    Enkephalin systems in diencephalon and brainstem of the rat

    The immunocytochemical distribution of [Leu]enkephalin and an adrenal enkephalin precursor fragment (BAM-22P) immunoreactivity was investigated in the diencephalon and brainstem of rats pretreated with relatively high doses of colchicine (300–400 Μg/10 Μl intracerebroventricularly). The higher ranges of colchicine pretreatment allowed the visualization of extensive enkephalin-containing systems in these brain regions, some of which are reported for the first time. Immunoreactive perikarya were found in many hypothalamic and thalamic nuclei, interpeduncular nucleus, substan-tia nigra, the colliculi, periaqueductal gray, parabrachial nuclei, trigeminal motor and spinal nuclei, nucleus raphe magnus and other raphe nuclei, nucleus reticularis paragigantocellularis, vestibular nuclei, several nor-adrenergic cell groups, nucleus tractus solitarius, as well as in the spinal cord dorsal horn. In addition to the above regions, immunoreactive fibers were also noted in the habenular nuclei, trigeminal sensory nuclei, locus coeruleus, motor facial nucleus, cochlear nuclei, dorsal motor nucleus of the vagus, and hypoglossal nucleus. When adjacent sections to those stained for [Leu]enkephalin were processed for BAM-22P immunoreactivity, it was found that these two immunoreactivities were distributed identically at almost all anatomical locations. B AM-22P immunoreactivity was generally less pronounced and was preferentially localized to neuronal perikarya. The results of the present as well as the preceding studies (Khachaturian et ai., '83) strongly suggest substantial structural similarity between the adrenal proenkephalin precursor and that which occurs in the brain.     The search for Hesperian organic matter on Mars: Pyrolysis studies of sediments rich in sulfur and iron

    Jarosite on Mars is of significant geological and astrobiological interest as it forms in acidic aqueous conditions that are potentially habitable for acidophilic organisms. Jarosite can provide environmental context and may host organic matter. The most common analytical technique used to search for organic molecules on the surface of Mars is pyrolysis. However, thermal decomposition of jarosite produces oxygen, which degrades organic signals. At pH values greater than 3 and high water to rock ratios jarosite has a close association with goethite. Hematite can form by dehydration of goethite or directly from jarosite under certain aqueous conditions. Goethite and hematite are significantly more amenable for pyrolysis experiments searching for organic matter than jarosite. Analysis of the mineralogy and organic chemistry of samples from a natural acidic stream revealed a diverse response for organic compounds during pyrolysis of goethite-rich layers but a poor response for jarosite-rich or mixed jarosite-goethite units. Goethite units that are associated with jarosite but do not contain jarosite themselves should be targeted for organic detection pyrolysis experiments on Mars. These findings are extremely timely as future exploration targets for Mars Science Laboratory include Hematite Ridge, which may have formed from goethite precursors

    Depression across pregnancy and the postpartum, antidepressant use and the association with female sexual function

    Background: There is an established relationship between depression and sexual functioning in women. However, there is limited research examining the relationship between perinatal depression and sexual functioning. Methods: This study draws on the Mercy Pregnancy and Emotional Wellbeing Study and reports on 211 women recruited in early pregnancy and followed to 12 months postpartum. Women were assessed for depression using the Structured Clinical Interview for the DSM-IV, repeated measurement of depressive symptoms using the Edinburgh Postnatal Depression Scale and sexual functioning using the Female Sexual Functioning Inventory. Data were also collected on antidepressant use, mode of delivery, history of childhood trauma, breastfeeding and partner support. Results: Women showed a decline in sexual functioning over pregnancy and the first 6 months postpartum, which recovered by 12 months. For women with depression, sexual functioning was lower throughout pregnancy and continued to be lower at 6 months postpartum than those without depression. Ongoing depressive symptoms at 12 months were also associated with lower sexual functioning. Sexual functioning was not predicted by mode of delivery, antidepressant use or childhood trauma. Breastfeeding predicted lower sexual functioning only at 6 months. Higher partner support predicted higher female sexual functioning. Conclusions: Pregnancy and the postpartum are a time of reduced sexual functioning for women; however, women with depression are more likely to have lower levels of sexual functioning and this was not predicted by antidepressant use. In women with perinatal depression, consideration of the impact on sexual functioning should be an integral part of care

    The role of trauma and partner support in perinatal depression and parenting stress: An Australian pregnancy cohort study

    Background: Improving our understanding of the relationship between maternal depression and parenting stress is likely to lie in the range of additional factors that are associated with vulnerability to depression and also to parenting stress. Objectives: To examine the role of trauma and partner support, in understanding the relationship between perinatal depression and parenting stress. Methods: This study utilises data from 246 women in a pregnancy cohort study that followed women from early pregnancy until their infant was 12 months. Included were both women with a diagnosis of depression and those without depression. The measures included Structured Clinical Interview for the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition, Edinburgh Postnatal Depression Scale, Childhood Trauma Questionnaire, Social Support Effectiveness Questionnaire and the Parenting Stress Index. Results: We found women with depression were more likely to report a history of childhood trauma. Depressive symptoms were positively associated with parenting stress while partner support was negatively associated with parenting stress. The protective role of partner support for parenting distress was observed in those with no history of childhood abuse and low depressive symptoms, but not in those with a trauma history and high depressive symptoms. Conclusions: These findings highlight the importance of early trauma in understanding the protective role of support on the relationship between parenting and depression. These findings can inform future studies and the refinement of future interventions aimed at both perinatal depression and parenting

    The composition and friction-reducing properties of leaf layers

    Every autumn rail networks across the world suffer delays, accidents and schedule changes due to low friction problems caused by leaves landing on the rails. These leaves form a layer that can reduce the friction between the wheel and the rail to a similar level as that between ice and an ice-skate (μ=0.01–0.05). Previous works have generated several hypotheses for the chemical reactions and low friction mechanism associated with these layers. In this work, the reaction between an aqueous extract of sycamore leaves and metallic iron is investigated. This reaction has been shown to produce a black precipitate, which matches field observations of leaf layers, while friction tests with these extracts produce characteristic ultra low friction. The reaction is investigated through FTIR, XPS, CHNS and ICP-MS analysis as well as wet chemical testing. The impact of the reaction on friction is investigated through three rounds of tribological testing. The results indicate that the black precipitate produced is iron tannate, formed by complexation of tannins with dissolved iron ions. Friction testing showed that eliminating tannins from the leaf extract resulted in a significant increase in the friction coefficient compared with the control

    HAR Inference: Recommendations for Practice

    The classic papers by Newey and West (1987) and Andrews (1991) spurred a large body of work on how to improve heteroscedasticity- and autocorrelation-robust (HAR) inference in time series regression. This literature finds that using a larger-than-usual truncation parameter to estimate the long-run variance, combined with Kiefer-Vogelsang (2002, 2005) fixed-b critical values, can substantially reduce size distortions, at only a modest cost in (size-adjusted) power. Empirical practice, however, has not kept up. This article therefore draws on the post-Newey West/Andrews literature to make concrete recommendations for HAR inference. We derive truncation parameter rules that choose a point on the size-power tradeoff to minimize a loss function. If Newey-West tests are used, we recommend the truncation parameter rule S = 1.3T 1/2 and (nonstandard) fixed-b critical values. For tests of a single restriction, we find advantages to using the equal-weighted cosine (EWC) test, where the long run variance is estimated by projections onto Type II cosines, using ν = 0.4T 2/3 cosine terms; for this test, fixed-b critical values are, conveniently, tν or F. We assess these rules using first an ARMA/GARCH Monte Carlo design, then a dynamic factor model design estimated using a 207 quarterly U.S. macroeconomic time series

    Time of origin of opioid peptide-containing neurons in the rat hypothalamus

    By using a combined technique of immunocytochemistry and [ 3 H]thymidine autoradiography, we have determined the “birth date” of opioid pep-tide-containing neurons in several hypothalamic nuclei and regions. These include proopiomelanocortin (POMC) neurons (represented by ACTH immunoreactivity) in the arcuate nucleus; dynorphin A neurons in the supraoptic and paraventricular nuclei and the lateral hypothalamic area; and leuenkephalin neurons in the periventricular, ventromedial, and medial mammillary nuclei, as well as in preoptic and perifornical areas. Arcuate POMC neurons were born very early in embryonic development, with peak heavy [ 3 H]thymidine nuclear labelling occurring on embryonic day E12. Supraoptic and paraventricular dynorphin A neurons were also labelled relatively early (peak at E13). The lateral hypothalamic dynorphin A neurons showed peak heavy labelling also on day E12, By contrast, leu-enkephalin neurons in the periventricular nucleus and medial preoptic area exhibited peak heavy nuclear labelling on day E14. Furthermore, perifornical and ventromedial leu-enkephalin neurons were also born relatively early (peak on days E12 and E13, respectively). However, the leu-enkephalin neurons in the medial mammillary nucleus were born the latest of all cell groups studied (i.e., peak at E15).     The immune environment of the mammary gland fluctuates during post-lactational regression and correlates with tumour growth rate

    Post-lactational mammary gland regression encompasses extensive programmed cell death and removal of milk-producing epithelial cells, breakdown of extracellular matrix components and redifferentiation of stromal adipocytes. This highly regulated involution process is associated with a transient increased risk of breast cancer in women. Using a syngeneic tumour model, we show that tumour growth is significantly altered depending on the stage of involution at which tumour cells are implanted. Tumour cells injected at day 3 involution grew faster than those in nulliparous mice, whereas tumours initiated at day 6 involution grew significantly slower. These differences in tumour progression correlate with distinct changes in innate immune cells, in particular among F4/80-expressing macrophages and among TCRδ(+) unconventional T cells. Breast cancer post-pregnancy risk is exacerbated in older first-time mothers and, in our model, initial tumour growth is moderately faster in aged mice compared with young mice. Our results have implications for breast cancer risk and the use of anti-inflammatory therapeutics for postpartum breast cancers

    A hierarchy of happiness? Mokken scaling analysis of the Oxford Happiness Inventory

    The items of the Oxford Happiness Inventory (OHI, a self-report assessment of happiness, are subjected to an analysis for hierarchy among its items. By using Mokken scaling analyses we can assess whether items can reliably be ordered between persons as severity indicators on a latent trait; in this case, a latent trait of Happiness. OHI item-level data from 1024 participants were entered into the Mokken Scaling Procedure (MSP) seeking reliable scales with H > 0.30. 12 OHI items formed a reliable and statistically significant hierarchy. However, the MSP values indicate a 'weak' scale. The 'most difficult' (happiest) item on the scale is 'feeling energetic' and the 'least difficult' (least happy) is 'I have fun'. Items in the scale are consistent with what is already known about both happiness and low mood. The reduction in the OHI's items from 29 to 12 in the Mokken scale may have utility making it more accessible to participants as well as identifying items with reliably different levels of 'difficulty'.
