    Statistical Evaluation of NDE Reliability in the Aerospace Industry

    The goal of this paper is to review the statistical methods used in the aerospace industries to evaluate NDE reliability. The techniques presented are consistent with the damage tolerant design and structural maintenance philosophies of the aerospace industry. The first part of this paper establishes the evaluation criteria and discusses the history of NDE reliability evaluations. The second part describes the state-of-the-art analysis methods through examples from the retirement for cause (RFC) inspection system evaluation. The last part of the paper discusses some techniques used to rate operator performance and deal with false calls

    Establishing consensus of position-specific predictors for elite youth soccer in England

    Purpose: To construct a valid and reliable methodology for the development of position- specific predictors deemed appropriate for talent identification purposes within elite youth soccer in England. Method: N = 10 experts participated in a three-step modified e-Delphi poll to generate consensus on a series of generic youth player attributes. A follow up electronic survey completed by coaches, scouts and recruitment staff (n = 99) ranked these attributes to specific player- positions. Results: A final list of 44 player attributes found consensus using the three-step modified e-Delphi poll and the findings indicated that player-positional attributes considered most important at the youth phase are more psychological and technical than physiological or anthropometric. Despite ‘hidden’ attributes (e.g. coachability, flair, versatility, vision etc) finding consensus on the e-Delphi poll, there was no evidence to support these traits when associated with a specific playing position. Conclusion: For those practitioners responsible for talent recruitment, our findings may provide greater understanding of the multiple attributes required for some playing positions. However, new ecological research is required to assess the veracity of our claims

    Validation of cognitive models for collaborative hybrid systems with discrete human input

    We present a method to validate a cognitive model, based on the cognitive architecture ACT-R, in dynamic humanautomation systems with discrete human input. We are inspired by the general problem of K-choice games as a proxy for many decision making applications in dynamical systems. We model the human as a Markovian controller based on gathered experimental data, that is, a non-deterministic control input with known likelihoods of control actions associated with certain configurations of the state-space. We use reachability analysis to predict the outcome of the resulting discrete-time stochastic hybrid system, in which the outcome is defined as a function of the system trajectory. We suggest that the resulting expected outcomes can be used to validate the cognitive model against actual human subject data. We apply our method to a twochoice game in which the human is tasked with maximizing net coverage of a robotic swarm that can operate under rendezvous or deployment dynamics. We validate the corresponding ACTR cognitive model generated with the data from eight human subjects. The novelty of this work is 1) a method to compute expected outcome in a hybrid dynamical system with a Markov chain model of the human's discrete choice, and 2) application of this method to validation of cognitive models with a database of actual human subject data

    Comparison between Suitable Priors for Additive Bayesian Networks

    Additive Bayesian networks are types of graphical models that extend the usual Bayesian generalized linear model to multiple dependent variables through the factorisation of the joint probability distribution of the underlying variables. When fitting an ABN model, the choice of the prior of the parameters is of crucial importance. If an inadequate prior - like a too weakly informative one - is used, data separation and data sparsity lead to issues in the model selection process. In this work a simulation study between two weakly and a strongly informative priors is presented. As weakly informative prior we use a zero mean Gaussian prior with a large variance, currently implemented in the R-package abn. The second prior belongs to the Student's t-distribution, specifically designed for logistic regressions and, finally, the strongly informative prior is again Gaussian with mean equal to true parameter value and a small variance. We compare the impact of these priors on the accuracy of the learned additive Bayesian network in function of different parameters. We create a simulation study to illustrate Lindley's paradox based on the prior choice. We then conclude by highlighting the good performance of the informative Student's t-prior and the limited impact of the Lindley's paradox.     Caregiver characteristics and bereavement needs: Findings from a population study

    © 2016, © The Author(s) 2016. Background: Globally, most care for people with life-limiting illnesses is provided by informal caregivers. Identifying characteristics of caregivers that may have unmet needs and negative outcomes can help provide better support to facilitate adjustment. Aim: We compared characteristics, expressed unmet needs and outcomes for spousal caregivers, with other caregivers at the end of life, by gender and age. Design: The South Australian Health Omnibus is an annual, random, face-to-face, cross-sectional survey wherein respondents are asked about end-of-life care. Setting/participants: Participants were aged over 15 years, resided in households in South Australia and had someone close to them die from a terminal illness in the last 5 years. Results: Of the 1540 respondents who provided hands-on care for someone close at the end of life, 155 were widows/widowers. Bereaved spousal caregivers were more likely to be older, female, better educated, have lower incomes, less full-time work, English as second language, sought help with grief and provided more day-to-day care for longer periods. Spousal caregivers were less likely to be willing to take on caregiving again, less able to ‘move on’ with life and needed greater emotional support and information about illness and services. The only difference between widows and widowers was older age of spouse in women. Younger spousal caregivers perceived greater unmet emotional needs and were significantly less likely to be able to ‘move on’. Conclusion: Spousal caregivers are different from other caregivers, with more intense needs that are not fully met. These have implications for bereavement, health and social services

    Understanding the stigma of psychosis in ethnic minority groups: A qualitative exploration

    Psychosis is a mental health difficulty which is widely stigmatized. The stigma of psychosis can lead to detrimental consequences and cause further distress. The aim of this study was to explore the experience of stigma and discrimination of psychosis from the perspective of service users from ethnic minority backgrounds. A total of 21 semistructured interviews were conducted with service users with psychosis from ethnic minority backgrounds examining their experiences of stigma from psychosis. Qualitative data were analyzed using thematic analysis. Five superordinate themes were identified, “social and cultural context of stigma,” “stigma is a family problem,” “stigma and discrimination within mental health services,” “intrapersonal impacts,” and “managing stigma within relationships.” Stigma is a significant concern for people with psychosis from an ethnic minority background. It is important that stigma and discrimination are appropriately assessed and considered within the care of people from ethnic minority backgrounds

    Testosterone enables growth and hypertrophy in fusion impaired myoblasts that display myotube atrophy: deciphering the role of androgen and IGF-I receptors

    We have previously highlighted the ability of testosterone to improve differentiation and myotube hypertrophy in fusion impaired myoblasts that display reduced myotube hypertrophy at 72hrs (after transfer to low serum media) via multiple population doublings (PD) vs. their parental controls (CON); an observation which is abrogated via PI3K/Akt inhibition (Deane et al. 2013). However, whether the most predominant molecular mechanism responsible for T induced hypertrophy occurs directly via androgen receptor or indirectly via IGF-IR/PI3K/Akt pathway is currently debated. PD and CON C2C12 muscle cells were exposed to low serum conditions in the presence or absence of T (100 nM) ± inhibitors of AR (flutamide/F, 40 Όm) and IGF-IR (Picropodophyllin/PPP, 150 nM) for 72 hrs and 7 days (early/late muscle differentiation respectively). T increased AR and Akt abundance, myogenin expression, and myotube hypertrophy, but not ERK1/2 activity in both CON and PD cell types. Akt activity was not increased significantly in either cell type with T. Testosterone was unable to promote early differentiation in the presence of IGF-IR inhibitor (PPP) yet still able to promote appropriate later increases in myotube hypertrophy and AR abundance despite IGF-IR inhibition. The addition of the AR inhibitor powerfully attenuated all T induced increases in differentiation and myotube hypertrophy with corresponding reductions in AR abundance, phosphorylated Akt, ERK1/2 and gene expression of IGF-I, myoD and myogenin with increases in myostatin mRNA both cell types. Interestingly, despite basally reduced differentiation and myotube hypertrophy, PD cells showed larger increased in AR abundance vs. CON cells, a response abrogated in the presence of AR but not IGF-IR inhibitors. Furthermore, T induced increases in Akt abundance were sustained despite the presence of IGF-IR inhibition in PD cells only. However, flutamide alone reduced IGF-IR mRNA in both cell types across time points, with an observed reduction in activity of ERK and Akt, perhaps suggesting that IGF-IR was transcriptionally regulated by AR. However, where testosterone increased AR protein content there was no increases observed in IGF-IR gene expression. Overall, this suggested that sufficient AR was important to enable normal gene expression of IGF-IR and downstream signalling, yet elevated levels of AR due to testosterone had no further effect on IGF-IR, despite testosterone increasing Akt abundance in the presence of IGF-IR inhibitor. In conclusion, testosterones ability to improve differentiation and myotube hypertrophy occurred predominately via increases in AR and Akt abundance in both CON and PD cells, with fusion impaired cells (PD) showing an increased responsiveness to T induced AR levels. Finally, T induced increases in myotube hypertrophy (but not early differentiation) occurred independently of upstream IGF-IR input, however it appears that normal AR function in basal conditions is required for adequate IGF-IR gene expression and downstream Akt abundance

    Impaired hypertrophy in myoblasts is improved with testosterone administration

    We investigated the ability of testosterone (T) to restore differentiation in multiple population doubled (PD) murine myoblasts, previously shown to have reduced differentiation in monolayer and bioengineered skeletal muscle cultures vs. their parental controls (CON) (Sharples et al., 2011, 2012 [7] and [26]). Cells were exposed to low serum conditions in the presence or absence of T (100 nM) ± PI3K inhibitor (LY294002) for 72 h and 7 days (early and late muscle differentiation respectively). Morphological analyses were performed to determine myotube number, diameter (ÎŒm) and myonuclear accretion as indices of differentiation and myotube hypertrophy. Changes in gene expression for myogenin, mTOR and myostatin were also performed. Myotube diameter in CON and PD cells increased from 17.32 ± 2.56 ÎŒm to 21.02 ± 1.89 ÎŒm and 14.58 ± 2.66 ÎŒm to 18.29 ± 3.08 ÎŒm (P ≀ 0.05) respectively after 72 h of T exposure. The increase was comparable in both PD (+25%) and CON cells (+21%) suggesting a similar intrinsic ability to respond to exogenous T administration. T treatment also significantly increased myonuclear accretion (% of myotubes expressing 5+ nuclei) in both cell types after 7 days exposure (P ≀ 0.05). Addition of PI3K inhibitor (LY294002) in the presence of T attenuated these effects in myotube morphology (in both cell types) suggesting a role for the PI3K pathway in T stimulated hypertrophy. Finally, PD myoblasts showed reduced responsiveness to T stimulated mRNA expression of mTOR vs. CON cells and T also reduced myostatin expression in PD myoblasts only. The present study demonstrates testosterone administration improves hypertrophy in myoblasts that basally display impaired differentiation and hypertrophic capacity vs. their parental controls, the action of testosterone in this model was mediated by PI3K/Akt pathway

    Precise measurement of positronium hyperfine splitting using the Zeeman effect

    Positronium is an ideal system for the research of the quantum electrodynamics (QED) in bound state. The hyperfine splitting (HFS) of positronium, ΔHFS\Delta_{\mathrm{HFS}}, gives a good test of the bound state calculations and probes new physics beyond the Standard Model. A new method of QED calculations has revealed the discrepancy by 15\,ppm (3.9σ\sigma) of ΔHFS\Delta_{\mathrm{HFS}} between the QED prediction and the experimental average. There would be possibility of new physics or common systematic uncertainties in the previous all experiments. We describe a new experiment to reduce possible systematic uncertainties and will provide an independent check of the discrepancy. We are now taking data and the current result of ΔHFS=203.395 1±0.002 4(stat.,12 ppm)±0.001 9(sys.,9.5 ppm) GHz\Delta_{\mathrm{HFS}} = 203.395\,1 \pm 0.002\,4 (\mathrm{stat.}, 12\,\mathrm{ppm}) \pm 0.001\,9 (\mathrm{sys.}, 9.5\,\mathrm{ppm})\,\mathrm{GHz} has been obtained so far.     Subcellular compartmentalisation of copper, iron, manganese, and zinc in the Parkinson's disease brain

    © 2017 The Royal Society of Chemistry. Elevated iron and decreased copper levels are cardinal features of the degenerating substantia nigra pars compacta in the Parkinson's disease brain. Both of these redox-active metals, and fellow transition metals manganese and zinc, are found at high concentrations within the midbrain and participate in a range of unique biological reactions. We examined the total metal content and cellular compartmentalisation of manganese, iron, copper and zinc in the degenerating substantia nigra, disease-affected but non-degenerating fusiform gyrus, and unaffected occipital cortex in the post mortem Parkinson's disease brain compared with age-matched controls. An expected increase in iron and a decrease in copper concentration was isolated to the soluble cellular fraction, encompassing both interstitial and cytosolic metals and metal-binding proteins, rather than the membrane-associated or insoluble fractions. Manganese and zinc levels did not differ between experimental groups. Altered Fe and Cu levels were unrelated to Braak pathological staging in our cases of late-stage (Braak stage V and VI) disease. The data supports our hypothesis that regional alterations in Fe and Cu, and in proteins that utilise these metals, contribute to the regional selectively of neuronal vulnerability in this disorder
