CCAAT/enhancer-binding proteins are key regulators of human type two deiodinase expression in a placenta cell line
An appropriate concentration of intracellular T(3) is a critical determinant of placenta development and function and is mainly controlled by the activity of type II deiodinase (D2). The levels of this enzyme are finely regulated in different tissues by coordinated transcriptional mechanisms, which rely on dedicated promoter sequences (e.g. cAMP response element and TATA elements) that impart inducibility and tissue specificity to Dio2 mRNA expression. Here we show that CCAAT enhancer-binding proteins α and β (C/EBPα and C/EBPβ) promote Dio2 expression in the trophoblastic cell line JEG3 through a conserved CCAAT element, which is a novel key component of the Dio2 promoter code that confers tissue-specific expression of D2 in these cells. Increased C/EBPs levels potently induce Dio2 transcription, whereas their ablation results in loss of Dio2 mRNA. By measuring the activity of several deletion and point mutant promoter constructs, we have identified the functional CCAAT element responsible for this effect, which is located in close proximity to the most 5' TATA box. Notably, this newly identified sequence is highly conserved throughout the species and binds in vivo and in vitro C/EBP, indicating the relevance of this regulatory mechanism. Together, our results unveil a novel mechanism of regulation of D2 expression in a trophoblastic cell line, which may play a relevant role during placenta development
Efeitos da ginástica abdominal hipopressiva no puerpério imediato - estudo de casos
Algumas modificações fisiológicas inerentes do período gestacional e parto podem perdurar no organismo materno, acarretando possíveis complicações em longo prazo. Objetivo: o objetivo do presente estudo foi verificar os efeitos da ginástica abdominal hipopressiva (GAH) no puerpério imediato. Método: trata-se de um estudo de caso com duas puérperas internadas em uma maternidade pública. Realizou-se mensuração da diástase do músculo reto abdominal (DMRA), cirtometria torácica e perimetria abdominal, antes e após o protocolo de GAH, aplicado dentro das 48 horas de internação. Resultados: após duas aplicações do protocolo de GAH, houve redução da DMRA e alterações na expansibilidade torácica e perímetro abdominal, porém não conclusivo devido o número limitado de participantes. Considerações finais: a GAH é um recurso fisioterapêutico que pode ser utilizado no puerpério imediato, atuando precocemente nas alterações decorrentes da gestação e parto, prevenindo posteriores complicações

Losartan counteracts the hyper-reactivity to angiotensin II and ROCK1 over-activation in aortas isolated from streptozotocin-injected diabetic rats
<p>Abstract</p> <p>Background</p> <p>In streptozotocin-injected rats (STZ-rats), we previously demonstrated a role for angiotensin II (AT-II) in cardiac remodelling and insulin resistance partially counteracted by <it>in vivo </it>treatment with losartan, an AT-II receptor antagonist.</p> <p>We now aimed to investigate the effect of treating diabetic STZ-rats with losartan on diabetes vascular response to vasoconstrictors.</p> <p>Methods</p> <p>Male Wistar rats were randomly divided in four groups, two of them were assigned to receive losartan in the drinking water (20 mg/kg/day) until the experiment ending (3 weeks afterward). After 1 week, two groups, one of which receiving losartan, were injected in the tail vein with citrate buffer (normoglycemic, N and normoglycemic, losartan-treated, NL). The remaining received a single injection of streptozotocin (50 mg/kg in citrate i.v.) thus becoming diabetic (D) and diabetic losartan-treated (DL). Plasma glycaemia and blood pressure were measured in all animals before the sacrifice (15 days after diabetes induction).</p> <p>In aortic strips isolated from N, NL, D and DL rats we evaluated i) the isometric concentration-dependent contractile response to phenylephrine (Phe) and to AT-II; ii) the RhoA-kinase (ROCK1) activity and expression by enzyme-immunoassay and Western blot respectively.</p> <p>Key results</p> <p>The concentration-dependent contractile effect of Phe was similar in aortas from all groups, whereas at all concentrations tested, AT-II contraction efficacy was 2 and half and 1 and half times higher in D and DL respectively in comparison with N and NL. AT-II contracture was similarly reduced in all groups by AT-II receptor antagonists, irbesartan or irbesartan plus PD123319. HA-1077 (10 μM), an inhibitor of ROCK1 activity, reduced AT-II efficacy (Δmg/mg tissue w.w.) by -3.5 ± 1.0, -4.6 ± 1.9, -22.1 ± 2.2 and -11.4 ± 1.3 in N, NL, D and DL respectively). ROCK1 activity and expression were higher in D than in N/NL and DL aortas.</p> <p>Conclusion and implications</p> <p>Aortas isolated from STZ-rats present hyper-contracture to AT-II mainly dependent on the up-regulation of ROCK1 expression/activity. In vivo losartan treatment partially corrects AT-II hyper-contracture, limiting the increase in ROCK1 expression/activity. These data offer a new molecular mechanism supporting the rationale for using losartan in the prevention of diabetic vascular complications.</p
Mycotoxins and mycotoxigenic fungi in poultry feed for food-producing animals
Moulds are capable of reducing the nutritional value of feedstuff as well as elaborating several mycotoxins. Mycotoxin-contaminated feed has adverse effects on animal health and productivity. Also, mycotoxins may be carried over into meat and eggs when poultry are fed with contaminated feed. In a point prevalence study feedstuff used for poultry nutrition in Argentina was analyzed for fungal flora, natural incidence of selected mycotoxins, and nutritional quality. Ten mould genera were recovered, six of them known to be mycotoxigenic. More than 28 species were determined. Fumonisins were detected in all the samples (median 1,750 ppb). Forty-four out of 49 samples (90%) were contaminated with DON (median 222 ppb) and OTA (median 5 ppb). Also, 44 out of 49 samples were contaminated with aflatoxins (median 2.685 ppb), 42 samples (86%) with ZEA (median 50 ppb), and 38 samples (78%) with T2-toxin (median 50 ppb). Ninety percent of the samples had at least one type of nutritional deficiency. This study indicates the need for continuous assessment of the mycological status of animal feed production, in order to feed animals for optimal performance ensuring food safety.Facultad de Ciencias Veterinaria
Maternal exposure to an enrichment environment promotes uterine vascular remodeling and prevents embryo loss in mice.
Implantation-related events are crucial for pregnancy success. In particular, defects in vascular remodeling at the maternal-fetal interface are associated with spontaneous miscarriage and recurrent pregnancy loss. Physical activity and therapies oriented to reduce stress improve pregnancy outcomes. In animal models, environmental stimulation and enrichment are associated with enhanced well-being, cognitive function and stress resilience. Here, we studied whether the exposure of BALB/c mice to an enriched environment (EE) regulates crucial events during early gestation at the maternal-fetal interface. Pregnant BALB/c mice were exposed to the EE that combines non-invasive stimuli from the sensory pathway with voluntary physical activity. The pregnancy rate was evaluated. Implantation sites were investigated microscopically and macroscopically. Vascular adaptation parameters at the maternal-fetal interface were analyzed. We found that exposure to the EE prevented pregnancy loss between gestational days 7 and 15. Also, it increased the diameter of the uterine artery and decreased the wall:lumen ratio of the mesometrial decidual vessels, suggesting that EE exposure promotes vascular remodeling. Moreover, it increased nitric oxide synthase activity and inducible nitric oxide synthase expression, as well as prostaglandin F2a production and endoglin expression in the implantation sites. Exposure of pregnant females to the EE regulates uterine physiology, promoting vascular remodeling during early gestation. These adaptations might contribute to preventing embryo loss. Our results highlight the importance of the maternal environment for pregnancy success. The design of an 'EE-like' protocol for humans could be considered as a new non-pharmacologic strategy to prevent implantation failure and recurrent miscarriage.Fil: de la Cruz Borthiry, Fernanda Luz. DEVELOPMENT OF A NOVEL METHOD FOR AMNIOTIC FLUID STEM CELL STORAGE
Background - Current procedures for collection of human
Amniotic Fluid Stem Cells (hAFSCs) imply that amniotic fluid cells were
cultured in flask for two weeks, than can be devoted to research purpose.
However, hAFSCs could be retrieved directly from a small amount of
amniotic fluid that can be obtained at the time of diagnostic
amniocentesis. The aim of the study was to verify if a direct freezing of
amniotic fluid cells is able to maintain and / or improve the potential
of the sub-population of stem cells. Methods - We compared the potential
of the hAFSCs depending on the moment in which they are frozen, cells
obtained directly from amniotic fluid aspiration (D samples) and cells
cultured in flask before freezing (C samples). Colony-forming-unit
ability, proliferation, morphology, stemness-related marker expression,
senescence, apoptosis, and differentiation potential of C and D samples
were compared. Results - hAFSCs isolated from D samples expressed MSC
markers until later passages, had a good proliferation rate, and
exhibited differentiation capacity similar to hAFSCs of C samples.
Interestingly, the direct freezing induce a higher concentration of cells
positive for pluripotency stem cell markers, without teratoma formation
in vivo.
Conclusions - This study suggests that minimal processing may be adequate
for the banking of amniotic fluid cells, avoiding in vitro passages
before the storage and exposure to high oxygen concentration affecting
stem cell properties. This technique might be a reasonable approach in
terms of costs and for the process of accreditation in GMP for a stem
cell bank
