    Sigma Model on de Sitter Space

    We discuss spherically symmetric, static solutions to the SU(2) sigma model on a de Sitter background. Despite of its simplicity this model reflects many of the features exhibited by systems of non-linear matter coupled to gravity e.g. there exists a countable set of regular solutions with finite energy; all of the solutions show linear instability with the number of unstable modes increasing with energy.Comment: 11 pages, latex2e, 1 figur

    Building on previous work on the critical behavior in gravitational collapse of the self-gravitating SU(2) σ\sigma-field and using high precision numerical methods we uncover a fine structure hidden in a narrow window of parameter space. We argue that this numerical finding has a natural explanation within a dynamical system framework of critical collapse

    Tracing Functional Antigen-Specific CCR6+ Th17 Cells after Vaccination

    BACKGROUND: The function of T helper cell subsets in vivo depends on their location, and one hallmark of T cell differentiation is the sequential regulation of migration-inducing chemokine receptor expression. CC-chemokine receptor 6 (CCR6) is a trait of tissue-homing effector T cells and has recently been described as a receptor on T helper type 17 (Th17) cells. Th17 cells are associated with autoimmunity and the defence against certain infections. Although, the polarization of Th cells into Th17 cells has been studied extensively in vitro, the development of those cells during the physiological immune response is still elusive. METHODOLOGY/PRINCIPAL FINDINGS: We analysed the development and functionality of Th17 cells in immune-competent mice during an ongoing immune response. In naïve and vaccinated animals CCR6(+) Th cells produce IL-17. The robust homeostatic proliferation and the presence of activation markers on CCR6(+) Th cells indicate their activated status. Vaccination induces antigen-specific CCR6(+) Th17 cells that respond to in vitro re-stimulation with cytokine production and proliferation. Furthermore, depletion of CCR6(+) Th cells from donor leukocytes prevents recipients from severe disease in experimental autoimmune encephalomyelitis, a model for multiple sclerosis in mice. CONCLUSIONS/SIGNIFICANCE: In conclusion, we defined CCR6 as a specific marker for functional antigen-specific Th17 cells during the immune response. Since IL-17 production reaches the highest levels during the immediate early phase of the immune response and the activation of Th17 cells precedes the Th1 cell differentiation we tent to speculate that this particular Th cell subset may represent a first line effector Th cell subpopulation. Interference with the activation of this Th cell subtype provides an interesting strategy to prevent autoimmunity as well as to establish protective immunity against infections

    Religious socialisation and fertility: transition to third birth in the Netherlands

    Although previous studies have demonstrated that religious people in Europe have larger families, the role played by religious socialisation in the context of contemporary fertility behaviour has not yet been analysed in detail. This contribution specifically looks at the interrelation between religious socialisation and current religiosity and their impact on the transition to the third child for Dutch women. It is based on data of the first wave of the Netherlands Kinship Panel Study (2002–2004) and uses event history analysis. The transitions to first, second and third birth are modelled jointly with a control for unobserved heterogeneity. The findings provide evidence for an impact of women’s current church attendance as well as religious socialisation measured by their fathers’ religious affiliation, when they were teenagers. A religious family background remains influential even when a woman has stopped attending church. The effects of religious indicators strengthen over cohorts. Moreover, the combined religious make-up of the respondent’s parents also significantly determines the progression to the third child.S’il est bien établi que les croyants en Europe ont plus d’enfants que les autres, le rôle de la socialisation religieuse dans le contexte de la fécondité contemporaine n’a pas encore été analysé à ce jour. Cette étude s’intéresse au lien entre la socialisation religieuse et la religiosité actuelle, et à leur impact sur la probabilité d’agrandissement de deux à trois enfants de la descendance des femmes néerlandaises. Les données exploitées sont celles de la première vague du Panel Néerlandais d’Etude de la Parenté (the Netherlands Kinship Panel Study, 2002–2004). A l’aide des techniques de l’analyse des biographies, les probabilités d’agrandissement de rang 1, rang 2 et rang 3 ont été modélisées de façon conjointe, en contrôlant l’hétérogénéité non observée. Les résultats mettent en évidence l’impact de la fréquentation actuelle de l’église par les femmes et de leur socialisation religieuse, mesurée par l’appartenance religieuse de leur père quand elles étaient adolescentes. Il apparaît que la religiosité du contexte familial exerce une influence, même quand la femme ne fréquente plus l’église, et que les effets des indicateurs de pratique religieuse se renforcent d’une génération à l’autre. Enfin, l’appartenance religieuse conjointe des parents de la femme détermine significativement la probabilité d’avoir un troisième enfant

    Cell cycle-dependent acetylation of Rb2/p130 in NIH3T3 cells

    The retinoblastoma protein (pRb) and the pRb-related proteins, p130 and p107, form the ‘pocket protein' family of cell cycle regulatory factors. A well characterized function of these proteins is the cell cycle-dependent regulation of E2F-responsive genes. The biological activity of pocket proteins is regulated by phosphorylation and for the founding member pRb it has been shown that acetylation also has an important role in modulating its function during the cell cycle. Here, we show that hyperphosphorylated retinoblastoma 2 (Rb2)/p130 also exists in an acetylated form in NIH3T3 cells. Acetylated p130 is present in the nucleus but not in the cytoplasm. Acetylation is cell cycle dependent, starting in S-phase and persisting until late G2-period. Using recombinant p130 and truncated forms for in vitro acetylation by the acetyltransferase p300, we could identify K1079 in the C-terminal part as the major acetylation site by mass spectrometry. Minor acetylation sites were pinpointed to K1068 and K1111 in the C-terminus, and K128 and K130 in the N-terminus.     Atomoxetine Enhances Connectivity of Prefrontal Networks in Parkinson's Disease.

    Cognitive impairment is common in Parkinson's disease (PD), but often not improved by dopaminergic treatment. New treatment strategies targeting other neurotransmitter deficits are therefore of growing interest. Imaging the brain at rest ('task-free') provides the opportunity to examine the impact of a candidate drug on many of the brain networks that underpin cognition, while minimizing task-related performance confounds. We test this approach using atomoxetine, a selective noradrenaline reuptake inhibitor that modulates the prefrontal cortical activity and can facilitate some executive functions and response inhibition. Thirty-three patients with idiopathic PD underwent task-free fMRI. Patients were scanned twice in a double-blind, placebo-controlled crossover design, following either placebo or 40-mg oral atomoxetine. Seventy-six controls were scanned once without medication to provide normative data. Seed-based correlation analyses were used to measure changes in functional connectivity, with the right inferior frontal gyrus (IFG) a critical region for executive function. Patients on placebo had reduced connectivity relative to controls from right IFG to dorsal anterior cingulate cortex and to left IFG and dorsolateral prefrontal cortex. Atomoxetine increased connectivity from the right IFG to the dorsal anterior cingulate. In addition, the atomoxetine-induced change in connectivity from right IFG to dorsolateral prefrontal cortex was proportional to the change in verbal fluency, a simple index of executive function. The results support the hypothesis that atomoxetine may restore prefrontal networks related to executive functions.
