    Cell surface-specific N-glycan profiling in breast cancer

    Aberrant changes in specific glycans have been shown to be associated with immunosurveillance, tumorigenesis, tumor progression and metastasis. In this study, the N-glycan profiling of membrane proteins from human breast cancer cell lines and tissues was detected using modified DNA sequencer-assisted fluorophore-assisted carbohydrate electrophoresis (DSA-FACE). The N-glycan profiles of membrane proteins were analyzed from 7 breast cancer cell lines and MCF 10A, as well as from 100 pairs of breast cancer and corresponding adjacent tissues. The results showed that, compared with the matched adjacent normal tissue samples, two biantennary N-glycans (NA2 and NA2FB) were significantly decreased (p <0.0001) in the breast cancer tissue samples, while the triantennary glycan (NA3FB) and a high-mannose glycan (M8) were dramatically increased (p = 0.001 and p <0.0001, respectively). Moreover, the alterations in these specific N-glycans occurred through the oncogenesis and progression of breast cancer. These results suggested that the modified method based on DSA-FACE is a high-throughput detection technology that is suited for analyzing cell surface N-glycans. These cell surface-specific N-glycans may be helpful in recognizing the mechanisms of tumor cell immunologic escape and could be potential targets for new breast cancer drugs

    The influence of delirium on mortality and length of ICU stay and analysis of risk factors for delirium after liver transplantation

    ObjectiveTo analyze the incidence, timing, risk factors and prognosis of delirium after liver transplantation (LT).MethodsThe clinical data of 321 patients undergoing LT in the Third Xiangya Hospital of Central South University from January 2018 to December 2022 were collected to investigate the incidence, onset, and risk factors for post-LT delirium and the impact of delirium on LT recipients’ prognosis by statistical analysis.ResultsThe incidence of post-LT delirium was 19.3% (62/321), and the median interval between LT and onset of delirium was 20.1 h. Univariate analysis showed that pre-LT variables (Model for End Stage Liver Disease (MELD) score, hospital stay, hepatic encephalopathy, infection, white blood cell (WBC) count, lymphocyte count, abnormal potassium, lactulose use), intraoperative variables (red blood cell transfusion, remimazolam use, dexmedetomidine use) and post-LT variables (hypernatraemia, acute rejection, reoperation, basiliximab use, tacrolimus concentration) were associated with post-LT delirium. Multivariate logistic regression analysis revealed that MELD score at LT ≥22 [OR = 3.400, 95% CI:1.468–7.876, p = 0.004], pre-LT hepatic encephalopathy [OR = 3.224, 95% CI:1.664–6.244, p = 0.001], infection within 2 months prior to LT [OR = 2.238, 95% CI:1.151–4.351, p = 0.018], acute rejection [OR = 2.974, 95% CI:1.322–6.690, p = 0.008], and reoperation [OR = 11.919, 95% CI:2.938–48.350, p = 0.001] were independent risk factors for post-LT delirium. Post-LT delirium was reduced in LT recipients exposing to intraoperative remimazolam [OR = 0.287, 95% CI: 0.113–0.733, p = 0.009] or ≥ 25 μg of intraoperative dexmedetomidine [OR = 0.441, 95% CI 0.225–0.867, p = 0.018]. As for clinical outcomes, patients with delirium had a higher percentage of staying at the (ICU) ≥7 d after LT than those without delirium [OR = 2.559, 95% CI 1.418–4.617, p = 0.002].ConclusionThe incidence of delirium was high and the onset of delirium was early after LT. Risk factors for post-LT delirium included high MELD score at LT, pre-LT hepatic encephalopathy and infections, acute rejection and reoperation. Intraoperative use of remimazolam or dexmedetomidine reduced post-LT delirium. Delirium had a negative impact on the length of ICU stay

    T2-GNN: Graph Neural Networks for Graphs with Incomplete Features and Structure via Teacher-Student Distillation

    Graph Neural Networks (GNNs) have been a prevailing technique for tackling various analysis tasks on graph data. A key premise for the remarkable performance of GNNs relies on complete and trustworthy initial graph descriptions (i.e., node features and graph structure), which is often not satisfied since real-world graphs are often incomplete due to various unavoidable factors. In particular, GNNs face greater challenges when both node features and graph structure are incomplete at the same time. The existing methods either focus on feature completion or structure completion. They usually rely on the matching relationship between features and structure, or employ joint learning of node representation and feature (or structure) completion in the hope of achieving mutual benefit. However, recent studies confirm that the mutual interference between features and structure leads to the degradation of GNN performance. When both features and structure are incomplete, the mismatch between features and structure caused by the missing randomness exacerbates the interference between the two, which may trigger incorrect completions that negatively affect node representation. To this end, in this paper we propose a general GNN framework based on teacher-student distillation to improve the performance of GNNs on incomplete graphs, namely T2-GNN. To avoid the interference between features and structure, we separately design feature-level and structure-level teacher models to provide targeted guidance for student model (base GNNs, such as GCN) through distillation. Then we design two personalized methods to obtain well-trained feature and structure teachers. To ensure that the knowledge of the teacher model is comprehensively and effectively distilled to the student model, we further propose a dual distillation mode to enable the student to acquire as much expert knowledge as possible.Comment: Accepted by AAAI2

    Measuring the Utility of Mobile Phone Video Service Based on A Service Value Network Model

    Service value networks (SVNs) are a new form of industrial cooperation. Within SVNs, service providers engage in loosely coupled relationships to jointly offer dynamically configured complex services to customers. Value is co-created through these joint complex services. By specialization, service providers leverage the knowledge and capital assets of their partners, and share the risk of operating in a changing and uncertain environment. However, as a new approach, SVNs lack theoretical foundations and empirical studies to validate its applicability. This paper seeks to address these problems by making three contributions. First, based on an existing theoretical SVN model, the paper presents a design for a mobile phone video service value network (MPVSVN) in China. Second, we describe a systematic approach to calculate the value and utility of services on the SVN. Third, we demonstrate our approach in detailed steps through real world service examples identified from the MPVSVN

    Effects of Pressure and Doping on Ruddlesden-Popper phases Lan+1NinO3n+1

    Recently the discovery of superconductivity with a critical temperature Tc up to 80 K in Ruddlesden-Popper phases Lan+1NinO3n+1 (n = 2) under pressure has garnered considerable attention. Up to now, the superconductivity was only observed in La3Ni2O7 single crystal grown with the optical-image floating zone furnace under oxygen pressure. It remains to be understood the effect of chemical doping on superconducting La3Ni2O7 as well as other Ruddlesden-Popper phases. Here, we systematically investigate the effect of external pressure and chemical doping on polycrystalline Ruddlesden-Popper phases. Our results demonstrate the application of pressure and doping effectively tunes the transport properties of Ruddlesden-Popper phases. We find pressure-induced superconductivity up to 86 K in La3Ni2O7 polycrystalline sample, while no signatures of superconductivity are observed in La2NiO4 and La4Ni3O10 systems under high pressure up to 50 GPa.     Molecular dynamics study on the adsorption and modification mechanism of polymeric sand-fixing agent

    Chemical sand-fixing technology has shown good potential in preventing desertification, but the effect is determined by materials. In this paper, the adsorption behavior of quartz and six common polymer sand-fixing agents under dry conditions was studied by molecular dynamics method. The results show that all polymers could be adsorbed on the surface of quartz and their functional groups play an important role in the adsorption process. Compared with other materials, the binding energy and the number of hydrogen bonds of PAA-quartz composites were improved by 30.7–65.6% and 8.3–333.3%, respectively. It was found that the number of hydrogen bonds formed under the unit molecular was positively correlated with the mechanical properties of the improved sandy soil. This study provides an accurate, efficient and inexpensive qualitative evaluation method for the curing effect of sand fixers, which will assist in the screening and development of new high performance sand fixers

    Effect of physical and chemical pressure on the superconductivity of caged-type quasiskutterudite Lu5Rh6Sn18

    Lu5Rh6Sn18 is one of the caged-type quasiskutterudite superconductors with superconducting transition temperature Tc = 4.12 K. Here, we investigate the effect of pressure on the superconductivity in Lu5Rh6Sn18 by combining high pressure electrical transport, synchrotron x-ray diffraction (XRD) and chemical doping. Application of high pressure can enhance both the metallicity and the superconducting transition temperature in Lu5Rh6Sn18. Tc is found to show a continuous increase reaching up to 5.50 K at 11.4 GPa. Our high pressure synchrotron XRD measurements demonstrate the stability of the pristine crystal structure up to 12.0 GPa. In contrast, Tc is suppressed after the substitution of La ions in Lu sites, inducing negative chemical pressure. Our study provides valuable insights into the improvement of superconductivity in caged compounds.Comment: 9 pages, 8 figure
