    Voluntary citizen attention and actions are key to successful public-sector communication. We investigated the conditions which increase such attention and actions using the situational theory of problem solving (STOPS) and government-citizen relationships (GCRs). Using three national issues consisting of an environmental issue, a social issue, and a political issue from South Korea (N=275), this study examined three hypotheses regarding public engagement effect (the effect of GCRs on political conversations on national issues), government empowerment effect (the effects of GCRs and issue-specific trust toward government on constraint recognition), and public serenity effect (the effect of issue-specific trust on problem recognition and involvement recognition). We found significant public engagement and government empowerment effects and partially significant public serenity effect. The results of the public serenity investigation found that issue-specific trust toward government was significant with problem recognition but insignificant with involvement recognition. Consequently, the findings illustrate strategic values in government-citizen relationships on public engagement, empowerment, and serenity to enable participatory democracy

    Improvement of Lipids and Reduction of Oxidative Stress With Octacosanol After Taekwondo Training

    Athletes in combat sports undergo rapid changes in body weight prior to competition in order to gain a size advantage over their opponent. However, these large weight changes with concomitant high-intensity exercise training create poor lipid profiles and high levels of oxidative stress, which can be detrimental to health and sport performance. Therefore, the purpose of this study was to investigate the ability of the nutritional supplement octacosanol to combat the physiological detriments that occur in taekwondo players during rapid weight loss with high-intensity exercise training. Methods: A total of 26 male taekwondo players were randomly divided into 2 groups: An experimental group performed a 5% weight-loss and taekwondo training program with 40-mg octacosanol intake (OCT; n = 13) for 6 d, and a control group performed the same weight-loss and taekwondo training program with a placebo (CON; n = 13). Results: There were significant (P \u3c .05) group × time interactions for low-density lipoprotein and triglycerides, which significantly decreased (Δ18 [5] mg/dL and Δ80 [7] mg/dL, respectively), and high-density lipoprotein, which significantly increased (Δ10 [7] mg/dL), in the OCT group compared with the CON group. There were also significant (P \u3c .05) group × time interactions for superoxide dismutase (SOD), glutathione peroxidase (GPx), and malondialdehyde (MDA), with SOD increasing (Δ226 [121] U/gHb) in the OCT group, while GPx decreased (Δ20 [13] U/gHb) and MDA increased (Δ72 [0.04] nmol/mL) in the CON group. Conclusion: These results suggest that octacosanol may be a beneficial supplement to protect against the poor cholesterol levels and oxidative stress that occurs during taekwondo training

    Cooperative roles of the suprachiasmatic nucleus central clock and the adrenal clock in controlling circadian glucocorticoid rhythm

    The mammalian circadian timing system consists of the central clock in the hypothalamic suprachiasmatic nucleus (SCN) and subsidiary peripheral clocks in other tissues. Glucocorticoids (GCs) are adrenal steroid hormones with widespread physiological effects that undergo daily oscillations. We previously demonstrated that the adrenal peripheral clock plays a pivotal role in circadian GC rhythm by driving cyclic GC biosynthesis. Here, we show that the daily rhythm in circulating GC levels is controlled by bimodal actions of central and adrenal clockwork. When mice were subjected to daytime restricted feeding to uncouple central and peripheral rhythms, adrenal GC contents and steroidogenic acute regulatory protein expression peaked around zeitgeber time 00 (ZT00), consistent with shifted adrenal clock gene expression. However, restricted feeding produced two distinct peaks in plasma GC levels: one related to adrenal GC content and the other around ZT12, which required an intact SCN. Light pulse-evoked activation of the SCN increased circulating GC levels in both wild-type and adrenal clock-disrupted mutant mice without marked induction of GC biosynthesis. In conclusion, we demonstrate that adrenal clock-dependent steroidogenesis and a SCN-driven central mechanism regulating GC release cooperate to produce daily circulatory GC rhythm. © The Author(s) 2017.1

    Novel next generation sequencing panel method for the multiple detection and identification of foodborne pathogens in agricultural wastewater

    Detecting and identifying the origins of foodborne pathogen outbreaks is a challenging. The Next-Generation Sequencing (NGS) panel method offers a potential solution by enabling efficient screening and identification of various bacteria in one reaction. In this study, new NGS panel primer sets that target 18 specific virulence factor genes from six target pathogens (Bacillus cereus, Yersinia enterocolitica, Staphylococcus aureus, Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus) were developed and optimized. The primer sets were validated for specificity and selectivity through singleplex PCR, confirming the expected amplicon size. Crosscheck and multiplex PCR showed no interference in the primer set or pathogenic DNA mixture. The NGS panel analysis of spiked water samples detected all 18 target genes in a single reaction, with pathogen concentrations ranging from 108 to 105 colony-forming units (CFUs) per target pathogen. Notably, the total sequence read counts from the virulence factor genes showed a positive association with the CFUs per target pathogen. However, the method exhibited relatively low sensitivity and occasional false positive results at low pathogen concentrations of 105 CFUs. To validate the detection and identification results, two sets of quantitative real-time PCR (qPCR) analyses were independently performed on the same spiked water samples, yielding almost the same efficiency and specificity compared to the NGS panel analysis. Comparative statistical analysis and Spearman correlation analysis further supported the similarity of the results by showing a negative association between the NGS panel sequence read counts and qPCR cycle threshold (Ct) values. To enhance NGS panel analysis for better detection, optimization of primer sets and real-time NGS sequencing technology are essential. Nonetheless, this study provides valuable insights into applying NGS panel analysis for multiple foodborne pathogen detection, emphasizing its potential in ensuring food safety

    Efekti korišćenja astaksantina u ishrani na rast, pigmentaciju mišića i antioksidantne aktivnosti mlađi kalifornijske pastrmke (oncorhynchus mykiss)

    This study was designed to test the effects of dietary astaxanthin on growth, muscle pigmentation, antioxidant activity and biochemical composition of juvenile rainbow trout (Oncorhynchus mykiss). Experimental diets were formulated to contain 50, 75 and 100 ppm astaxanthin (designed as AS50, AS75 and AS100). The diet without supplementation of astaxanthin was considered as the control diet. Each experimental diet was fed to three replicate groups of fish (18.5 g/fish) to visual satiation two times a day for 10 weeks. Growth performance and proximate composition of muscle of fish were not affected by dietary AS levels (P> 0.05). Total carotenoid concentration in the muscle of fish fed the AS50 diet was higher than that of fish fed the control diet, but no different to that of fish fed the AS75 and AS100 diets. The astaxanthin concentration in the muscle of fish fed AS50, AS75 and AS100 diets were higher than that of control diet. The redness (a*) of the muscle of fish fed AS50, AS75 and AS100 diets were higher than that of fish fed the control diet (P 0.05) nije uticao na performansu rasta i hemijski sastav mišića ribe. Ukupna koncentracija karotenoida u mišiću ribe koja je hranjena AS50 hranom bila je viša nego kod riba hranjenih kontrolnom hranom, ali nije bila drugačija od riba hranjenih AS75 i AS100 hranom. Koncentracija astaksantina u mišiću riba hranjenih AS50, AS75 i AS100 hranom bila je viša nego kod riba hranjenih kontrolnom hranom. Crvena boja (a*) mišića ribe koja je hranjena AS50, AS75 i AS100 hranom bila je jača nego kod ribe hranjene kontrolnom hranom (P< 0.05). Antioksidantna aktivnost DPPH, radikala hidroksila i alkila u plazmi i jetri riba nisu zavisili od nivoa astaksantina osim kod plazme koja je imala antioksidantnu aktivnost alkilnih radikala. Rezultati ove studije nagoveštavaju da se hrana koja sadrži 50 ppm astaksantina može koristiti da bi se poboljšala crvena boja mišične pigmentacije kod mlađi kalifornijske pastrmke
