    Further evidences concerning the systemic spreading of Agrobacterium tumefaciens in the vascular system of grapevines

    The isolation of pathogen, A. tumefaciens, was successful first from the buds, callus tissue and fresh root-tips of forced short cuttings made from symptom-free canes, and furthermore from fresh root-tips and callus tissue of forced, symptomfree root pieces.Both the canes and roots were collected from natural infected, self-rooted vines (cuttings). The results give further evidence regarding systemic spreading of the pathogen in the vascular system of grapevine and to its transmissibility by vegetative propagating material. The tumor-formation on green-wood grafted vines, of which the shoots were collected in a vineyard which was more than 25 percent infected with crown gall, gives further convincing evidence

    Liquid encapsulated float zone process and apparatus

    The process and apparatus for growing crystals using float zone techniques are described. A rod of crystalline materials is disposed in a cylindrical container, leaving a space between the rod and container walls. This space is filled with an encapsulant, selected to have a slightly lower melting point than the crystalline material. The rod is secured to a container end cap at one end and to a shaft at its other end. A piston slides over the rod and provides pressure to prevent loss of volatile components upon melting of the rod. Prior to melting the rod the container is first heated to melt the encapsulant, with any off-gas from this step being vented to a cavity behind the piston. The piston moves slightly forward owing to volume change upon melting of the encapsulant, and the vent passageway is closed. The container is then moved longitudinally through a heated zone to progressively melt sections of the rod as in conventional float zone processes. The float zone technique may be used in the microgravity environment of space

    Effect of the grape chrome-mosaic and grape fanleaf yellow-mosaic virus-infection on the photosynthetical carbon dioxide fixation in vine-leaves

    The intensity of photosynthetical C02-fixation in red Veltliner leaves, strongly or slightly infected with GCM, was compared with leaves of healthy individuals. Leaves of white Sultana, slightly infected with GFYM virus were also compared with leaves of healthy plants. The photosynthetic activity was calculated by measuring the CO2-fixation in dark and in light and the data thus obtained were compared with the content of chlorophyll. In GCM infected leaves with slight symptoms the chlorophyll level and the intensity of photosynthetical CO2 -fixation gradually decreased; in leaves with severe symptoms, the specific activity (CO2-fixation/mg chlorophyll) was relatively higher, presumably· due to the increased transparency of the tissues. After GFYM infection the photosynthetical CO2-fixation decreased strinkingly, independently of the strength of leaf symptoms. It is supposed that the destruction of the chlorophyll-protein-complexe caused by the virus infection asserts an inhibiting effect on the photosynthetic activity

    The Advanced Automated Directional Solidification Furnace

    The Advanced Automated Directional Solidification Furnace (AADSF) is a five zone tubular furnace designed for Bridgman-Stockbarger, other techniques of crystal growth involving multiple temperature zones such as vapor transport experiments and other materials science experiments. The five zones are primarily designed to produce uniform hot and cold temperature regions separated by an adiabatic region constructed of a heat extraction plate and an insert to reduce radiation from the hot to the cold zone. The hot and cold zone temperatures are designed to reach 1600 C and 1100 C, respectively. AADSF operates on a Multi-Purpose Experiment Support Structure (MPESS) within the cargo bay of the Space Shuttle on the United States Microgravity Payload (USMP) missions. Two successful flights, both employing the directional solidification or Bridgman Stockbarger technique for crystal growth have been made, and crystals of HgCdTe and PbSnTe grown in microgravity have been produced on USMP-2 and USMP-3, respectively. The addition of a Sample Exchange Mechanism (SEM) will enable three different samples to be processed on future flights including the USMP-4 mission

    Implication of long-distance regulation of the HOXA cluster in a patient with postaxial polydactyly

    Apparently balanced chromosomal inversions may lead to disruption of developmentally important genes at the breakpoints of the inversion, causing congenital malformations. Characterization of such inversions may therefore lead to new insights in human development. Here, we report on a de novo inversion of chromosome 7 (p15.2q36.3) in a patient with postaxial polysyndactyly. The breakpoints do not disrupt likely candidate genes for the limb phenotype observed in the patient. However, on the p-arm the breakpoint separates the HOXA cluster from a gene desert containing several conserved noncoding elements, suggesting that a disruption of a cis-regulatory circuit of the HOXA cluster could be the underlying cause of the phenotype in this patient

    Genetic Variation and Population Substructure in Outbred CD-1 Mice: Implications for Genome-Wide Association Studies

    Outbred laboratory mouse populations are widely used in biomedical research. Since little is known about the degree of genetic variation present in these populations, they are not widely used for genetic studies. Commercially available outbred CD-1 mice are drawn from an extremely large breeding population that has accumulated many recombination events, which is desirable for genome-wide association studies. We therefore examined the degree of genome-wide variation within CD-1 mice to investigate their suitability for genetic studies. The CD-1 mouse genome displays patterns of linkage disequilibrium and heterogeneity similar to wild-caught mice. Population substructure and phenotypic differences were observed among CD-1 mice obtained from different breeding facilities. Differences in genetic variation among CD-1 mice from distinct facilities were similar to genetic differences detected between closely related human populations, consistent with a founder effect. This first large-scale genetic analysis of the outbred CD-1 mouse strain provides important considerations for the design and analysis of genetic studies in CD-1 mice
