    Formative Assessment and Professional Training: Reflections from a Mathematics course in Bioengineering

    Bioengineering is currently considered an interdisciplinary professional field which provides solutions to different problems arising in the area of health care. Its strategic importance is widely acknowledged since its developments and proposals could help diminish the level of technological dependence in the sector. The fast pace of innovation in the area of biomedical technology gives rise to permanent reflection on the learning goals and teaching strategies proposed by educators in the different training stages of a bioengineer. In this context, learning assessment appears as a controversial issue which needs to be debated and rethought. This paper describes the reflections of teachers of a Mathematics course within a Bioengineering program around the question, What approach to assessment favors the student's participation, autonomy and training as a future bioengineer? The investigation was carried out in the framework of a Participatory Research Action project and helped us to redesign assessment activities from a different perspective.Fil: Carrere, C..     Characterization of the molecular changes associated with the overexpression of a novel epithelial cadherin splice variant mRNA in a breast cancer model using proteomics and bioinformatics approaches: identification of changes in cell metabolism and an increased expression of lactate dehydrogenase B

    Breast cancer (BC) is the most common female cancer and the leading cause of cancer death in women worldwide. Alterations in epithelial cadherin (E-cadherin) expression and functions are associated to BC, but the underlying molecular mechanisms have not been fully elucidated. We have previously reported a novel human E-cadherin splice variant (E-cadherin variant) mRNA. Stable transfectants in MCF-7 human BC cells (MCF7Ecadvar) depicted fibroblast-like cell morphology, E-cadherin wild-type downregulation, and other molecular changes characteristic of the epithelial-to-mesenchymal transition process, reduced cell-cell adhesion, and increased cell migration and invasion. In this study, a two-dimensional differential gel electrophoresis (2D-DIGE) combined with mass spectrometry (MS) protein identification and bioinformatics analyses were Results: By 2D-DIGE and MS analysis, 50 proteins were found differentially expressed (≥ Δ1.5) in MCF7Ecadvar compared to control cells. Validation of transcript expression was done in the ten most overexpressed and underexpressed proteins. Bioinformatics analyses revealed that 39 of the 50 proteins identified had been previously associated to BC. Moreover, metabolic processes were the most affected, and glycolysis the canonical pathway most altered. The lactate dehydrogenase B (LDHB) was the highest overexpressed protein, and transcript levels were higher in MCF7Ecadvar than in control cells. In agreement with these findings, MCF7Ecadvar conditioned media had lower glucose and higher lactate levels than control cells. MCF7Ecadvar cell treatment with 5 mM of the glycolytic inhibitor 2-deoxy-glucose led to decreased cell viability, and modulation of LDHB expression in MCF7Ecadvar cells with a specific small interfering RNA resulted in decreased cell proliferation. Finally, a positive association between expression levels of the E-cadherin variant and LDHB transcripts was demonstrated in 21 human breast tumor tissues, and breast tumor samples with higher Ki67 expression showed higher LDHB mRNA levels. Conclusions: Results from this investigation contributed to further characterize molecular changes associated to the novel E-cadherin splice variant expression in BC cells. They also revealed an association between expression of the novel variant and changes related to BC progression and aggressiveness, in particular those associated to cell metabolism.     Understanding formative assessment in a mathematics course for Bioengineering students

    [EN] This paper describes how formative assessment was introduced into a bioengineering mathematics course. This change was carried out within a Participatory Action Research project and involved a shift from an assessment system based on midterm tests, final exams and laboratory assignments to a new system which emphasizes debate, feedback and student participation. In this new assessment approach the students complete written reports and present them orally in class, giving rise to a rich exchange in which peer assessment, self-assessment and teacher assessment are combined. They are also asked to submit drafts for their laboratory assignments so as to receive teacher feedback; and in a further instance of self-assessment, they are asked to complete checklists and questionnaires on their performance in the course. The first results of this new system are encouraging as student performance has considerably improved and, furthermore, a healthy change of attitude has been observed in both students and teachers.[ES] Este trabajo describe cómo la evaluación formativa fue introducida en un curso de matemáticas para futuros bioingenieros. Este cambio se desarrolló en el contexto de una indagación enmarcada en los principios metodológicos de una Investigación Acción Participativa. Como resultado de la indagación el sistema tradicional de evaluación basado en exámenes parciales, evaluaciones finales y trabajos de laboratorio fue reemplazado por un nuevo sistema que incorpora nuevas actividades enfatizando el debate, la retroalimentación y la participación de los estudiantes. En este nuevo sistema se solicita a los estudiantes completar informes escritos y presentarlos oralmente en la clase, dando lugar a un intercambio enriquecido por la combinación de la evaluación entre pares, la autoevaluación y la evaluación del docente. Por otro lado también se les propone realizar entregas de borradores de los trabajos de laboratorio para recibir retroalimentación del docente, y en otra instancia de autoevaluación, se les presentan listas de cotejo y cuestionarios para reflexionar sobre su desempeño en el curso.     Bases moleculares da interação celular em modelos de reprodução e câncer: identificação de proteínas e mecanismos envolvidos

    La interacción entre las células somáticas y entre las gametas involucra una serie de eventos moleculares que no han sido dilucidados totalmente. Nuestro grupo de investigación ha desarrollado proyectos dirigidos a profundizar el conocimiento de dichos eventos. Los estudios han comprendido el análisis de moduladores de la funcionalidad espermática (ej. efecto de la temperatura de incubación, las concentraciones del ión calcio, los anticuerpos antiespermáticos de fluidos biológicos en la motilidad, la capacitación y la exocitosis acrosomal). Asimismo, hemos caracterizado componentes del espermatozoide (ej. CaM Kinasa IV, proacrosina/acrosina) y de secreciones del tracto femenino (ej. Grp78/BiP), evaluado su rol en el desarrollo de capacidad fecundante y, en algunos casos, investigado su relación con la infertilidad. En años recientes, nuestros proyectos se han extendido al estudio de las cadherinas en eventos de adhesión celular durante la fecundación; hemos caracterizado la expresión de cadherina epitelial y neural en tejidos reproductivos y gametas y evaluado su participación en la fecundación. Dada su reconocida relevancia en el cáncer, hemos abordado estudios en diversos modelos tumorales. Nuestras investigaciones han contribuido a la comprensión de los eventos de interacción de las gametas durante la fecundación así como entre las células somáticas durante la progresión tumoral.Cell-cell interaction between somatic cells as well as gametes involves molecular events that have not been completely elucidated. Our research group has developed projects aimed at studying proteins and mechanisms participating in these interactions. Several modulators of sperm functions have been analyzed (i.e. incubation temperature, calcium ion concentration, and antisperm antibodies present in biological fluids upon sperm motility, capacitation and acrosomal exocytosis). In addition, proteins from spermatozoa (i.e. CaM Kinase IV, proacrosin/acrosin) and from secretions of the female tract (Grp78/BiP) have been characterized, and their role in the development of sperm fertilizing ability assessed. In some cases, their relationship with infertility was evaluated. In recent years, our projects have been extended to study members of the cadherin superfamily and related proteins; in particular, the expression of epithelial and neural cadherin in reproductive tissues and gametes was characterized and evidence of their participation in fertilization-related cell-cell adhesion events shown. Based on the vast evidence of the role of these proteins in tumor progression, our current research also involves studies of cancer models. Our projects have contributed to the understanding of the molecular basis of cell-cell interaction during fertilization as well as during tumor progression.A interação entre as células somáticas e entre os gametas envolve uma série de eventos moleculares que não têm sido elucidados totalmente. Nosso grupo de pesquisa tem desenvolvido projetos encaminhados a aprofundar o conhecimento de tais eventos. Os estudos têm compreendido a análise de moduladores da funcionalidade espermática (ex. efeito da temperatura de incubação, as concentrações do íon cálcio, os anticorpos antiespermáticos de fluidos biológicos na motilidade, a capacitação e a exocitose acrossomal). Do mesmo modo, caracterizamos componentes do espermatozoide (ex. CaM Kinase IV, proacrosina /acrosina) e de secreções do trato feminino (ex. Grp78/BiP), avaliamos seu papel no desenvolvimento de capacidade fecundante e, em alguns casos, investigamos sua relação com a infertilidade. Em anos recentes, nossos projetos se têm estendido ao estudo das caderinas em eventos de adesão celular durante a fecundação; temos caracterizado a expressão de caderina epitelial e neural em tecidos reprodutivos e gametas e avaliamos sua participação na fecundação.
