Fabrication of the DESI Corrector Lenses
The Dark Energy Spectroscopic Instrument (DESI) is under construction to
measure the expansion history of the Universe using the Baryon Acoustic
Oscillation technique. The spectra of 35 million galaxies and quasars over
14000 square degrees will be measured during the life of the experiment. A new
prime focus corrector for the KPNO Mayall telescope will deliver light to 5000
fiber optic positioners. The fibers in turn feed ten broad-band spectrographs.
We describe the DESI corrector optics, a series of six fused silica and
borosilicate lenses. The lens diameters range from 0.8 to 1.1 meters, and their
weights 84 to 237 kg. Most lens surfaces are spherical, and two are challenging
10th-order polynomial aspheres. The lenses have been successfully polished and
treated with an antireflection coating at multiple subcontractors, and are now
being integrated into the DESI corrector barrel assembly at University College
London. We describe the final performance of the lenses in terms of their
various parameters, including surface figure, homogeneity, and others, and
compare their final performance against the demanding DESI corrector
requirements. Also we describe the reoptimization of the lens spacing in their
corrector barrel after their final measurements are known. Finally we assess
the performance of the corrector as a whole, compared to early budgeted
estimates
Biomarkers of seafood intake during pregnancy â Pollutants versus fatty acids and micronutrients
Intake of fish and seafood during pregnancy may have certain beneficial effects on fetal development, but measurement of intake using questionnaires is unreliable. Here, we assessed several candidate biomarkers of seafood intake, including long-chain omega 3 fatty acids (n-3 LCPUFA), selenium, iodine, methylmercury, and different arsenic compounds, in 549 pregnant women (gestational week 29) in the prospective birth cohort NICE (Nutritional impact on Immunological maturation during Childhood in relation to the Environment). Proportions of the fatty acids eicosapentaenoic acid (EPA), docosapentaenoic acid (DPA), and docosahexaenoic acid (DHA) in erythrocytes were measured using gas chromatography with flame ionization detector. Selenium was measured in blood plasma and erythrocytes, mercury and arsenic in erythrocytes, and iodine and several arsenic compounds in urine, using inductively coupled plasma mass spectrometry, arsenic compounds after first being separated by ion exchange high-performance liquid chromatography (HPLC). Each biomarker was related to intake of total seafood and to intake of fatty and lean fish, and shellfish in third trimester, estimated from a semi-quantitative food frequency questionnaire filled out in gestational week 34. The pregnant women reported a median total seafood intake of 184 g/week (5th-95th percentiles: 34â465 g/week). This intake correlated most strongly with erythrocyte mercury concentrations (rho = 0.49, p < 0.001), consisting essentially of methylmercury, followed by total arsenic in erythrocytes (rho = 0.34, p < 0.001), and arsenobetaine in urine (rho = 0.33, p < 0.001), the main form of urinary arsenic. These biomarkers correlated well with intake of both fatty fish, lean fish, and shellfish. Erythrocyte DHA and plasma selenium correlated, although weakly, mainly with fatty fish (rho = 0.25 and 0.22, respectively, both p < 0.001). In conclusion, elevated concentrations of erythrocyte mercury and urinary arsenobetaine can be useful indicators of seafood intake, more so than the n-3 LCPUFAs. However, the relative importance of the biomarkers may differ depending on the type and amount of seafood consumed
Unbiased clustering estimates with the DESI fibre assignment
The Emission Line Galaxy survey made by the Dark Energy Spectroscopic
Instrument (DESI) survey will be created from five passes of the instrument on
the sky. On each pass, the constrained mobility of the ends of the fibres in
the DESI focal plane means that the angular-distribution of targets that can be
observed is limited. Thus, the clustering of samples constructed using a
limited number of passes will be strongly affected by missing targets. In two
recent papers, we showed how the effect of missing galaxies can be corrected
when calculating the correlation function using a weighting scheme for pairs.
Using mock galaxy catalogues we now show that this method provides an unbiased
estimator of the true correlation function for the DESI survey after any number
of passes. We use multiple mocks to determine the expected errors given one to
four passes, compared to an idealised survey observing an equivalent number of
randomly selected targets. On BAO scales, we find that the error is a factor 2
worse after one pass, but that after three or more passes, the errors are very
similar. Thus we find that the fibre assignment strategy enforced by the design
of DESI will not affect the cosmological measurements to be made by the survey,
and can be removed as a potential risk for this experiment.Comment: 11 pages, 8 figure

Reduced local mutation density in regulatory DNA of cancer genomes is linked to DNA repair
Carcinogenesis and neoplastic progression are mediated by the accumulation of somatic mutations. Here we report that the local density of somatic mutations in cancer genomes is highly reduced specifically in accessible regulatory DNA defined by DNase I hypersensitive sites. This reduction is independent of any known factors influencing somatic mutation density and is observed in diverse cancer types, suggesting a general mechanism. By analyzing individual cancer genomes1, we show that the reduced local mutation density within regulatory DNA is linked to intact global genome repair machinery, with nearly complete abrogation of the hypomutation phenomenon in individual cancers that possess mutations in multiple nucleotide excision repair components. Together, our results connect chromatin structure, gene regulation and cancer-associated somatic mutation
Association of maternal urinary fluoride concentrations during pregnancy with size at birth and the potential mediation effect by maternal thyroid hormones: The Swedish NICE birth cohort
Background: Observational studies have indicated that elevated maternal fluoride exposure during pregnancy may impair child neurodevelopment but a potential impact on birth outcomes is understudied. Objectives: To evaluate the impact of gestational fluoride exposure on birth outcomes (birth size and gestational age at birth) and to assess the potential mediating role of maternal thyroid hormones. Methods: We studied 583 mother-child dyads in the NICE cohort in northern Sweden. Maternal fluoride exposure was assessed by measuring urinary concentrations at late pregnancy (median: 29th gestational week) using an ion selective electrode. Plasma levels of free and total thyroxine (fT4, tT4) and triiodothyronine (fT3, tT3), and thyroid stimulating hormone (TSH) were measured with electrochemiluminescence immunoassays. The infant\u27s weight, length, head circumference, and gestational age at birth were extracted from hospital records. Results: Median urinary fluoride concentration was 0.71 mg/L (5th-95th percentile 0.31â1.9 mg/L; specific gravity adjusted). In multivariable-adjusted regression models, every 1 mg/L increase of maternal urinary fluoride was associated with a mean increase in birth weight by 84 g (95%CI: 30, 138), length by 0.41 cm (95%CI: 0.18, 0.65), head circumference by 0.3 cm (95%CI: 0.1, 0.4), and with increased odds of being born large for gestational age (OR = 1.39, 95%CI: 1.03, 1.89). Every 1 mg/L increase of maternal urinary fluoride was also associated with a mean increase of the plasma fT3:fT4 ratio (B = 0.007, 95%CI: 0.000, 0.014), but not with the hormones or TSH. In mediation analyses, the maternal fT3:fT4 ratio did not explain the urinary fluoride-birth size relationships. Discussion: Gestational urinary fluoride concentrations were associated with increased size at birth and even with increased odds of being born large for gestational age. The fluoride-related associations with increased size at birth were not explained by changes in maternal thyroid hormone levels

High-throughput identification of genotype-specific cancer vulnerabilities in mixtures of barcoded tumor cell lines.
Hundreds of genetically characterized cell lines are available for the discovery of genotype-specific cancer vulnerabilities. However, screening large numbers of compounds against large numbers of cell lines is currently impractical, and such experiments are often difficult to control. Here we report a method called PRISM that allows pooled screening of mixtures of cancer cell lines by labeling each cell line with 24-nucleotide barcodes. PRISM revealed the expected patterns of cell killing seen in conventional (unpooled) assays. In a screen of 102 cell lines across 8,400 compounds, PRISM led to the identification of BRD-7880 as a potent and highly specific inhibitor of aurora kinases B and C. Cell line pools also efficiently formed tumors as xenografts, and PRISM recapitulated the expected pattern of erlotinib sensitivity in vivo
Acarbose improves health and lifespan in aging HET3 mice
To followâup on our previous report that acarbose (ACA), a drug that blocks postprandial glucose spikes, increases mouse lifespan, we studied ACA at three doses: 400, 1,000 (the original dose), and 2,500Â ppm, using genetically heterogeneous mice at three sites. Each dose led to a significant change (by logârank test) in both sexes, with larger effects in males, consistent with the original report. There were no significant differences among the three doses. The two higher doses produced 16% or 17% increases in median longevity of males, but only 4% or 5% increases in females. Age at the 90th percentile was increased significantly (8%â11%) in males at each dose, but was significantly increased (3%) in females only at 1,000Â ppm. The sex effect on longevity is not explained simply by weight or fat mass, which were reduced by ACA more in females than in males. ACA at 1,000Â ppm reduced lung tumors in males, diminished liver degeneration in both sexes and glomerulosclerosis in females, reduced blood glucose responses to refeeding in males, and improved rotarod performance in aging females, but not males. Three other interventions were also tested: ursolic acid, 2â(2âhydroxyphenyl) benzothiazole (HBX), and INTâ767; none of these affected lifespan at the doses tested.
