    Shared Identity, Trustworthiness, and Social Distance in Online Training

    This research examines shared identity in the context of online training. Specifically, we examine the impact of perceived shared identity on social distance, operationalized as one's willingness to work with another person and their willingness to recommend that person for a job. We conduct two studies of simulated online training and find that increased shared identity plays an important role in the online training process and that perceptions of trustworthiness mediate the relationship between shared identity and social distance.     Resident Skin-specific γδ T Cells Provide Local, Nonredundant Regulation of Cutaneous Inflammation

    The function of the intraepithelial lymphocyte (IEL) network of T cell receptor (TCR) γδ+ (Vγ5+) dendritic epidermal T cells (DETC) was evaluated by examining several mouse strains genetically deficient in γδ T cells (δ−/− mice), and in δ−/− mice reconstituted with DETC or with different γδ cell subpopulations. NOD.δ−/− and FVB.δ−/− mice spontaneously developed localized, chronic dermatitis, whereas interestingly, the commonly used C57BL/6.δ−/− strain did not. Genetic analyses indicated a single autosomal recessive gene controlled the dermatitis susceptibility of NOD.δ−/− mice. Furthermore, allergic and irritant contact dermatitis reactions were exaggerated in FVB.δ−/−, but not in C57BL/6.δ−/− mice. Neither spontaneous nor augmented irritant dermatitis was observed in FVB.β−/− δ−/− mice lacking all T cells, indicating that αβ T cell–mediated inflammation is the target for γδ-mediated down-regulation. Reconstitution studies demonstrated that both spontaneous and augmented irritant dermatitis in FVB.δ−/− mice were down-regulated by Vγ5+ DETC, but not by epidermal T cells expressing other γδ TCRs. This study demonstrates that functional impairment at an epithelial interface can be specifically attributed to absence of the local TCR-γδ+ IEL subset and suggests that systemic inflammatory reactions may more generally be subject to substantial regulation by local IELs

    Intellectual ability in tuberous sclerosis complex correlates with predicted effects of mutations on TSC1 and TSC2 proteins.

    BACKGROUND: Tuberous sclerosis complex is a multisystem genetic disease, caused by mutation in the TSC1 or TSC2 genes, associated with many features, including intellectual disability (ID). We examined psychometric profiles of patients with TSC1 or TSC2 mutations and tested whether different mutation types were associated with different degrees of intellectual ability. METHODS: One hundred subjects with known TSC1/TSC2 mutations were assessed using a range of IQ or developmental quotient (DQ) measures. Effects of mutations on TSC1/TSC2 proteins were inferred from sequence data and published biochemical studies. RESULTS: Most individuals with TSC1 mutations fell on a normal distribution identical to the general population, with ∼10% showing profound ID. Of individuals with TSC2 mutations, 34% showed profound ID, and the remainder a pattern of IQ/DQ more variable and shifted to the left than in TSC1 or the general population. Truncating TSC1 mutations were all predicted to be subject to nonsense-mediated mRNA decay. Mutations predicted to result in unstable protein were associated with less severe effects on IQ/DQ. There was a statistically significant negative correlation between length of predicted aberrant C-terminal tails arising from frameshift mutations in TSC1 and IQ/DQ; for TSC2 a positive but not statistically significant correlation was observed. CONCLUSION: We propose a model where (i) IQ/DQ correlates inversely with predicted levels and/or deleterious biochemical effects of mutant TSC1 or TSC2 protein, and (ii) longer aberrant C-terminal tails arising from frameshift mutations are more detrimental for TSC1 and less for TSC2.     The Distinct Contributions of Murine T Cell Receptor (TCR)γδ+ and TCRαβ+ T Cells to Different Stages of Chemically Induced Skin Cancer

    Epithelial tissues in which carcinomas develop often contain systemically derived T cell receptor (TCR)αβ+ cells and resident intraepithelial lymphocytes that are commonly enriched in TCRγδ+ cells. Recent studies have demonstrated that γδ cells protect the host against chemically induced cutaneous malignancy, but the role of αβ T cells has been enigmatic, with both protective and tumor-enhancing contributions being reported in different systems. This study aims to clarify the contributions of each T cell type to the regulation of squamous cell carcinoma induced in FVB mice by a two-stage regimen of 7,12-dimethylbenz[a]anthracene initiation followed by repetitive application of the tumor promoter 12-O-tetradecanoylphorbol 13-acetate. This protocol permits one to monitor the induction of papillomas and the progression of those papillomas to carcinomas. The results show that whereas γδ cells are strongly protective, the nonredundant contributions of αβ T cells to the host's protection against papillomas are more modest. Furthermore, at both high and low doses of carcinogens, αβ T cells can contribute to rather than inhibit the progression of papillomas to carcinomas. As is likely to be the case in humans, this study also shows that the contribution of T cells to tumor immunosurveillance is regulated by modifier genes

    Comparing Water Level Estimation in Coastal and Shelf Seas From Satellite Altimetry and Numerical Models

    Accurately resolving coastal Total Water Levels (TWL) is crucial for socio-economic and environmental reasons. Recent efforts in satellite altimetry and numerical modelling have improved accuracy over near-shore areas. In this study we used data from tide gauges (TGs), SAR-mode altimetry from two satellites (Sentinel-3A (S3) and CryoSat-2 (C2)), and a state-of-the-art high-resolution regional coupled environmental prediction model (Amm15) to undertake an inter-comparison between the observations and the model. The aim is to quantify our capability to measure TWL around the UK coast, and to quantify the capacity of the model to represent coastal TWL. Results show good agreement between the satellite and TG data (the mean correlation (R) over seventeen TGs between June 2016 and September 2017 is 0.85 for S3 and 0.80 for C2). The satellite-model comparison shows that the variability is well captured (R=0.98 for both satellite), however there is an offset (-0.23m for S3, -0.15m for C2) between the satellite and model data, that is near-constant across the domain. This offset is partly attributed to the difference in the reference level used by the satellites and the model, and residual differences linked to the temporal resolution of the model. The best agreement between model and satellite is seen away from the coast, further than 3-4km offshore. However, even within the coastal band, R remains high, ~0.95 (S3) and ~0.96 (C2). In conclusion, models are still essential to represent TWL in coastal regions where there is no cover from in-situ observations, but satellite altimeters can now provide valuable observations that are reliable much closer to the coast than before

    Discovery and Timing of Millisecond Pulsars with the Arecibo 327 MHz Drift-Scan Survey

    We present the discovery and timing solutions of four millisecond pulsars (MSPs) discovered in the Arecibo 327 MHz Drift-Scan Pulsar Survey. Three of these pulsars are in binary systems, consisting of a redback (PSR J2055+1545), a black widow (PSR J1630+3550), and a neutron star-white dwarf binary (PSR J2116+1345). The fourth MSP, PSR J2212+2450, is isolated. We present the multi-year timing solutions as well as polarization properties across a range of radio frequencies for each pulsar. We perform a multi-wavelength search for emission from these systems and find an optical counterpart for PSR J2055+1545 in Gaia DR3, as well as a gamma-ray counterpart for PSR J2116+1345 with the Fermi-LAT telescope. Despite the close co-location of PSR J2055+1545 with a Fermi source, we are unable to detect gamma-ray pulsations, likely due to the large orbital variability of the system. This work presents the first two binaries found by this survey with orbital periods shorter than a day; we expect to find more in the 40% of the survey data which have yet to be searched.Comment: 20 pages, 11 figures, 4 tables.     Defining geo-habitats for groundwater ecosystem assessments: an example from England and Wales (UK)

    Groundwater ecosystems comprising micro-organisms and metazoans provide an important contribution to global biodiversity. Their complexity depends on geology, which determines the physical habitat available, and the chemical conditions within it. Despite this, methods of classifying groundwater habitats using geological data are not well established, and researchers have called for higher resolution habitat frameworks. A novel habitat typology for England and Wales (UK) is proposed, which distinguishes 11 geological habitats (geo-habitats) on hydrogeological principles and maps their distribution. Hydrogeological and hydrochemical data are used to determine the characteristics of each geo-habitat, and demonstrate their differences. Using these abiotic parameters, a new method to determine abiotic habitat quality is then developed. The geo-habitats had significantly different characteristics validating the classification system. Karstic and porous habitats were generally of higher quality than fractured habitats. All geo-habitats were highly heterogeneous, containing both high quality habitat patches that are likely to be suitable for fauna, and areas of low quality that may limit faunal distributions. Overall, 70 % of England and Wales are covered by lower quality fractured habitats, with only 13 % covered by higher quality habitats. The main areas of high quality habitats occur in central England as north-south trending belts, possibly facilitating dispersal along this axis. They are separated by low quality geo-habitats that may prevent east-west dispersal of fauna. In south-west England and in Wales suitable geo-habitats occur as small isolated patches. Overall, this paper provides a new national-scale typology that is adaptable for studies in other geographic areas
