    Novel Polyoxyethylene-Containing Glycolipids Are Synthesized in Corynebacterium matruchotii and Mycobacterium smegmatis Cultured in the Presence of Tween 80

    The addition of polyoxyethylene sorbitan monooleate (Tween 80) to a culture of mycobacteria greatly influences cell permeability and sensitivity to antibiotics but very little is known regarding the underlying mechanism. Here we show that Corynebacterium matruchotii (surrogate of mycobacteria) converts Tween 80 to a structural series of polyoxyethylenic acids which are then used to form novel series-2A and series-2B glycolipids. Minor series-3 glycolipids were also synthesized. The polyoxyethylenic acids replaced corynomycolic acids in the cell wall. Correspondingly the trehalose dicorynomycolate content was reduced. MALDI mass spectrometry, MS-MS, 1H-NMR, and 13C-NMR were used to characterize the series-2 glycolipids. Series-2A glycolipid is trehalose 6-C36:2-corynomycolate-6′-polyoxyethylenate and series-2B glycolipid is trehalose 6-C36:2-corynomycolate-6′-furan ring-containing polyoxyethylenate. Mycobacterium smegmatis grown in the presence of Tween 80 also synthesizes series-2 type glycolipids. The synthesis of these novel glycolipids in corynebacteria and mycobacteria should result in gross changes in the cell wall permeability and drug sensitivity

    Novel Reproductive Behavior in an Asian Frog: Sex‐Reversed Inguinal Amplexus

    Amphibians exhibit diverse reproductive behaviors, including nine documented types of amplexus, the behavior in which male and female frogs position themselves for courtship, oviposition, and fertilization. All known forms of amplexus involve the male on top of or in line horizontally (cloacal apposition) with the female. Here, we report a novel form of amplexus observed in Lau’s leaf litter toad (Leptobrachella laui; Megophryidae) in Hong Kong, China. Termed “sex‐reversed inguinal amplexus,” the female climbs on top of a male and the male transports the female to a concealed breeding site. We were unable to determine whether this was the amplectant position in which frogs engaged during oviposition or solely during courtship and prior to oviposition, but there are a number of possible evolutionary drivers that may have given rise to this behavior, including limiting suitable oviposition sites or strong competition for males among females. Further research will be necessary to understand the evolutionary origins of this novel reproductive behavior

    When memory falls short: The effect of suggestibility and repeated questioning on 3- to 5-year-old children’s recognition of event details

    The present study investigated children’s event recognition as a function of feedback, suggestible question type, and repeated questioning. Forty-three 3- to 5-year-old children read a book and were asked twelve positive-leaning, negative-leaning, and additive-misinformation questions. After six and twelve questions, children were presented with positive or negative feedback and asked the same questions again. Feedback did not affect response accuracy; however, children’s likelihood of changing their answer when questioned a second time varied as a function of question type. Children answered negative-leaning questions more accurately the second time, additive-misinformation questions less accurately the second time, and showed no change with positive-leaning questions. These findings extend prior knowledge of factors that influence children’s recollection of an event

    An Initial Assessment of a SMAP Soil Moisture Disaggregation Scheme Using TIR Surface Evaporation Data over the Continental United States

    The Soil Moisture Active Passive (SMAP) mission is dedicated toward global soil moisture mapping. Typically, an L-band microwave radiometer has spatial resolution on the order of 36-40 km, which is too coarse for many specific hydro-meteorological and agricultural applications. With the failure of the SMAP active radar within three months of becoming operational, an intermediate (9-km) and finer (3-km) scale soil moisture product solely from the SMAP mission is no longer possible. Therefore, the focus of this study is a disaggregation of the 36-km resolution SMAP passive-only surface soil moisture (SSM) using the Soil Evaporative Efficiency (SEE) approach to spatial scales of 3-km and 9-km. The SEE was computed using thermal-infrared (TIR) estimation of surface evaporation over Continental U.S. (CONUS). The disaggregation results were compared with the 3 months of SMAP-Active (SMAP-A) and Active/Passive (AP) products, while comparisons with SMAP-Enhanced (SMAP-E), SMAP-Passive (SMAP-P), as well as with more than 180 Soil Climate Analysis Network (SCAN) stations across CONUS were performed for a 19 month period. At the 9-km spatial scale, the TIR-Downscaled data correlated strongly with the SMAP-E SSM both spatially (r = 0.90) and temporally (r = 0.87). In comparison with SCAN observations, overall correlations of 0.49 and 0.47; bias of 0.022 and 0.019 and unbiased RMSD of 0.105 and 0.100 were found for SMAP-E and TIR-Downscaled SSM across the Continental U.S., respectively. At 3-km scale, TIR-Downscaled and SMAP-A had a mean temporal correlation of only 0.27. In terms of gain statistics, the highest percentage of SCAN sites with positive gains (>55%) was observed with the TIR-Downscaled SSM at 9-km. Overall, the TIR-based downscaled SSM showed strong correspondence with SMAP-E; compared to SCAN, and overall both SMAP-E and TIR-Downscaled performed similarly, however, gain statistics show that TIR-Downscaled SSM slightly outperformed SMAP-E

    Knot Theory: from Fox 3-colorings of links to Yang-Baxter homology and Khovanov homology

    This paper is an extended account of my "Introductory Plenary talk at Knots in Hellas 2016" conference We start from the short introduction to Knot Theory from the historical perspective, starting from Heraclas text (the first century AD), mentioning R.Llull (1232-1315), A.Kircher (1602-1680), Leibniz idea of Geometria Situs (1679), and J.B.Listing (student of Gauss) work of 1847. We spend some space on Ralph H. Fox (1913-1973) elementary introduction to diagram colorings (1956). In the second section we describe how Fox work was generalized to distributive colorings (racks and quandles) and eventually in the work of Jones and Turaev to link invariants via Yang-Baxter operators, here the importance of statistical mechanics to topology will be mentioned. Finally we describe recent developments which started with Mikhail Khovanov work on categorification of the Jones polynomial. By analogy to Khovanov homology we build homology of distributive structures (including homology of Fox colorings) and generalize it to homology of Yang-Baxter operators. We speculate, with supporting evidence, on co-cycle invariants of knots coming from Yang-Baxter homology. Here the work of Fenn-Rourke-Sanderson (geometric realization of pre-cubic sets of link diagrams) and Carter-Kamada-Saito (co-cycle invariants of links) will be discussed and expanded.     Localized abnormalities in the cingulum bundle in patients with schizophrenia: A Diffusion Tensor tractography study

    The cingulum bundle (CB) connects gray matter structures of the limbic system and as such has been implicated in the etiology of schizophrenia. There is growing evidence to suggest that the CB is actually comprised of a conglomeration of discrete sub-connections. The present study aimed to use Diffusion Tensor tractography to subdivide the CB into its constituent sub-connections, and to investigate the structural integrity of these sub-connections in patients with schizophrenia and matched healthy controls. Diffusion Tensor Imaging scans were acquired from 24 patients diagnosed with chronic schizophrenia and 26 matched healthy controls. Deterministic tractography was used in conjunction with FreeSurfer-based regions-of-interest to subdivide the CB into 5 sub-connections (I1 to I5). The patients with schizophrenia exhibited subnormal levels of FA in two cingulum sub-connections, specifically the fibers connecting the rostral and caudal anterior cingulate gyrus (I1) and the fibers connecting the isthmus of the cingulate with the parahippocampal cortex (I4). Furthermore, while FA in the I1 sub-connection was correlated with the severity of patients' positive symptoms (specifically hallucinations and delusions), FA in the I4 sub-connection was correlated with the severity of patients' negative symptoms (specifically affective flattening and anhedonia/asociality). These results support the notion that the CB is a conglomeration of structurally interconnected yet functionally distinct sub-connections, of which only a subset are abnormal in patients with schizophrenia. Furthermore, while acknowledging the fact that the present study only investigated the CB, these results suggest that the positive and negative symptoms of schizophrenia may have distinct neurobiological underpinnings

    Acute Sterol O-Acyltransferase 2 (SOAT2) Knockdown Rapidly Mobilizes Hepatic Cholesterol for Fecal Excretion

    The primary risk factor for atherosclerotic cardiovascular disease is LDL cholesterol, which can be reduced by increasing cholesterol excretion from the body. Fecal cholesterol excretion can be driven by a hepatobiliary as well as a non-biliary pathway known as transintestinal cholesterol efflux (TICE). We previously showed that chronic knockdown of the hepatic cholesterol esterifying enzyme sterol O-acyltransferase 2 (SOAT2) increased fecal cholesterol loss via TICE. To elucidate the initial events that stimulate TICE, C57Bl/6 mice were fed a high cholesterol diet to induce hepatic cholesterol accumulation and were then treated for 1 or 2 weeks with an antisense oligonucleotide targeting SOAT2. Within 2 weeks of hepatic SOAT2 knockdown (SOAT2HKD), the concentration of cholesteryl ester in the liver was reduced by 70% without a reciprocal increase in hepatic free cholesterol. The rapid mobilization of hepatic cholesterol stores resulted in a ∼ 2-fold increase in fecal neutral sterol loss but no change in biliary cholesterol concentration. Acute SOAT2HKD increased plasma cholesterol carried primarily in lipoproteins enriched in apoB and apoE. Collectively, our data suggest that acutely reducing SOAT2 causes hepatic cholesterol to be swiftly mobilized and packaged onto nascent lipoproteins that feed cholesterol into the TICE pathway for fecal excretion
