Long-term in vitro 3D hydrogel co-culture model of inflammatory bowel disease
The in vitro study of the pathogenesis of inflammatory bowel disease (IBD) requires a cell model which closely reflects the characteristics of the in vivo intestinal epithelium. This study aimed to investigate the application of L-pNIPAM hydrogel as a scaffold to develop a long-term 3D co-culture model of Caco-2 and HT29-MTX cells under conditions analogous to inflammation, to determine its potential use in studying IBD. Monocultures and co-cultures were layered on L-pNIPAM hydrogel scaffolds and maintained under dynamic culture conditions for up to 12 weeks. Treatments with IL-1β, TNFα, and hypoxia for 1 week were used to create an inflammatory environment. Following prolonged culture, the metabolic activity of Caco-2 monoculture and 90% Caco-2/10% HT29-MTX co-cultures on L-pNIPAM hydrogels were increased, and finger-like structures, similar in appearance to villi were observed. Following treatment with IL-1β, TNFα and hypoxia, ALP and ZO-1 were decreased, MUC2 increased, and MUC5AC remained unchanged. ADAMTS1 was increased in response to hypoxia. Caspase 3 expression was increased in response to TNFα and hypoxic conditions. In conclusion, L-pNIPAM hydrogel supported long-term co-culture within a 3D model. Furthermore, stimulation with factors seen during inflammation recapitulated features seen during IBD
A four-season prospective study of muscle strain reoccurrences in a professional football club
The aim of this investigation was to characterise muscle strain reinjuries and examine their impact on playing resources in a professional football club. Muscle strains and reoccurrences were prospectively diagnosed over four seasons in first-team players (n = 46). Altogether, 188 muscle strains were diagnosed with 44 (23.4%) of these classed as reinjuries, leading to an incidence of 1.32 strain reoccurrences per 1,000 hours exposure (95% Confidence Interval [CI], 0.93–1.71). The incidence of recurrent strains was higher in match-play compared with training (4.51, 95% CI, 2.30–6.72 vs 0.94, 95% CI, 0.59–1.29). Altogether, 50.0% of players sustained at least 1 reoccurrence of a muscle strain, leading to approximately 3 days lost and 0.4 matches missed per player per season. The incidence of recurrent strains was highest in centre-forwards (2.15, 95% CI, 1.06–3.24), peaked in May (3.78, 95% CI, 0.47–7.09), and mostly affected the hamstrings (38.6% of all reoccurrences). Mean layoff for nonreoccurrences and recurrences was similar: ∼7.5 days. These results provide greater insight into the extent of the problem of recurrent muscle strains in professional football
High Metallicity Mg II Absorbers in the z < 1 Lyman alpha Forest of PKS 0454+039: Giant LSB Galaxies?
We report the discovery of two iron-group enhanced high-metallicity Mg II
absorbers in a search through 28 Lyman Alpha forest clouds along the PKS
0454+039 sight line. Based upon our survey and the measured redshift number
densities of W_r(MgII) <= 0.3 A absorbers and Lyman Alpha absorbers at z ~ 1,
we suggest that roughly 5% of Lyman Alpha absorbers at z < 1 will exhibit
"weak" Mg II absorption to a 5-sigma W_r(2796) detection limit of 0.02 A. The
two discovered absorbers, at redshifts z = 0.6248 and z = 0.9315, have W_r(Lya)
= 0.33 and 0.15 A, respectively. Based upon photoionization modeling, the H I
column densities are inferred to be in the range 15.8 <= log N(HI) <= 16.8
cm^-2. For the z = 0.6428 absorber, if the abundance pattern is solar, then the
cloud has [Fe/H] > -1; if its gas-phase abundance follows that of depleted
clouds in our Galaxy, then [Fe/H] > 0 is inferred. For the z = 0.9315 absorber,
the metallicity is [Fe/H] > 0, whether the abundance pattern is solar or
suffers depletion. Imaging and spectroscopic studies of the PKS 0454+039 field
reveal no candidate luminous objects at these redshifts. We discuss the
possibility that these Mg II absorbers may arise in the class of "giant" low
surface brightness galaxies, which have [Fe/H] >= -1, and even [Fe/H] >= 0, in
their extended disks. We tentatively suggest that a substantial fraction of
these "weak" Mg II absorbers may select low surface brightness galaxies out to

AEGIS: Enhancement of Dust-enshrouded Star Formation in Close Galaxy Pairs and Merging Galaxies up to z ~ 1
Using data from the DEEP2 Galaxy Redshift Survey and HST/ACS imaging in the
Extended Groth Strip, we select nearly 100 interacting galaxy systems including
kinematic close pairs and morphologically identified merging galaxies. Spitzer
MIPS 24 micron fluxes of these systems reflect the current dusty star formation
activity, and at a fixed stellar mass (M_{*}) the median infrared luminosity
(L_{IR}) among merging galaxies and close pairs of blue galaxies is twice (1.9
+/- 0.4) that of control pairs drawn from isolated blue galaxies. Enhancement
declines with galaxy separation, being strongest in close pairs and mergers and
weaker in wide pairs compared to the control sample. At z ~ 0.9, 7.1% +/- 4.3%
of massive interacting galaxies (M_{*} > 2*10^{10} M_{solar}) are found to be
ULIRGs, compared to 2.6% +/- 0.7% in the control sample. The large spread of IR
luminosity to stellar mass ratio among interacting galaxies suggests that this
enhancement may depend on the merger stage as well as other as yet unidentified
factors (e.g., galaxy structure, mass ratio, orbital characteristics, presence
of AGN or bar). The contribution of interacting systems to the total IR

Functional plasticity of antibacterial EndoU toxins.
Bacteria use several different secretion systems to deliver toxic EndoU ribonucleases into neighboring cells. Here, we present the first structure of a prokaryotic EndoU toxin in complex with its cognate immunity protein. The contact-dependent growth inhibition toxin CdiA-CTSTECO31 from Escherichia coli STEC_O31 adopts the eukaryotic EndoU fold and shares greatest structural homology with the nuclease domain of coronavirus Nsp15. The toxin contains a canonical His-His-Lys catalytic triad in the same arrangement as eukaryotic EndoU domains, but lacks the uridylate-specific ribonuclease activity that characterizes the superfamily. Comparative sequence analysis indicates that bacterial EndoU domains segregate into at least three major clades based on structural variations in the N-terminal subdomain. Representative EndoU nucleases from clades I and II degrade tRNA molecules with little specificity. In contrast, CdiA-CTSTECO31 and other clade III toxins are specific anticodon nucleases that cleave tRNAGlu between nucleotides C37 and m2 A38. These findings suggest that the EndoU fold is a versatile scaffold for the evolution of novel substrate specificities. Such functional plasticity may account for the widespread use of EndoU effectors by diverse inter-bacterial toxin delivery systems
Production of Biodiesel from Vietnamese Waste Coffee Beans: Biofuel Yield, Saturation and Stability are All Elevated Compared with Conventional Coffee Biodiesel
The suitability of biodiesel produced from spent Vietnamese coffee was examined. Previous work shows that the geographical origin of coffee beans has little effect on the composition and physical properties of the biodiesel produced Jenkins et al. [1]. Vietnamese coffee, however, is roasted in a range of fats and oils for flavour enhancement and therefore has a unique fatty acid profile. The oil yield and biodiesel properties of three Vietnamese coffees were assessed and compared to a coffee of more typical composition—Colombian—and traditional biodiesel feedstocks (rapeseed, sunflower and palm). The oil yield from fresh Vietnamese coffee was higher (12.0–14.0 %) than Colombian coffee (9.3 %), while the oil yield from spent Vietnamese coffee (9.3–10.4 %) was comparable to the Colombian coffee (9.5 %). The unsaponifiable matter was only present in low levels in the Vietnamese coffee (1.9–4.9 %) compared to Colombian coffee (30.4 % fresh, 21.4 % spent). Vietnamese coffee biodiesel was more saturated than Columbian coffee biodiesel. It was therefore more viscous and had a higher pour point than the Colombian coffee, and possessed properties more akin to palm biodiesel. Vietnamese coffee biodiesel would therefore be a suitable feedstock for use locally due to the more suitable climate and compatibility with the palm feedstock that is currently used
Asthma control in severe asthma and occupational exposures to inhalable asthmagens
INTRODUCTION: Work-related asthma accounts for ≥25% of asthma in working-age populations, though the relationship between work exposures and symptoms is frequently missed, leading to poor health and employment outcomes. We hypothesised that inhalable exposures at work are associated with poor asthma control in severe asthma (SA).METHODS: We searched the Birmingham (UK) Regional NHS SA Service clinical database (n=1453 records; 1 March 2004 to 1 March 2021) and undertook a cross-sectional study using baseline data collected at diagnosis. We included all employed patients aged 16-64 with documented current occupation (n=504), and collected socio-demographic, general health and asthma-specific data, including Asthma Control Questionnaire 7 (ACQ7) score. The Occupational Asthma Specific Job-Exposure Matrix (OAsJEM) was employed to determine the likelihood of exposure to respiratory sensitisers, irritants, cleaning agents and detergents; associations between exposures and ACQ7 were investigated using binary and multinomial regression.RESULTS: Frequently reported occupations were care assistants (7%) and nurses (6%); 197/504 (39%) patients were exposed to an asthmagen, including respiratory sensitisers (30%), airway irritants (38%) and cleaning products/disinfectants (29%). ACQ7 score was available for 372/504 (74%) patients, of whom 14% had adequate control (ACQ7=0-1.5). After adjustment for major confounders there were no significant associations between inhaled asthmagens and ACQ7 score (either as binary or multinomial outcomes).CONCLUSION: JEM-determined workplace exposures to inhaled asthmagens are not associated with asthma control in SA; 29-39% of patients may have current exposure to workplace asthmagens. Routine collection of lifetime occupational data including current job role and level of exposure, in the national asthma registry, would give further insights into this relationship.</p
Anodised TiO2 nanotubes as a scaffold for antibacterial silver nanoparticles on titanium implants

All rights reserved.Medical grade titanium alloy is widely used for bone/dental implants, but the material alone has no innate antimicrobial properties that would reduce infection risk following surgery. However, silver nanoparticles (Ag NPs) are known to be antibacterial. This study investigated the growth of Ag NPs on titanium dioxide nanotubes (TiO2 NTs) on Ti-6Al-4V discs. The TiO2 NTs were grown on the Ti alloy using an electrochemical method, and then decorated with Ag NPs. The Ag NPs were synthesised by chemical reduction using δ-gluconolactone. A silver ammonia solution (silver nitrate + liquid ammonia) was used as the source of silver. Two separate approaches were used: (1) The δ-gluconolactone was mixed with the silver ammonia and then exposed to the TiO2 NTs (the 'mixing method'), which produced micron-sized clusters of the Ag NPs. (2) The TiO2 NTs were exposed to the silver ammonia first and then to δ-gluconolactone (the 'sequential addition method'), which resulted in the formation of nano-sized clusters of the nanoparticles. The Ag-TiO2 composites were confirmed by scanning electron microscopy and the elements analysed using energy dispersive X-ray spectroscopy (EDS). The composite coatings were exposed to a simulated body fluid for 24 h in order to determine the total Ag released. The release from the micron-sized clusters from the mixing method (14.6 ± 0.67 ppm) was higher than that from the nano-sized clusters (4.05 ± 0.36 ppm) when 0.015 M of silver ammonia was used. Additionally, Staphylococcus aureus, was cultured on the composite coatings for 24 h. Both the micron- and nano-sized clusters of the Ag NPs were found to be antibacterial using the Live/Dead assay. Overall, δ-gluconolactone was successfully used to reduce silver to Ag NPs on the surface of TiO2 NTs. The sequential addition method was the preferred method of synthesis because of its slower silver release, better coverage of the Ag-NPs on the TiO2 NTs and strong antibacterial properties
Investigating the Effect of Carbon Nanotube Diameter and Wall Number in Carbon Nanotube/Silicon Heterojunction Solar Cells

(CC BY 4.0).Suspensions of single-walled, double-walled and multi-walled carbon nanotubes (CNTs) were generated in the same solvent at similar concentrations. Films were fabricated from these suspensions and used in carbon nanotube/silicon heterojunction solar cells and their properties were compared with reference to the number of walls in the nanotube samples. It was found that single-walled nanotubes generally produced more favorable results; however, the double and multi-walled nanotube films used in this study yielded cells with higher open circuit voltages. It was also determined that post fabrication treatments applied to the nanotube films have a lesser effect on multi-walled nanotubes than on the other two types
High stakes and low bars: How international recognition shapes the conduct of civil wars
When rebel groups engage incumbent governments in war for control of the state, questions of international recognition arise. International recognition determines which combatants can draw on state assets, receive overt military aid, and borrow as sovereigns—all of which can have profound consequences for the military balance during civil war. How do third-party states and international organizations determine whom to treat as a state's official government during civil war? Data from the sixty-one center-seeking wars initiated from 1945 to 2014 indicate that military victory is not a prerequisite for recognition. Instead, states generally rely on a simple test: control of the capital city. Seizing the capital does not foreshadow military victory. Civil wars often continue for many years after rebels take control and receive recognition. While geopolitical and economic motives outweigh the capital control test in a small number of important cases, combatants appear to anticipate that holding the capital will be sufficient for recognition. This expectation generates perverse incentives. In effect, the international community rewards combatants for capturing or holding, by any means necessary, an area with high concentrations of critical infrastructure and civilians. In the majority of cases where rebels contest the capital, more than half of its infrastructure is damaged or the majority of civilians are displaced (or both), likely fueling long-term state weakness
