Stimulatory Effect of 5-Hydroxytryptamine (5-HT) on Rat Capsaicin-Sensitive Lung Vagal Sensory Neurons via Activation of 5-HT\u3csub\u3e3\u3c/sub\u3e Receptors
5-hydroxytryptamine (5-HT) is an inflammatory mediator known to be released in lung. Capsaicin-sensitive lung vagal (CSLV) afferents function as a primary sensor for detecting chemical stimuli and produce consequent reflexes during lung inflammation. To characterize the effect of 5-HT on CSLV afferents, responses of cardiorespiratory reflexes and single-unit C-fiber afferents to right-atrial injections of 5-HT were investigated in anesthetized Sprague-Dawley rats. Bolus injection of 5-HT (8 ÎĽg/kg) caused an immediate augmented breath and apnea, accompanied by hypotension and bradycardia. These initial responses were then followed by a brief pressor response and a more sustained depressor response. After a perineural treatment of both cervical vagi with capsaicin to block the conduction of C fibers, 5-HT still triggered the augmented breath, but no longer evoked the apnea, bradycardia and hypotension, indicating an involvement of C-fiber activation. The remaining augmented breath induced by 5-HT after perineural capsaicin treatment was totally eliminated by vagotomy. To further study the effect of 5-HT on CSLV afferents, activities arising from these afferents were determined using the single-fiber recording technique. Right-atrial injection of 5-HT evoked an intense discharge in CSLV afferents in a dose-dependent manner. The highest dose of 5-HT (16 ÎĽg/kg) activated 79% (19/24) of CSLV afferents which were also sensitive to capsaicin (0.8 ÎĽg/kg). The pretreatment of tropisetron, a selective antagonist of the 5-HT3 receptor, completely blocked CSLV-afferents stimulation induced by 5-HT but did not affect that by capsaicin. Furthermore, a similar afferent response of CSLV afferents was mimicked by phenylbiguanide, a selective agonist of the 5-HT3 receptor. In isolated rat lung vagal C neurons, 5-HT induced intense calcium transients in a dose-dependent manner. The highest concentration (3 ÎĽM) of 5-HT activated 67% (18/27) of the CSLV neurons. The 5-HT-induced response was totally abolished by pretreatment of tropisetron. In conclusion, 5-HT exerts an intense stimulatory effect on lung C-fiber terminals mediated through an activation of the 5-HT3 receptor, which may contribute to the airway hypersensitivity under lung inflammation
Transthyretin Stimulates Tumor Growth through Regulation of Tumor, Immune, and Endothelial Cells
Early detection of lung cancer offers an important opportunity to decrease mortality while it is still treatable and curable. Thirteen secretory proteins that are Stat3 downstream gene products were identified as a panel of biomarkers for lung cancer detection in human sera. This panel of biomarkers potentially differentiates different types of lung cancer for classification. Among them, the transthyretin (TTR) concentration was highly increased in human serum of lung cancer patients. TTR concentration was also induced in the serum, bronchoalveolar lavage fluid, alveolar type II epithelial cells, and alveolar myeloid cells of the CCSP-rtTA/(tetO)7-Stat3C lung tumor mouse model. Recombinant TTR stimulated lung tumor cell proliferation and growth, which were mediated by activation of mitogenic and oncogenic molecules. TTR possesses cytokine functions to stimulate myeloid cell differentiation, which are known to play roles in tumor environment. Further analyses showed that TTR treatment enhanced the reactive oxygen species production in myeloid cells and enabled them to become functional myeloid-derived suppressive cells. TTR demonstrated a great influence on a wide spectrum of endothelial cell functions to control tumor and immune cell migration and infiltration. TTR-treated endothelial cells suppressed T cell proliferation. Taken together, these 13 Stat3 downstream inducible secretory protein biomarkers potentially can be used for lung cancer diagnosis, classification, and as clinical targets for lung cancer personalized treatment if their expression levels are increased in a given lung cancer patient in the blood
Complementary Therapy with Traditional Chinese Medicine for Neonatal Hypoxic Ischemic Encephalopathy
Hypoxic ischemic encephalopathy (HIE) is one of the most significant causes of morbidity, mortality, and lifelong disability in newborns. The diagnosis of neonatal HIE is based on the dysfunction of neurogenic signs and classification according to the Sarnat staging system, which evaluates conscious level, neuromuscular control, complex reflexes, autonomic function, seizures, electroencephalogram readings, and duration of neurologic sign. There is no standard treatment for neonatal HIE, but it is widely accepted that hypothermia therapy is a safe and effective method for treating neonates with HIE. Traditional Chinese medicine (TCM) has recently been used to treat cases of neonatal HIE, especially herbal medicine prescriptions. Acupuncture is a common method used in TCM and is another promising therapy for neonatal HIE due to its demonstrated effective treatment of the disease in animal models. While there is a lack of direct evidence in clinical practice, we have observed acupuncture to be useful in adult HIE and in animal studies; therefore, we believe a clinical trial designed to evaluate the effectiveness of acupuncture in neonatal HIE treatment is worthwhile. Taken together, TCM is a promising technique that can be integrated into the conventional therapies for neonatal HIE
Structural Engineering on Pt-Free Electrocatalysts for Dye-Sensitized Solar Cells
In recent decades, plenty of nanomaterials have been investigated as electrocatalysts for the replacement of the expensive platinum (Pt) counter electrode in dye-sensitized solar cells (DSSCs). The key function of the electrocatalyst is to reduce tri-iodide ions to iodide ions at the electrolyte/counter electrode interface. The performance of the electrocatalyst is usually determined by two key factors, i.e., the intrinsic heterogeneous rate constant and the effective electrocatalytic surface area of the electrocatalyst. The intrinsic heterogeneous rate constant of the electrocatalyst varies by different types of materials, which can be roughly divided into five groups: non-Pt metals, carbons, conducting polymers, transition metal compounds, and their composites. The effective electrocatalytic surface area is determined by the nanostructure of the electrocatalyst. In this chapter, the nanostructural design and engineering on different types of Pt-free electrocatalysts will be systematically introduced. Also, the relationship between various nanostructures of electrocatalysts and the pertinent physical/electrochemical properties will be discussed
The influence of the technical dimension, functional dimension, and tenant satisfaction on tenant loyalty: an analysis based on the theory of planned behavior
This study primarily explored the influence of the technical dimension, functional dimension, and tenant satisfaction on tenant loyalty. The theory of planned behavior served as the basis of this study, and the three aforementioned factors (the technical dimension, the functional dimension, and tenant satisfaction) were incorporated into a conceptual framework for tenant loyalty. Structural equation modeling (SEM) was employed for parameter estimation. The participants consisted of tenants residing in eight administrative districts in Kaohsiung City. 315 questionnaires were administered, all of which were returned. After removing 15 invalid responses, there were 300 valid responses, which indicated an effective recovery rate of 95.2%. The results showed that the technical dimension, the functional dimension, and attitude significantly and positively influenced tenant satisfaction. Tenant satisfaction, perceived behavioral control, and social norms significantly and positively influenced tenant loyalty. Tenant satisfaction mediated the influence of the technical dimension and the functional dimension on tenant loyalty; the mediating effect of the functional dimension on tenant loyalty was greater than that of the technical dimension. The findings of this study highlight the measures rental companies should adopt in order to enhance the technical dimension, functional dimension, and tenant satisfaction, as this is crucial to maintaining sustainable operations
Exposing the Functionalities of Neurons for Gated Recurrent Unit Based Sequence-to-Sequence Model
The goal of this paper is to report certain scientific discoveries about a
Seq2Seq model. It is known that analyzing the behavior of RNN-based models at
the neuron level is considered a more challenging task than analyzing a DNN or
CNN models due to their recursive mechanism in nature. This paper aims to
provide neuron-level analysis to explain why a vanilla GRU-based Seq2Seq model
without attention can achieve token-positioning. We found four different types
of neurons: storing, counting, triggering, and outputting and further uncover
the mechanism for these neurons to work together in order to produce the right
token in the right position.Comment: 9 pages (excluding reference), 10 figure
Virtual Guidance as a Mid-level Representation for Navigation
In the context of autonomous navigation, effectively conveying abstract
navigational cues to agents in dynamic environments poses challenges,
particularly when the navigation information is multimodal. To address this
issue, the paper introduces a novel technique termed "Virtual Guidance," which
is designed to visually represent non-visual instructional signals. These
visual cues, rendered as colored paths or spheres, are overlaid onto the
agent's camera view, serving as easily comprehensible navigational
instructions. We evaluate our proposed method through experiments in both
simulated and real-world settings. In the simulated environments, our virtual
guidance outperforms baseline hybrid approaches in several metrics, including
adherence to planned routes and obstacle avoidance. Furthermore, we extend the
concept of virtual guidance to transform text-prompt-based instructions into a
visually intuitive format for real-world experiments. Our results validate the
adaptability of virtual guidance and its efficacy in enabling policy transfer
from simulated scenarios to real-world ones.Comment: Tsung-Chih Chiang, Ting-Ru Liu, Chun-Wei Huang, and Jou-Min Liu
