 11, type Ⅱa in 35, type Ⅱb in 87, type Ⅲ in 171 and absence in 66. The morphology of A-1 segment in 296 patients with other cerebral vascular diseases displayed type Ⅰa in 195 sides, type Ⅰb in 20, type Ⅱa in 47, type Ⅱ b in 74, type Ⅲ in 217 and absence in 39. The non-visualization of A-1 segment in the group of ACoAA occurred more than in the control group (χ2=11.482, p=0.001). The classifications of ACoAAs in 264 patients were confirmed as anterior-superior type in 121 cases, anterior-inferior type in 105, complicated type in 16, posterior-inferior type in 12 and posterior-superior type in 10. The correlation between morphology of A-1 segment of ACA and classifications of ACoAA was significant (p=0.000; C=0.619, p=0.000). The direction of ACoAA was downward when the A-1 segment of ACA was Type Ⅰa or Type Ⅱa, and was upward when it was Type Ⅰb or Type Ⅱb,and was upward or downward or complicated when it was Type Ⅲ.     Co-methylated Genes in Different Adipose Depots of Pig are Associated with Metabolic, Inflammatory and Immune Processes

    It is well established that the metabolic risk factors of obesity and its comorbidities are more attributed to adipose tissue distribution rather than total adipose mass. Since emerging evidence suggests that epigenetic regulation plays an important role in the aetiology of obesity, we conducted a genome-wide methylation analysis on eight different adipose depots of three pig breeds living within comparable environments but displaying distinct fat level using methylated DNA immunoprecipitation sequencing. We aimed to investigate the systematic association between anatomical location-specific DNA methylation status of different adipose depots and obesity-related phenotypes. We show here that compared to subcutaneous adipose tissues which primarily modulate metabolic indicators, visceral adipose tissues and intermuscular adipose tissue, which are the metabolic risk factors of obesity, are primarily associated with impaired inflammatory and immune responses.     A Rapid and Sensitive Europium Nanoparticle-Based Lateral Flow Immunoassay Combined with Recombinase Polymerase Amplification for Simultaneous Detection of Three Food-Borne Pathogens

    Food-borne pathogens have become an important public threat to human health. There are many kinds of pathogenic bacteria in food consumed daily. A rapid and sensitive testing method for multiple food-borne pathogens is essential. Europium nanoparticles (EuNPs) are used as fluorescent probes in lateral flow immunoassays (LFIAs) to improve sensitivity. Here, recombinase polymerase amplification (RPA) combined with fluorescent LFIA was established for the simultaneous and quantitative detection of Listeria monocytogenes, Vibrio parahaemolyticus, and Escherichia coli O157:H7. In this work, the entire experimental process could be completed in 20 min at 37 degrees C. The limits of detection (LODs) of EuNP-based LFIA-RPA were 9.0 colony-forming units (CFU)/mL for Listeria monocytogenes, 7.0 CFU/mL for Vibrio parahaemolyticus, and 4.0 CFU/mL for Escherichia coli O157:H7. No cross-reaction could be observed in 22 bacterial strains. The fluorescent LFIA-RPA assay exhibits high sensitivity and good specificity. Moreover, the average recovery of the three food-borne pathogens spiked in food samples was 90.9-114.2%. The experiments indicate the accuracy and reliability of the multiple fluorescent test strips. Our developed EuNP-based LFIA-RPA assay is a promising analytical tool for the rapid and simultaneous detection of multiple low concentrations of food-borne pathogens

    MicroRNAs miR-27a and miR-143 Regulate Porcine Adipocyte Lipid Metabolism

    MicroRNAs (miRNAs) are non-coding small RNAs that play roles in regulating gene expression. Some miRNAs have been classed as epigenetic regulators of metabolism and energy homeostasis. Previous reports indicated that the miRNAs miR-27a and miR-143 were involved in lipid metabolism in human and rodents. To determine the roles of porcine miR-27a and miR-143 in adipocyte lipid metabolism, porcine adipocytes were cultured and allowed to induce differentiation for 10 days. The lipid-filled adipocytes were then transfected with miRNA mimics and inhibitors. We measured how the indicators of adipogenesis and adipolysis in porcine adipocytes were affected by the over-expression and by the inhibition of both miR-27a and miR-143. The results indicated that the over-expression of miR-27a could accelerate adipolysis releasing significantly more glycerol and free fatty acids than the negative control (P < 0.001), while the mimic of miR-143 expression, promoted adipogenesis by accumulating more triglycerides (P < 0.001) in the adipocytes. In addition, we demonstrated that there was good correlation (r > 0.98, P < 0.001) between the indicators of adipolysis in cell lysates and in the culture medium
