Identification of the PTPN22 functional variant R620W as susceptibility genetic factor for giant cell arteritis
Objective: To analyse the role of the PTPN22 and CSK genes, previously associated with autoimmunity, in the predisposition and clinical phenotypes of giant cell arteritis (GCA). Methods: Our study population was composed of 911 patients diagnosed with biopsy-proven GCA and 8136 unaffected controls from a Spanish discovery cohort and three additional independent replication cohorts from Germany, Norway and the UK. Two functional PTPN22 polymorphisms (rs2476601/R620W and rs33996649/R263Q) and two variants of the CSK gene (rs1378942 and rs34933034) were genotyped using predesigned TaqMan assays. Results: The analysis of the discovery cohort provided evidence of association of PTPN22 rs2476601/R620W with GCA (PFDR=1.06E-04, OR=1.62, CI 95% 1.29 to 2.04). The association did not appear to follow a specific GCA subphenotype. No statistically significant differences between allele frequencies for the other PTPN22 and CSK genetic variants were evident either in the case/control or in stratified case analysis. To confirm the detected PTPN22 association, three replication cohorts were genotyped, and a consistent association between the PTPN22 rs2476601/R620W variant and GCA was evident in the overall meta-analysis (PMH=2.00E-06, OR=1.51, CI 95% 1.28 to 1.79). Conclusions: Our results suggest that the PTPN22 polymorphism rs2476601/R620W plays an important role in the genetic risk to GCA
Influence of the IL17A locus in giant cell arteritis susceptibility
Objective: Different lines of evidence have highlighted the role of IL-17A in the inflammatory process occurring in giant cell arteritis (GCA). The aim of the present study was to assess whether the IL17A locus influences GCA susceptibility and its clinical subphenotypes. Methods: We carried out a large meta-analysis including a total of 1266 biopsy-proven GCA patients and 3779 healthy controls from four European populations (Spain, Italy, Germany and Norway). Five IL17A polymorphisms (rs4711998, rs8193036, rs3819024, rs2275913 and rs7747909) were selected by tagging and genotyped using TaqMan assays. Allelic combination and dependency tests were also performed. Results: In the pooled analysis, two of the five analysed polymorphisms showed evidence of association with GCA (rs2275913: PMH=1.85E−03, OR=1.17 (1.06-1.29); rs7747909: PMH=8.49E-03, OR=1.15 (1.04-1.27)). A clear trend of association was also found for the rs4711998 variant (PMH=0.059, OR=1.11 (1.00-1.23)). An independent effect of rs2275913 and rs4711998 was evident by conditional regression analysis. In addition, the haplotype harbouring the risk alleles better explained the observed association than the polymorphisms independently (likelihood p value <10−05). Conclusions: Polymorphisms within the IL17A locus show a novel association with GCA. This finding supports the relevant role of the Th17 cells in this vasculitis pathophysiology
Quantitative Untersuchungen zur NO Aufnahme durch Pflanzen aus der Luft-Konstruktion eines Expositionssystems und Ergebnisse an Sonnenblumen (Helianthus Annuus L)
Anthropogene Luftverunreinigungen aus Industrieanlagen, Verkehr und Haushalt, und zwar speziell NO-Emissionen sind an dem Ursachenkomplex der "neuartigen" Waldschäden beteiligt. Trotz eingeleiteter Maßnahmen zur Emissionsminderung ist der Gesamtausstoß seit 1982 weiter angestiegen und liegt in der Bundesrepublik bei 3 Mio. t NO pro Jahr. In Zusammenhang mit der Ursachenforschung der "neuartigen" Waldschäden wird nach dem möglichen Effekt dieser seit den letzten zwei bis drei Jahrzehnten stark angestiegenen NO-Emissionen gefragt. Sind sie für die Pflanzen eine zusätzliche Stickstoffquelle, werden sie direkt aufgenommen, wird der Stickstoff in den Blattorganen verwertet, führt dies zu internen Nährstoffungleichgewichten? Es war daher das Ziel der Arbeit, die Stickstoffaufnahme aus dem NO der Luft in die oberirdischen Pflanzenteile unter den in der Atmosphäre realistisch vorkommenden NO-Konzentrationen (3 bis 40 ppb NO) experimentell zu ermitteln. Voraussetzungen für solche Untersuchungen sind: 1. Eine Expositionsanlage zu entwickeln, in der Pflanzen unter geeigneten Klimabedingungen einer Atmosphäre mit diesem Stickstoffdioxidgehalt ausgesetzt werden können, 2. eine Analysentechnik einzusetzen, mit der eine quantitative Stickstoffbestimmung in Pflanzen möglich ist und 3. ein Meßverfahren aufzubauen, mit dem N als Tracer aus dem NO der Luft in Pflanzen eindeutig nachgewiesen werden kann. Für die Experimente wurde deshalb eine zylindrische Expositionskammer mit 1641 Volumen angefertigt. Diese wurde in einer Klimakammer installiert, so daß reproduzierbare Temperatur- und Beleuchtungsbedingungen gewährleistet waren. Der Wurzelraum war durch eine Teflonplatte vom oberirdischen Expositionsraum abgetrennt, um einerseits einen NO-Eintrag in die Nährlösung zu verhindern und andererseits N-Umsetzungen der Mikroorganismen im Wurzelraum nicht im Luftraum der Sproßatmosphäre zu erfassen. Um bei diesen geringen NO-Konzentrationen Reaktionen von NO mit den Materialien auszuschließen, wurden NO-inerte Werkstoffe, Glas für alle lichtdurchlässigen Teile und Teflon für Dichtungs- und Leitungsmaterial, verwendet. Der NO-Stickstoff in der Pflanze wurde mit Hilfe der Stabilisotopenmeßtechnik auf der Grundlage von N als Tracer untersucht. Die zugeführte Luft wurde daher von natürlichem NO gereinigt und NO kontrolliert zudosiert. Genome-Wide Transcriptional Profiling of the Purple Sulfur Bacterium Allochromatium vinosum DSM 180T during Growth on Different Reduced Sulfur Compounds
The purple sulfur bacterium Allochromatium vinosum DSM 180(T) is one of the best-studied sulfur-oxidizing anoxygenic phototrophic bacteria, and it has been developed into a model organism for laboratory-based studies of oxidative sulfur metabolism. Here, we took advantage of the organism's high metabolic versatility and performed whole-genome transcriptional profiling to investigate the response of A. vinosum cells upon exposure to sulfide, thiosulfate, elemental sulfur, or sulfite compared to photoorganoheterotrophic growth on malate. Differential expression of 1,178 genes was observed, corresponding to 30% of the A. vinosum genome. Relative transcription of 551 genes increased significantly during growth on one of the different sulfur sources, while the relative transcript abundance of 627 genes decreased. A significant number of genes that revealed strongly enhanced relative transcription levels have documented sulfur metabolism-related functions. Among these are the dsr genes, including dsrAB for dissimilatory sulfite reductase, and the sgp genes for the proteins of the sulfur globule envelope, thus confirming former results. In addition, we identified new genes encoding proteins with appropriate subcellular localization and properties to participate in oxidative dissimilatory sulfur metabolism. Those four genes for hypothetical proteins that exhibited the strongest increases of mRNA levels on sulfide and elemental sulfur, respectively, were chosen for inactivation and phenotypic analyses of the respective mutant strains. This approach verified the importance of the encoded proteins for sulfur globule formation during the oxidation of sulfide and thiosulfate and thereby also documented the suitability of comparative transcriptomics for the identification of new sulfur-related genes in anoxygenic phototrophic sulfur bacteria
