    Potential of Sentinel-2 satellite images to monitor vine fields grown at a territorial scale

    Aim: The aim of this short note is to provide first insights into the ability of Sentinel-2 images to monitor vine growth across a whole season. It focuses on verifying the practical temporal resolution that can be reached with Sentinel-2 images, the main stages of Mediterranean vineyard development as well as potential relevant agronomic information that can be seen on the temporal vegetation curves arising from Sentinel-2 images. Methods and results: The study was carried out in 2017 in a production vineyard located in southern France, 2 km from the Mediterranean seashore. Sentinel-2 images acquired during the whole vine growing cycle were considered, i.e. between the 3rd of March 2017 and the 10th of October 2017. The images were used to compute the classical normalized difference vegetation index (NDVI). Time series of NDVI values were analyzed on four blocks chosen for exhibiting different features, e.g. age, missing plants, weeding practices. The practical time lag between two usable images was closer to 16 days than to the 10 theoretical days (with only one satellite available at the date of the experiment), i.e. near 60% of the theoretical one. Results show that it might be possible to identify i) the main steps of vine development (e.g. budburst, growth, trimming, growth stop and senescence), ii) weed management and inter-row management practices, and iii) possible reasons for significant inter-block differences in vegetative expression (e.g. young vines that have recently been planted, low-productive blocks affected by many missing vines). Conclusions: Although this experiment was conducted at a time when Sentinel-2b was not fully operational, results showed that a sufficient number of usable images was available to monitor vine development. The availability of two Sentinel satellites (2a and 2b) in upcoming seasons should increase the number of usable images and the temporal resolution of the time series. This study also showed the limitations of the Sentinel-2 images’ resolution to provide within-block information in the case of small blocks or blocks with complex borders or both. Significance and impact of the study: This technical note demonstrated the potential of Sentinel-2 images to characterize vineyard blocks’ vigor and to monitor winegrowers’ practices at a territorial (regional) scale. The impact of management operations such as weeding and trimming, along with their incidence on canopy size, were observed on the NDVI time series. Some relevant parameters (slope, maximum values) may be derived from the NDVI time series, providing new insights into the monitoring of vineyards at a large scale. These results provided areas for further investigation, especially regarding the development of new indicators to characterize block-climate relationships

    Surveillance of endoscopes : comparison of different sampling techniques

    OBJECTIVE: To compare different techniques of endoscope sampling to assess residual bacterial contamination. DESIGN: Diagnostic study. SETTING: The endoscopy unit of an 1,100-bed university hospital performing similar to 13,000 endoscopic procedures annually. METHODS: In total, 4 sampling techniques, combining flushing fluid with or without a commercial endoscope brush, were compared in an endoscope model. Based on these results, sterile physiological saline flushing with or without PULL THRU brush was selected for evaluation on 40 flexible endoscopes by adenosine triphosphate (ATP) measurement and bacterial culture. Acceptance criteria from the French National guideline (<25 colony-forming units [CFU] per endoscope and absence of indicator microorganisms) were used as part of the evaluation. RESULTS: On biofilm-coated PTFE tubes, physiological saline in combination with a PULL THRU brush generated higher mean ATP values (2,579 relative light units [RLU]) compared with saline alone (1,436 RLU; P=.047). In the endoscope samples, culture yield using saline plus the PULL THRU (mean, 43 CFU; range, 1-400 CFU) was significantly higher than that of saline alone (mean, 17 CFU; range, 0-500 CFU; P<.001). In samples obtained using the saline+PULL THRU brush method, ATP values of samples classified as unacceptable were significantly higher than those of samples classified as acceptable (P=.001). CONCLUSION: Physiological saline flushing combined with PULL THRU brush to sample endoscopes generated higher ATP values and increased the yield of microbial surveillance culture. Consequently, the acceptance rate of endoscopes based on a defined CFU limit was significantly lower when the saline+PULL THRU method was used instead of saline alone

    Combined oropharyngeal/nasal swab is equivalent to nasopharyngeal sampling for SARS-CoV-2 diagnostic PCR

    BackgroundEarly 2020, a COVID-19 epidemic became a public health emergency of international concern. To address this pandemic broad testing with an easy, comfortable and reliable testing method is of utmost concern. Nasopharyngeal (NP) swab sampling is the reference method though hampered by international supply shortages. A new oropharyngeal/nasal (OP/N) sampling method was investigated using the more readily available throat swab.Results35 patients were diagnosed with SARS-CoV-2 by means of either NP or OP/N sampling. The paired swabs were both positive in 31 patients. The one patient who tested negative on both NP and OP/N swab on admission, was ultimately diagnosed on bronchoalveolar lavage fluid. A strong correlation was found between the viral RNA loads of the paired swabs (r=0.76; P<0.05). The sensitivity of NP and OP/N analysis in hospitalized patients (n=28) was 89.3% and 92.7% respectively.ConclusionsThis study demonstrates equivalence of NP and OP/N sampling for detection of SARS-CoV-2 by means of rRT-PCR. Sensitivity of both NP and OP/N sampling is very high in hospitalized patients

    City of things : enabling resource provisioning in smart cities

    In the last few years, traffic over wireless networks has been increasing exponentially due to the impact of IoT. IoT is transforming a wide range of services in different domains of urban life, such as environmental monitoring, home automation, and public transportation. The so-called smart city applications will introduce a set of stringent requirements, such as low latency and high mobility, since services must be allocated and instantiated on demand, simultaneously, close to multiple devices at different locations. Efficient resource provisioning functionalities are needed to address these demanding constraints introduced by smart city applications while minimizing resource costs and maximizing QoS. In this article, the CoT framework is presented, which provides not only data collection and analysis functionalities but also automated resource provisioning mechanisms for future smart city applications. CoT is deployed as a smart city test-bed in Antwerp, Belgium, which allows researchers and developers to easily set up and validate IoT experiments. A smart city use case of air quality monitoring through the deployment of air quality sensors in moving cars is presented showing the full applicability of the CoT framework for a flexible and scalable resource provisioning in the smart city ecosystem

    The Protozoan Parasite Toxoplasma gondii Selectively Reprograms the Host Cell Translatome

    The intracellular parasite Toxoplasma gondii promotes infection by targeting multiple host cell processes; however, whether it modulates mRNA translation is currently unknown. Here, we show that infection of primary murine macrophages with type I or II T. gondii strains causes a profound perturbation of the host cell translatome. Notably, translation of transcripts encoding proteins involved in metabolic activity and components of the translation machinery was activated upon infection. In contrast, the translational efficiency of mRNAs related to immune cell activation and cytoskeleton/cytoplasm organization was largely suppressed. Mechanistically, T. gondii bolstered mechanistic target of rapamycin (mTOR) signaling to selectively activate the translation of mTOR-sensitive mRNAs, including those with a 5'-terminal oligopyrimidine (5' TOP) motif and those encoding mitochondrion-related proteins. Consistent with parasite modulation of host mTOR-sensitive translation to promote infection, inhibition of mTOR activity suppressed T. gondii replication. Thus, selective reprogramming of host mRNA translation represents an important subversion strategy during T. gondii infection
