    Porcine reproductive and respiratory syndrome virus (PRRSV) in GB pig herds : farm characteristics associated with heterogeneity in seroprevalence

    Background: The between- and within-herd variability of porcine reproductive and respiratory syndrome virus (PRRSV) antibodies were investigated in a cross-sectional study of 103 British pig herds conducted 2003–2004. Fifty pigs from each farm were tested for anti-PRRSV antibodies using ELISA. A binomial logistic model was used to investigate management risks for farms with and without pigs with PRRSV antibodies and multilevel statistical models were used to investigate variability in pigs' log ELISA IRPC (relative index × 100) in positive herds. Results: Thirty-five herds (34.0%) were seronegative, 41 (39.8%) were seropositive and 27 (26.2%) were vaccinated. Herds were more likely to be seronegative if they had < 250 sows (OR 3.86 (95% CI 1.46, 10.19)) and if the nearest pig herd was ≥ 2 miles away (OR 3.42 (95% CI 1.29, 9.12)). The mean log IRPC in seropositive herds was 3.02 (range, 0.83 – 5.58). Sixteen seropositive herds had only seropositive adult pigs. In these herds, pigs had -0.06 (95% CI -0.10, -0.01) lower log IRPC for every mile increase in distance to the nearest pig unit, and -0.56 (95% CI -1.02, -0.10) lower log IRPC when quarantine facilities were present. For 25 herds with seropositive young stock and adults, lower log IRPC were associated with isolating purchased stock for ≥ 6 days (coefficient - 0.46, 95% CI -0.81, -0.11), requesting ≥ 48 hours 'pig-free time' from humans (coefficient -0.44, 95% CI -0.79, -0.10) and purchasing gilts (coefficient -0.61, 95% CI -0.92, -0.29). Conclusion: These patterns are consistent with PRRSV failing to persist indefinitely on some infected farms, with fadeout more likely in smaller herds with little/no reintroduction of infectious stock. In new research Raymond Foxworth, Laura E. Evans, Gabriel R. Sanchez, Cheryl Ellenwood, and Carmela M. Roybal contextualize the history of colonization and policy neglect by federal and state governments to explain the unequal impact of the pandemic. They find that this disparity is related to a lack of basic infrastructure like safe running water, a shortage of health information available in Native languages, and the high rate of non-tribal members visiting tribal lands during the pandemic. State-level partisanship also plays an important role; Republican dominated states were less likely to implement pandemic mitigation policies such as mask mandates, which in turn has put Native American lives in danger

    A Sr-Rich Star on the Main Sequence of Omega Centauri

    Abundance ratios relative to iron for carbon, nitrogen, strontium and barium are presented for a metal-rich main sequence star ([Fe/H]=--0.74) in the globular cluster omega Centauri. This star, designated 2015448, shows depleted carbon and solar nitrogen, but more interestingly, shows an enhanced abundance ratio of strontium [Sr/Fe] ~ 1.6 dex, while the barium abundance ratio is [Ba/Fe]<0.6 dex. At this metallicity one usually sees strontium and barium abundance ratios that are roughly equal. Possible formation scenarios of this peculiar object are considered.Comment: 13 pages, 3 figures.     Groundwater and Subsurface Water in a Landscape with Shallow Bedrock: Implications for Agricultural Nutrient Export

    Controls on nutrient export from agricultural landscapes include regional hydrological and geological conditions. In many central Kentucky basins shallow fractured bedrock produces focused groundwater discharge and flashy runoff events. Eastern Kentucky University’s Meadowbrook Farm is currently experimenting with nutrient sequestration techniques; however the effectiveness of these techniques given the local hydrogeology needs to be quantified. Our objective was to determine the potential pathways for nutrient export by mapping key hydrologic features and using a watershed approach. The key features mapped were springs, tile drains, runoff channels, land use and the underlying geology. We chose a 0.46 km2 watershed to study closely because it contained a good representation of farm activities and hydrology. Within the watershed we constructed a V-notch weir at the outlet to determine the flow from surface runoff and groundwater outputs and conducted a water budget. Next, we installed gaging stations along a main fourth order stream that the farm drains into. Finally, we selected sites within the stream to conduct differential stream gaging to quantify the input and outputs of groundwater to the stream. Mapping indicated that the Boyle Dolomite contained 100% of the springs. This means that the Boyle Dolomite is controlling groundwater export off of the farm. Hydrographs indicated flashy runoff events consisting of ~15% of precipitation volume. Tributaries dried approximately 5 days after a precipitation even. Nutrient export must be confined within these flashy events. Differential stream gaging indicated both gaining and losing stream reaches

    Bite force and cranial bone strain in four species of lizards

    In vivo bone strain data provide direct evidence of strain patterns in the cranium during biting. Compared to mammals, in vivo bone strains in lizard skulls are poorly documented. This paper presents strain data from the skulls of Anolis equestris, Gekko gecko, Iguana iguana and Salvator merianae during transducer biting. Analysis of variance was used to investigate effects of bite force, bite point, diet, cranial morphology and cranial kinesis on strain magnitudes. Within individuals the most consistent determinants of variance in bone strain magnitudes are gage location and bite point, with the importance of bite force varying between individuals. Inter-site variance in strain magnitudes—strain gradient—is present in all individuals, and varies with bite point. Between individuals within species, variance in strain magnitude is driven primarily by variation in bite force, not gage location or bite point, suggesting that inter-individual variation in patterns of strain magnitude is minimal. Between species, variation in strain magnitudes is significantly impacted by bite force and species membership, as well as by interactions between gage location, species, and bite point. Independent of bite force, species differences in cranial strain magnitudes may reflect selection for different cranial morphology in relation to feeding function, but what these performance criteria are is not clear. The relatively low strain magnitudes in Iguana and Uromastyx compared to other lizards may be related to their herbivorous diet. Cranial kinesis and the presence or absence of postorbital and supratemporal bars are not important determinants of inter-specific variation in strain magnitudes

    The Effect of Cumulus Cloud Field Anisotropy on Domain-Averaged Solar Fluxes and Atmospheric Heating Rates

    Cumulus clouds can become tilted or elongated in the presence of wind shear. Nevertheless, most studies of the interaction of cumulus clouds and radiation have assumed these clouds to be isotropic. This paper describes an investigation of the effect of fair-weather cumulus cloud field anisotropy on domain-averaged solar fluxes and atmospheric heating rate profiles. A stochastic field generation algorithm was used to produce twenty three-dimensional liquid water content fields based on the statistical properties of cloud scenes from a large eddy simulation. Progressively greater degrees of x-z plane tilting and horizontal stretching were imposed on each of these scenes, so that an ensemble of scenes was produced for each level of distortion. The resulting scenes were used as input to a three-dimensional Monte Carlo radiative transfer model. Domain-average transmission, reflection, and absorption of broadband solar radiation were computed for each scene along with the average heating rate profile. Both tilt and horizontal stretching were found to significantly affect calculated fluxes, with the amount and sign of flux differences depending strongly on sun position relative to cloud distortion geometry. The mechanisms by which anisotropy interacts with solar fluxes were investigated by comparisons to independent pixel approximation and tilted independent pixel approximation computations for the same scenes. Cumulus anisotropy was found to most strongly impact solar radiative transfer by changing the effective cloud fraction, i.e., the cloud fraction when the field is projected on a surface perpendicular to the direction of the incident solar beam

    The Acinetobacter baumannii two-component system aders regulates genes required for multidrug efflux, biofilm formation, and virulence in a strain-specific manner

    The opportunistic pathogen Acinetobacter baumannii is able to persist in the environment and is often multidrug resistant (MDR), causing difficulties in the treatment of infections. Here, we show that the two-component system AdeRS, which regulates the production of the AdeABC multidrug resistance efflux pump, is required for the formation of a protective biofilm in an ex vivo porcine mucosal model, which mimics a natural infection of the human epithelium. Interestingly, deletion of adeB impacted only on the ability of strain AYE to form a biofilm on plastic and only on the virulence of strain Singapore 1 for Galleria mellonella. RNA-Seq revealed that loss of AdeRS or AdeB significantly altered the transcriptional landscape, resulting in the changed expression of many genes, notably those associated with antimicrobial resistance and virulence interactions. For example, A. baumannii lacking AdeRS displayed decreased expression of adeABC, pil genes, com genes, and a pgaC-like gene, whereas loss of AdeB resulted in increased expression of pil and com genes and decreased expression of ferric acinetobactin transport system genes. These data define the scope of AdeRS-mediated regulation, show that changes in the production of AdeABC mediate important phenotypes controlled by AdeRS, and suggest that AdeABC is a viable target for antimicrobial drug and antibiofilm discovery. IMPORTANCE Acinetobacter baumannii is a nosocomial pathogen and is an increasing problem in hospitals worldwide. This organism is often multidrug resistant, can persist in the environment, and forms a biofilm on environmental surfaces and wounds. Overproduction of efflux pumps can allow specific toxic compounds to be pumped out of the cell and can lead to multidrug resistance. This study demonstrates the role of the A. baumannii efflux pump AdeB, and its regulator AdeRS, in multidrug resistance, epithelial cell killing, and biofilm formation. Deletion of the genes encoding these systems led to increased susceptibility to antibiotics, decreased biofilm formation on biotic and abiotic surfaces, and decreased virulence. Our data suggest that inhibition of AdeB could prevent biofilm formation or colonization in patients by A. baumannii and provides a good target for drug discovery

    Abundances on the Main Sequence of Omega Centauri

    Abundance ratios of carbon, nitrogen and strontium relative to iron, calculated using spectrum synthesis techniques, are given for a sample of main sequence and turnoff stars that belong to the globular cluster omega Centauri. The variations of carbon, nitrogen and/or strontium show several different abundance patterns as a function of [Fe/H]. The source of the enhancements/depletions in carbon, nitrogen and/or strontium may be enrichment from asymptotic giant branch stars of low (1--3 solar masses) and intermediate (3--8 solar masses) mass. Massive rotating stars which produce excess nitrogen without carbon and oxygen overabundances may also play a role.     ADAPT study: adaptation of evidence-informed complex population health interventions for implementation and/or re-evaluation in new contexts: protocol for a Delphi consensus exercise to develop guidance

    Introduction Complex population health interventions that are effective in one context may not be effective elsewhere, and may even be harmful. As such, an intervention may require adaptation to ensure it fits with a new context. To date, there is no overarching guidance to help researchers to adapt and evaluate interventions in new contexts, and no criteria to support research funders or journals assess proposed or reported adaptations or evaluation. There is limited assistance for policy-makers and practitioners to decide if evidence-informed interventions are appropriate to their context, or if adaptation and further evaluation is needed. This Delphi exercise will contribute to the development of guidance for these communities to support the adaptation, implementation and/or re-evaluation of complex population health interventions in new contexts. Methods We will conduct a Delphi consensus exercise to gather expert opinion from researchers, research funders, journal editors and policy-makers. Expert opinion will be sought on: appropriate definitions and concepts, identifying key methodological considerations and establishing adaptations and processes to be undertaken during adaptation of complex population health interventions in new contexts. Ethics and dissemination Ethics approval for the Delphi exercise has been obtained from the University of Glasgow and and the RAND institutional research board. Dissemination of the results of this study will be through peer-reviewed publications, workshops at national and international conferences, and a summary of the guidance developed for key organisations and stakeholders
