    Characterization of femtosecond laser written waveguides for integrated biochemical sensing

    Fluorescence detection is known to be one of the most sensitive among the different optical sensing techniques. This work focuses on excitation and detection of fluorescence emitted by DNA strands labeled with fluorescent dye molecules that can be excited at a specific wavelength. Excitation occurs via optical channel waveguides written with femtosecond laser pulses applied coplanar with a microfluidic channel on a glass chip. The waveguides are optically characterized in order to facilitate the design of sensing structures which can be applied for monitoring the spatial separation of biochemical\ud species as a result of capillary electrophoresis

    Satellite cells are skeletal muscle stem cells capable of self-renewal and differentiation after transplantation, but whether they contribute to endogenous muscle fiber repair has been unclear. The transcription factor Pax7 marks satellite cells and is critical for establishing the adult satellite cell pool. By using a lineage tracing approach, we show that after injury, quiescent adult Pax7(+) cells enter the cell cycle; a subpopulation returns to quiescence to replenish the satellite cell compartment, while others contribute to muscle fiber formation. We demonstrate that Sprouty1 (Spry1), a receptor tyrosine kinase signaling inhibitor, is expressed in quiescent Pax7(+) satellite cells in uninjured muscle, downregulated in proliferating myogenic cells after injury, and reinduced as Pax7(+) cells re-enter quiescence. We show that Spry1 is required for the return to quiescence and homeostasis of the satellite cell pool during repair. Our results therefore define a role for Spry1 in adult muscle stem cell biology and tissue repair

    We report on a new optical implementation of primary gas thermometry based on laser absorption spectrometry in the near infrared. The method consists in retrieving the Doppler broadening from highly accurate observations of the line shape of the R(12) ν1+2ν210+ν3\nu_{1} + 2 \nu_{2}^{\phantom{1}0} + \nu_{3} transition in CO2_{2} gas at thermodynamic equilibrium. Doppler width measurements as a function of gas temperature, ranging between the triple point of water and the gallium melting point, allowed for a spectroscopic determination of the Boltzmann constant with a relative accuracy of 1.6×104\sim1.6\times10^{-4}.Comment: Submitted to Physical Review Letter

    Coherent tunneling by adiabatic passage in an optical waveguide system

    We report on the first experimental demonstration of light transfer in an engineered triple-well optical waveguide structure which provides a classic analogue of Coherent Tunnelling by Adiabatic Passage (CTAP) recently proposed for coherent transport in space of neutral atoms or electrons among tunneling-coupled optical traps or quantum wells [A.D. Greentree et al., Phys. Rev. B 70, 235317 (2004); K. Eckert et al., Phys. Rev. A 70, 023606 (2004)]. The direct visualization of CTAP wavepacket dynamics enabled by our simple optical system clearly shows that in the counterintuitive passage scheme light waves tunnel between the two outer wells without appreciable excitation of the middle well.Comment: submitted for publicatio

    Heavy-quark contribution to the proton's magnetic moment

    We study the contribution to the proton's magnetic moment from a heavy quark sea in quantum chromodynamics. The heavy quark is integrated out perturbatively to obtain an effective dimension-6 magnetic moment operator composed of three gluon fields. The leading contribution to the matrix element in the proton comes from a quadratically divergent term associated with a light-quark tensor operator. With an approximate knowledge of the proton's tensor charge, we conclude that a heavy sea-quark contribution to the proton's magnetic moment is positive in the asymptotic limit. We comment on the implication of this result for the physical strange quark.Comment: 4 pages, 2 figure

    Carrier-envelope offset frequency measurement by means of an external optical resonator

    A general-purpose method based on the implementation of the asymmetric Pound-Drever-Hall (PDH) technique is proposed to measure the carrier-envelope offset (CEO) frequency of a mode-locked laser using an external optical cavity. By analyzing the synchronously demodulated signal of the spectrally filtered cavity reflection when the optical resonator is locked to the mode-locked laser, a discriminating signal depending on the relative frequency offset between the mode-locked and optical cavity comb-like spectra is obtained. For a given geometry and group delay dispersion (GDD) of the cavity parameters (i.e., a known cavity mode offset), this signal can be used to retrieve the laser CEO. This approach turns out to be advantageous in terms of setup complexity with respect to other well-known techniques that rely on non-linear frequency generation, such as f-2f interferometers. In addition, this method can be used to precisely determine the laser-cavity spectral coupling, which is an important topic in cavity-enhanced spectroscopy and non-linear optics applications. After the theoretical description of the generalized asymmetric PDH signal, an experimental validation of the proposed method is reported using an Er-doped fiber frequency comb source centered at 1,550 nm, with a repetition rate of 250 MHz, locked to a linear optical cavity with a 1 GHz free spectral range. The theoretical effect of the GDD is confirmed experimentally using different cavity configurations. Moreover, the comparison with the CEO frequency values measured using an f-2f interferometer demonstrates the feasibility of the proposed method
