    TURBIDITY REMOVAL BY MUCILAGE FROM BASELLA ALBA

    Coagulation and flocculation are preliminary used in water treatment processes for turbidity removal, using popularily synthetic chemicals with health and environmental concerns. The use of natural flocculants has known to be a promising alteratives for chemical ones due to their environmental friendly behavior. This research investigated turbidity removal efficiency of mucilage extracted from Basella alba - an indigenous species in Vietnam - in the role of a flocculant. The removal efficiency of mucilage was investigated in combination with PAC or Alum on To Lich river water by mean of Jar tests. PAC or Alum alone can remove maximum 97% and 90% turbidity of To Lich river water at its original pH for the sedimentation time of 30 minutes. The combination of mucilage and PAC or Alum increased the efficiencies of turbidity removal and reduced the amount of chemicals needed. The corresponding increases were maximum 7% and 18%, respectively; while the reduction of PAC/Alum used was 75-80%

    Conservation, Exploitation And Use Of The Ginger Family (Zingiberaceae) At The National Genebank In Vietnam

    There are 741 accessions in the ginger family such as turmeric, ginger, galangal, and pinecone ginger collections conserved at Plant Resources Center, Vietnam. These collections were highly diverse in the number of accessions and species composition. The results of the Zingiberaceae exploitation shown the G10 ginger variety plants reached around 63.7 - 77.5 cm in height, 400 - 500 g/clumps in fresh weight, 25 - 27 tonnes/ha in fresh yield. The G10 ginger variety contained 4.37% oil, 1.2 mg/kg Zn and 9.31 mg/100g Vitamin C. This G10 ginger variety resistance to leaf spot and green aphids. In addition, the N8 turmeric variety plants reached 130 - 170 cm in height; 880 - 1000 g/clumps in fresh weight and 35 tonnes/ha in yield, curcumin content 6.2 - 6.6%, essential oil contents 2.5 - 2.7%. This turmeric resistance to heat stress, leaf spot, and green aphids. The conserved root and tuber crops at PRC are very diverse both of the amount of sample in collections and species. Conservation and evaluation on root and tuber crops reviewed the promising G10 ginger and N8 turmeric varieties had high yield, good oil content to release for production

    Cloning and expression of gene FanC-2NT encoding K99-2NT fimbrial antigen of enterotoxigenic Escherichia coli from diarrheic post-weaning piglets

    Background and Purpose: The K99 (F5) is one pilus adhesin that mediates the attachment of enterotoxigenic E. coli (ETEC) strains to small intestines to cause to diarrhea in piglets, lambs and newborn calves. In this work, we carried out cloning and expression of the mature peptide of FanC subunit, K99 fimbriae, one of the most common adhesive antigens in E. coli. Materials and Methods: E. coli 2NT strain was isolated from fecal samples of post-weaning piglets with diarrhea. The coding sequence of the mature peptide of K99-2NT subunit was isolated by PCR amplification and cloned into pGEM®-T Easy vector for sequencing using fluorescent dideoxy-terminator method. Expression of K99-2NT protein which was inserted into pET200/D-TOPO vector induced with IPTG. The PCR product and expression level of protein was examined by agarose gel electrophoresis and sodium dodecyl sulfate-polyacrylamide gel electrophoresis, respectively. Results and Conclusions: We cloned and expressed successfully the mature peptide of K99 subunit with molecular weight of approximately 17.5 kDa from E. coli 2NT strain (named K99-2NT). Nucleotide sequence of the K99-2NT subunit coding region of fanC-2NT gene is 477 bp in length and is 99% similarity with that of fanC gene (accession no: M35282). Highest expression level occurred after 12 h of induction with 0.75 mM IPTG at 37oC. This subunit antigen will be tested for immune response of rat in the next time

    Pedagogical Challenges in Online Teaching: A Qualitative Study on Vietnamese University Teachers of Social Sciences

    In recent years, online teaching has gained significant traction in higher education institutions across the globe, primarily due to the widespread impact of the Covid-19 pandemic. Before the pandemic, online teaching was mainly associated with distance learning which aims to provide further education for in-service learners. In this context, our study aims to explore the pedagogical obstacles encountered by Vietnamese university teachers when implementing online teaching in social sciences. Employing qualitative research methods, such as snowball sampling and semi-structured in-depth interviews, the researchers gathered data from 23 university teachers specializing in social sciences across 12 public and private universities in Ho Chi Minh City, Vietnam. The findings of this investigation shed light on various pedagogical barriers that impede the teaching process, encompassing aspects such as lesson plan preparation, lecture delivery (limited interactions, teaching methods and activities, students’ distraction and passive participation, managing virtual classrooms), student assessment, and overall teaching ineffectiveness. This study contributes to the existing literature on contextualized online instructions in higher education, specifically focusing on the social sciences domain. Furthermore, the discussion delves into the critical points, providing a comprehensive understanding of the challenges faced by educators in the online teaching landscape

    Comparative study on vision based rice seed varieties identification

    Abstract-This paper presents a system for automated classification of rice variety for rice seed production using computer vision and image processing techniques. Rice seeds of different varieties are visually very similar in color, shape and texture that make the classification of rice seed varieties at high accuracy challenging. We investigated various feature extraction techniques for efficient rice seed image representation. We analyzed the performance of powerful classifiers on the extracted features for finding the robust one. Images of six different rice seed varieties in northern Vietnam were acquired and analyzed. Our experiments have demonstrated that the average accuracy of our classification system can reach 90.54% using Random Forest method with a simple feature extraction technique. This result can be used for developing a computer-aided machine vision system for automated assessment of rice seeds purity

    GENETIC POLYMORPHISM OF 23 Y-CHROMOSOME SHORT TANDEM REPEAT LOCI IN THE KINH POPULATION OF VIETNAM

    Y-chromosome microsatellites or short tandem repeats (STRs) have been proved to be ideal markers to delineate the differences between individuals in human population. Nowadays, Y-STR testing using the PowerPlex® Y23 amplification kit is considered as an extremely sensitive analysis method and has the potential to be used to perform forensic caseworks, and to explore the complexity in population substructures. However, little is known about the forensic Y-chromosome databases in the Vietnam population. In this study, 23 Y-STR loci (DYS576, DYS389I, DYS389 II, DYS448, DYS19, DYS391, DYS481, DYS549, DYS533, DYS438, DYS437, DYS570, DYS635, DYS390, DYS439, DYS392, DYS393, DYS458 DYS456, DYS643, YGATAH4, and DYS385a/b) were investigated in 120 non-related males of the Kinh population in Northern Vietnam using PowerPlex® Y23 system kit (Promega). Our results showed that allele frequencies of 23 loci in the sample population, with the calculated average gene diversity (GD) for each locus, ranged from 0.24 (DYS438) to 0.92 (DYS385a/b). In addition, a total of 120 different haplotypes were found, all of them were unique. Therefore, we found that the haplotype diversity was 1 with a discrimination capacity of 100%, which serves as an essential prerequisite for using Y-chromosomal STR with PowerPlex® Y23 System kit in forensic application in Vietnam. We also compared genetic distances between Kinh population and 10 other neighboring populations from Y-chromosome haplotype reference database (YHRD). The Kinh population is significantly different from other populations. In conclusion, it was indicated that the 23 Y-STR loci were highly genetically polymorphic in the Kinh population in Vietnam and might be of great value in forensic application

    Orange Peel Essential Oil Nanoemulsions Supported by Nanosilver for Antibacterial Application

    This study is devoted to the synthesis of King Orange peel essential oil in water nanoemulsions combined with nanosilver by ultrasonic method supported by mechanical stirring for antibacterial application. The samples were characterized by Gas chromatography-mass spectrometry (GC-MS), Ultraviolet–visible (UV-Vis), Transmission electron microscopy (TEM), and Dynamic light scattering (DLS) methods. According to the above-experimental results, it was found that the emulsions based on orange essential oil (without nanosilver) and the ones combined with nanosilver have the average particle size from 66.2 nm and 42.9 nm, respectively. The obtained nanoemulsions were also tested to examine their antibacterial ability against Escherichia coli (E. coli) by the disc diffusion method. It has been also found that nanoemulsions based on orange essential oil supported by nanosilver have the superior antibacterial ability, compared with individual components of the materials

    Assessment of Antioxidant and Antibacterial Activities of Lactobacillus farciminis HN11 as a Probiotic Candidate

    Background and Objective: Lactobacillus farciminis is an obligate homofermentative bacterial species in fermented foods. Although other species such as Lactobacillus plantarum, Lactobacillus acidophilus, Lactobacillus casei and Lactobacillus rhamnosus in Lactobacillus genus have been well characterized, probiotic characteristics of Lactobacillus farciminis still need to investigate. Thus, the objective of this study was to investigate probiotic characteristics and antibacterial activity of Lactobacillus farciminis HN11 isolated from Solanum macrocarpon sauces in Hue city, Vietnam. Material and Methods: Lactobacillus farciminis HN11 was cultured in Rogosa and Sharpe media and antibacterial activity of the free-cell suspension was assessed against Escherichia coli, Vibrio parahaemolyticus K5, Vibrio parahaemolyticus KS-02, Vibrio owensii KS-05, Vibrio alginolyticus KS-08, Vibrio alginolyticus A1-1 and Staphylococcus aureus using agar well diffusion method. Various probiotic characteristics of the isolate including antibacterial and antioxidant activities, autoaggregation and coaggregation were assessed. Furthermore, hemolytic and amino-acid dercarboxylase activities were assessed for biosafety assessment. The strain abilities to resist sodium chloride and bile salts were assessed as well. Results and Conclusion: Lactobacillus farciminis HN11 exhibited significant resistance to NaCl and bile salts. The strain expressed high coaggregation abilities for Escherichia coli, significant autoaggregation abilities and antibacterial activities against Vibrio spp. and Escherichia coli. Antioxidant assessment showed that Lactobacillus farciminis HN11 contained high antioxidant activities. This strain was negative for hemolytic and amino-acid dercarboxylase activities. Moreover, ampicillin and chloramphenicol inhibited growth of Lactobacillus farciminis HN11. This study assessed characteristics of Lactobacillus farciminis HN11 and showed its great potentials as a probiotic in fermented foods, enhancing antioxidant and decreasing harmful foodborne bacteria. Although, encapsulation of the strain within acid resistance materials is suggested to better protect it against high-acid contents of the stomach.     A Novel Thermostable Cytochrome P450 from Sequence-Based Metagenomics of Binh Chau Hot Spring as a Promising Catalyst for Testosterone Conversion

    Biotechnological applications of cytochromes P450 show difficulties, such as low activity, thermal and/or solvent instability, narrow substrate specificity and redox partner dependence. In an attempt to overcome these limitations, an exploitation of novel thermophilic P450 enzymes from nature via uncultured approaches is desirable due to their great advantages that can resolve nearly all mentioned impediments. From the metagenomics library of the Binh Chau hot spring, an open reading frame (ORF) encoding a thermostable cytochrome P450—designated as P450-T3—which shared 66.6% amino acid sequence identity with CYP109C2 of Sorangium cellulosum So ce56 was selected for further identification and characterization. The ORF was synthesized artificially and heterologously expressed in Escherichia coli C43(DE3) using the pET17b system. The purified enzyme had a molecular weight of approximately 43 kDa. The melting temperature of the purified enzyme was 76.2 ◦C and its apparent half-life at 60 ◦C was 38.7 min. Redox partner screening revealed that P450-T3 was reduced well by the mammalian AdR-Adx4-108 and the yeast Arh1-Etp1 redox partners. Lauric acid, palmitic acid, embelin, retinoic acid (all-trans) and retinoic acid (13-cis) demonstrated binding to P450-T3. Interestingly, P450-T3 also bound and converted testosterone. Overall, P450-T3 might become a good candidate for biocatalytic applications on a larger scale

    Quantifying antimicrobial access and usage for paediatric diarrhoeal disease in an urban community setting in Asia.

    OBJECTIVES: Antimicrobial-resistant infections are a major global health issue. Ease of antimicrobial access in developing countries is proposed to be a key driver of the antimicrobial resistance (AMR) epidemic despite a lack of community antimicrobial usage data. METHODS: Using a mixed-methods approach (geospatial mapping, simulated clients, healthcare utilization, longitudinal cohort) we assessed antimicrobial access in the community and quantified antimicrobial usage for childhood diarrhoea in an urban Vietnamese setting. RESULTS: The study area had a pharmacy density of 15.7 pharmacies/km2 (a pharmacy for every 1316 people). Using a simulated client method at pharmacies within the area, we found that 8% (3/37) and 22% (8/37) of outlets sold antimicrobials for paediatric watery and mucoid diarrhoea, respectively. However, despite ease of pharmacy access, the majority of caregivers would choose to take their child to a healthcare facility, with 81% (319/396) and 88% (347/396) of responders selecting a specialized hospital as one of their top three preferences when seeking treatment for watery and mucoid diarrhoea, respectively. We calculated that at least 19% (2688/14427) of diarrhoea episodes in those aged 1 to <5 years would receive an antimicrobial annually; however, antimicrobial usage was almost 10 times greater in hospitals than in the community. CONCLUSIONS: Our data question the impact of community antimicrobial usage on AMR and highlight the need for better education and guidelines for all professionals with the authority to prescribe antimicrobials
