    Characterization of phychoactive plant pollen: a new tool in forensic investigations

    Palynology, based on the micromorphological characterization of pollen and spores, can be a valid ally for the resolution of forensic cases. Since its first application in 1959 for a murder in Austria, this discipline has been applied to cases of illegal smuggling involving drugs, weapons, medicines, and money (1). Also in Italy, several examples of palynology applied to kidnapping and homicide crime solving have been reported. In particular, a close connection was found between forensic palynology and aerobiology, as a reference tool for establishing the season when unknown death occurred (2-4).The use of palynology is also useful to trace possible pollen contamination of drugs: e.g. in a seizure of a batch of drugs in New York in 1991 (5). It is also possible to solve cases of murder, rape, theft, and food fraud (6). Pollen can adhere firmly to any surface and resist decomposition, while its morphology can be a distinctive feature for the identification of plant species and habitats (1,4,6). The present study concerns the pollen of 12 species of psychoactive plants belonging to 9 families, including illegal drug plants (e.g. Erythroxylum coca; Lophophora williamsii), and plants used as legal alternatives to drugs (e.g. Leonotis leonurus; Brugmansia suaveolens), some of which are currently marketed on internet. Given the lack of literature in this study, we started to compile a palynological atlas of psychoactive plants, aimed at providing support in the forensic field. These data can been useful to trace the origin of illegal drugs, and to identify the most likely route of drug shipping (2). The selected pollen grains was sampled from herbarium specimens, the Reference Pollen Collection (Palinoteca), or prepared as fresh samples collected from the Botanical Garden of Genoa University and the Hanbury Botanical Garden of Ventimiglia. Unacetolyzed pollen was prepared in glycerin jelly containing basic fuchsin dye. The analysis was carried out on 40 pollen grains for each species at the magnification of 40x.The diagnostic features of pollen grains studied were: length of polar axis (P) and equatorial axis (E), pollen shape (P/E ratio), number and type of apertures, exine thickness and sculpture. For the two species belonging to Convolvulaceae (Ipomoea violacea and Argyreia nervosa), also spine length, interspinal distance, pore diameter, exine and nexine thickness, have been considered. Observations were carried out by using light and scanning electron microscopy. In some cases, the analyses showed slight morphological differences compared to literature data, probably due to a different number of measurements, while in other cases new morphological features were recorded. A web repository is being created to make morphometric data and pictures of psychoactive plant pollen available, thus providing a useful tool for forensic investigations

    Emerging Exotic Fruits: New Functional Foods in the European Market

    The consumption of exotic fruits is rapidly increasing in European countries. Some of these products have attracted much interest due to their alleged properties of preventing malnutrition, over-nutrition, and disease, maintaining a healthy body. Scientific studies on these fruits are multiplying, including chemical characterizations and biological investigations on in vitro and in vivo experimental models. This review concerns four edible fruits: Hylocereus undatus (dragon fruit), Annona cherimola (cherimoya), Citrus australasica (finger lime), and Averrhoa carambola (carambola or star fruit). By screening biomedical databases, viz. Scopus, WOS, and PubMed, a total of 131 papers have been selected. Data reveals a wide series of biological effects that confirm traditional medicinal uses or suggest new therapeutic applications. Most studies concern problems related to nutrition, such as body redox balance, metabolic syndrome, and hepatoprotective effects, but other properties have been highlighted, including anticancer, antimicrobial, anti-inflammatory, and neuroprotective effects, as well as cardiovascular and skin protection. Pharmacological investigations have also been focused on specific compounds, assuming a potential role in drug discovery. In summary, food products, byproducts, and single compounds derived from these plants could be exploited in the prevention of disease or for specific treatments of health problems

    Plants Used In Artisanal Fisheries On The Western Mediterranean Coasts Of Italy

    Background Artisanal fisheries in the Mediterranean, especially in Italy, have been poorly investigated. There is a long history of fishing in this region, and it remains an important economic activity in many localities. Our research entails both a comprehensive review of the relevant literature and 58 field interviews with practitioners on plants used in fishing activities along the Western Mediterranean Italian coastal regions. The aims were to record traditional knowledge on plants used in fishery in these regions and to define selection criteria for plant species used in artisanal fisheries, considering ecology and intrinsic properties of plants, and to discuss the pattern of diffusion of shared uses in these areas. Methods Information was gathered both from a general review of ethnobotanical literature and from original data. A total of 58 semi-structured interviews were carried out in Liguria, Latium, Campania and Sicily (Italy). Information on plant uses related to fisheries were collected and analyzed through a chi-square residual analysis and the correspondence analysis in relation to habitat, life form and chorology. Results A total of 60 plants were discussed as being utilized in the fisheries of the Western Italian Mediterranean coastal regions, with 141 different uses mentioned. Of these 141 different uses, 32 are shared among different localities. A multivariate statistical analysis was performed on the entire dataset, resulting in details about specific selection criteria for the different usage categories (plants have different uses that can be classified into 11 main categories). In some uses, species are selected for their features (e.g., woody), or habitat (e.g., riverine), etc. The majority of uses were found to be obsolete (42%) and interviews show that traditional fishery knowledge is in decline. There are several reasons for this, such as climatic change, costs, reduction of fish stocks, etc. Conclusions Our research correlates functional characteristics of the plants used in artisanal fishery and habitats, and discusses the distribution of these uses. This research is the first comprehensive outline of plant role in artisanal fisheries and traditional fishery knowledge in the Mediterranean, specifically in Italy

    The bioactivity of Hedysarum coronarium extracts on skin enzymes and cells correlates with phenolic content

    CONTEXT: The search for bioactive compounds from botanical sources is attracting much interest. However, differences in chemical composition may occur within the same species depending on different geographical origins. OBJECTIVES: We evaluated the properties on skin enzymes and cells of extracts from sulla legume crop Hedysarum coronarium L. (Fabaceae), collected at two Italian sites near Pisa and Ventimiglia, for possible dermatological and cosmetic applications. MATERIAL AND METHODS: Plant aerial portions were extracted in MTBE/ethyl acetate/acetone, obtaining two extracts named Pisa sulla extract (PSE) and Ventimiglia sulla extract (VSE). Extracts were subjected to chemical characterization, LC-MS/MS analysis and biological assays. RESULTS: PSE showed stronger antiradical scavenging and higher phenolic and flavonoid contents with respect to VSE. LC-MS/MS analysis revealed similar composition for the two extracts, but PSE was richer in condensed tannins and flavonoids, principally rhoifolin, quercetin, naringenin and derivatives. PSE induced stronger inhibition on collagenase and elastase by in vitro enzyme assays, possibly due to higher levels of condensed tannins and quercetin. ELISA bioassay on human dermal fibroblasts revealed stronger PSE induction of collagen production. Determination of glycerol release from adipocytes disclosed stronger stimulation of lipolysis by PSE, allegedly ascribed to higher charge of quercetin and derivatives. In summary, the higher richness in phenolics of PSE is strictly related to stronger bioactivity. DISCUSSION AND CONCLUSIONS: Data indicate that aerial H. coronarium material is suitable for the development of dermatological and cosmeceutical products, but the geographical origin is an important factor for maximally exploiting the biological properties of this species

    Characterization and Phytotoxicity Assessment of Essential Oils from Plant Byproducts

    The present work describes the chemical characterization and the phytotoxicity assessment of essential oils (EOs) obtained from spent materials or pruning waste of four plant species: Zingiber officinale Roscoe used in the juicing industry, Pistacia vera L. var. Bronte used in the food industry, discarded material of industrial hemp (Cannabis sativa L. var. Futura 75), and pruning waste from Cupressus sempervirens L. The phytochemical profile of the EOs was evaluated by gas chromatographic flame ionization detection (GC-FID) and GC-MS analyses, which highlighted the presence of several compounds with a wide range of biological activities. Among them, application possibilities in agriculture were evaluated by studying the phytotoxic activity in vitro against germination and initial radical growth of several seeds such as Raphanus sativus L., Lepidium sativum L., Lactuca sativa L., Solanum lycopersicum L., Lolium multiflorum Lam., and Portulaca oleracea L

    Coriandrum sativum and Lavandula angustifolia essential oils: Chemical composition and activity on central nervous system

    The aims of this study are to determine the chemical composition of Lavandula angustifolia Mill. and Coriandrum sativum L. essential oils, to evaluate their cytotoxic effects in SH-SY5Y human neuroblastoma cells, to investigate whether an alteration of adenylate cyclase 1 (ADCY1) and of extracellular signal-regulated kinase (ERK) expression can take part in the molecular mechanisms of the essential oils, and to study their possible neuronal electrophysiological effects. The essential oils were obtained by hydrodistillation, and studied by GC and GC-MS. In the oils from L. angustifolia and C. sativum, linalool was the main component (33.1% and 67.8%, respectively). SH-SY5Y cells were incubated with different concentrations of essential oils and of linalool. Cell viability and effects on ADCY1 and ERK expression were analyzed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide MTT andWestern blotting, respectively. Variation in cellular electrophysiology was studied in primary cultures of rat cortical neurons with a multi-electrode array (MEA)-based approach. The essential oils and linalool revealed different cytotoxic activities. Linalool inhibited ADCY1 and ERK expression. Neuronal networks subjected to L. angustifolia and C. sativum essential oils showed a concentration-dependent inhibition of spontaneous electrical activity

    Anti-bacterial prenylated phenols from the Kurdish medicinal plant Onobrychis carduchorum (Fabaceae)

    Onobrychis carduchorum C.C. Towns. is a plant widely employed in the Kurdish traditional medicine, to cure inflammations and other skin diseases. We isolated ten different phenolic metabolites from an acetone extract of leaves and flowers. The phenolic compounds belong to three different classes, i.e.: 1. iso-flavones, having a genistein skeleton; 2. flavanones, bearing a naringenin skeleton; 3. dihydro-stilbenes. Many of them have a prenyl unit on an aromatic ring. The above compounds have been found to date mostly in other Fabaceae, as Glycyrrhiza glabra L. (liquorice)1. However, their bioactivities are largely unknown. In this work we reported a strong inhibition activity on the growth of Staphylococcus aureus, a well-known human pathogen. In particular, compound (Fig. 1) shows an inhibitory activity on growth, comparable to that of vancomycin, using the agar-diffusion standard method

    Feijoa Fruit Peel: Micro-morphological Features, Evaluation of Phytochemical Profile, and Biological Properties of Its Essential Oil

    Acca sellowiana (O. Berg) Burret (Feijoa) is an evergreen shrub, belonging to the Mirtaceae family. The aim of this study was to investigate the micromorphological features of the feijoa fruit peel and to evaluate the phytochemical profile, as well as the antioxidant, cytoprotective, and antimicrobial properties of its essential oil (EO), by several in vitro cell-free and cell-based assays. The micromorphological analysis showed several schizogenic secretory cavities, immediately below the epidermal layer. Forty compounds were identified and quantified by GC-FID and GC-MS analyses. Sesquiterpenes were the most abundant ones (76.89%), followed by monoterpene hydrocarbons (3.26%), and oxygenated monoterpenes (0.34%). The main compounds were \u3b3-Selinene (17.39%), \u3b1-Cariophyllene (16.74%), \u3b2-Cariophyllene (10.37%), and Germacene D (5.32%). The EO showed a strong and dose-dependent antioxidant, and free-radical scavenging activity. Furthermore, it showed cytoprotective activity on the lymphocytes, that have been pre-treated with 100 \u3bcM tert-butyl-hydroperoxide (t-BOOH), as well as a decrease in intracellular reactive oxygen species (ROS), induced by t-BOOH on erythrocytes. A preliminary antimicrobial screening against GRAM+ and GRAM- bacteria, as well as on fungi highlighted that EO showed the best activity against S. aureus and C. albicans (MIC 2.7 mg/mL). In light of these results, feijoa fruit EO could find various applications, especially in the food, nutraceutical, and pharmaceutical fields

    Eucalyptus cinerea and E. nicholii by-Products as Source of Bioactive Compounds for Agricultural Applications

    The cultivation of different species of Eucalyptus has recently expanded in Liguria (Italy) due to the growing demand of the North European floricultural market. Eucalyptus tree branches are cut and selected for their quality, resulting in large amounts of waste biomass to be disposed of. The aim of our study was to evaluate the phytotoxic and antimicrobial activities of essential oils (EOs) from pruning wastes of E. cinerea (EC) and E. nicholii (EN), for potential applications in agriculture. Phytochemical analyses showed eucalyptol (1,8-cineole) as the major component in both EOs, but the EO yield of EN was higher than that of EC, in agreement with a significantly higher oil gland density on EN leaves. EOs from both species showed phytotoxicity on both weeds tested, but no significant inhibition on horticultural crop seed germination, except for Raphanus sativus. The EO from EC showed the strongest antibacterial activity, while the EO from EN showed the strongest antifungal activity. Concluding, EOs from Eucalyptus pruning may be used as possible alternatives to synthetic herbicides and pesticides, acting as antimicrobial and antifungal agents, thus representing a safe strategy for crop management programs
