    Seasonal variation in the diet and the morphometric parameters of the genus Pseudophoxinus sp. (Cyprinidae) in Eastern Algeria

    The diet and the morphometric parameters of the Pseudophoxinus sp. (Cyprinidae) in the El Mellah ravine (M’sila, Algeria) were studied by determining body measurements and analyzing the contents of digestive tracts of 320 fish from December 2018 to November 2019. The obtained results show marked diversity within the fish species diets, which can be grouped into three main fractions: (i) a plant fraction; (ii) an animal fraction composed mainly of invertebrate prey; and (iii) fraction consisting of fish eggs (digested by males during the breeding season). The plant fraction was present in all the analyzed digestive tracts, mainly being composed of phytoplankton (algae), namely, Spirogyra sp., Melosira sp. and Navicula sp. with the total relative abundances of 38.7%, 34.4% and 4.1%, respectively. On the other hand, the animal fraction was represented by the Chironomus sp., Austrosimulium sp., and Ecdyonurus sp. dipterans with a total relative abundance of 7.3%, 6.7% and 5.9%, respectively. Pseudophoxinus sp. eggs can also be considered as part of this fraction with the total relative abundance of 2.9%. The age of the fish seems to have an influence on their feeding behaviour. The juveniles of Pseudophoxinus sp. in the study area had a purely phytophagous diet, while adults showed a phytophagous behaviour with an omnivorous tendency. The present study shows that not only the age of the fish but also the seasonal changes (climatic conditions and entry into the breeding season) may have a significant influence on the diet and morphometric parameters of Pseudophoxinus sp. in El Mellah ravine

    Seeds Potentialities of Medicks in Sub-Humid Area to be used in Steppe Zone

    New pulse crops varieties more productive as medics should be made available to breeders located in semi-arid region of Algeria. So, and through two years of testing, pods yield and seeds production of twenty populations belonging to species M.intertexta , M.ciliaris, M.polymorpha , M.Truncatula and an introduced population M.muricoleptis is an Australian cultivar (Jemalong) are appreciated.Pods' yields of year 2013 vary between 78,66g/m² with poly27 and 3637,33g/m² with I107. Values of the second essay (2015) are different, they vary between 40,89g /m² with Poly236 and 464,36 g/m² with I11.The cultivar Jemalong offers a yield of 172,4 g/m². The corresponding seed yields also show a low production in year 2015. The ratio leaf / stem varies between 1,03 and 5. The average of yields in dry matter of 27 populations in 2013 was of 457,79g/m2 against 127,41 g/m² in 18 populations in 2015. Jemalong cultivar records a yield of 12,8g/m². For the same dose of seed, number of plants by square meter varies between 44 and 112. Number of stems by square meter varies between 136 in C204 and 420 in I52..The average width of population's ramifications varies between 13cm in Tr334 and 44 cm in I52, The Jemalong cultivar offers an average of 17cm. So, production results of medicks depend of weather conditions in littoral zone of lower altitude than 600-700m. In steppe zones, we recommend to make tests in situ with these same populations

    Chemical composition analysis of essential oils of four plants from Aurès region of Algeria and their antibacterial and antibiofilm activities against coagulase-negative staphylococci

    Background: The altitudinal and geographical variability of the Aurès mountains of Algeria favored the existence of some endemic and rare varieties of medicinal plants. The aim of the present work is to determine the chemical composition, antimicrobial and antibiofilm properties of the  essential oils (EOs) from aerial parts of four medicinal plants from Aurès region of Algeria; Juniperus thurifera L., Juniperus oxycedrus L., Salvia   officinalis L. and Thymus ciliatus ssp. munbyanus (Boiss. & Reut.) Batt. on coagulase negative staphylococci (CoNS) isolates. Methodology: Extraction of EOs from the four plant materials was carried out by hydro-distillation, and the EO yield expressed in gram of the distillate per 100 grams of dry matter. The chemical composition of the EOs was analyzed by gas chromatography-mass spectrometry (GC-MS) method. In vitro antibacterial and antibiofilm activities of the EOs were evaluated against CoNS previously isolated at the Anti-Cancer Center of Batna, Algeria using the agar disc diffusion assay and biofilm inhibition study, respectively. Minimum inhibitory concentration (MIC) and minimum  bacterial concentration (MBC) of the EOs of S. officinalis L. and T. ciliatus ssp. munbyanus were determined by the dilution method. Results: Twenty-seven and 41 compounds rich in monoterpene hydrocarbons were identified from J. oxycedrus and J. thurifera plants respectively, while 45 and 32 compounds, constituted mainly by oxygenated monoterpenes, were identified from S. officinalis L. and T. ciliatus ssp. munbyanus, respectively. The EOs of T. ciliatus ssp. munbyanus showed the most inhibitory activity of all the four plants on CoNS isolates (n=66) with mean  inhibition zone diameter of 24.99±6.29mm, and mean MIC and MBC values of 2.65±3.77mg/ml and 5.31±7.41mg/ml respectively, followed by S.  officinalis L., with mean inhibition zone diameter of 13.38± 6.52mm, and mean MIC and MBC values of 27.53±28.2 mg/ml and 31.97±33.19 mg/ml  respectively (p<0.0001 by one-way ANOVA). Also, percentage biofilm inhibition of CoNS isolates (n=59) was high for EOs of T. ciliatus ssp. munbyanus  (65.63±10.71%) and S. officinalis L. (53.13±5.83%), although was significantly higher for T. ciliatus ssp. munbyanus compared to S. officinalis L. (p<0.0001, t=7.874). Conclusion: Essential oils from T. ciliatus ssp. munbyanus and S. officinalis L. could represent an alternative to classical antibiotics against planktonic cells and biofilms of CoNS

    Caractéristiques pétrologiques et géochimiques des roches magmatiques d’El Aouana, NE algérien = Petrological and geochemical characteristics of the magmatic rocks from El Aouana, NE Algeria

    The El Aouana igneous rocks are part of the Miocene magmatic suite that extends from Northern Tunisia to Morocco through the Algerian coast in the Maghrebin chain. These rocks are composed of volcanic (andesites and dacites) and subvolcanic (micodiorites and microgranodiorites) lithologies that intruded both the Cretaceous and Oligo-Miocene nappes, and the Miocene post-nappe sediments. The andesites are composed of plagioclase, amphibole and pyroxene phenocrysts that are set in a microlithic groundmass. The dacites are plagioclase-rich and rare, highly altered ferromagnesian minerals. The microdiorites and microgranodiorites are hornblende-bea- ring rocks with plagioclase, pyroxene and rare biotite phenocrysts that are set in a microcrystalline groundmass. Geochemical observations show that the rocks are calc-alkaline with I-type affinity. They are enriched in large ion lithophile elements (LILE) and light rare earth elements (LREE) compared to high field strength elements (HFSE) and heavy rare earth elements (HREE). The negative Nb, P and Ti anomalies observed on the multi-element patterns are imprints of magmas originated from subduction zones. Field, petrological and geochemical investi- gations show that the El Aouana Miocene igneous rocks are emplaced in a post-collisionnal setting. These rocks show similarities with the metaluminous, post-collisionnel granitoids of north-eastern Algeria which are thought to have been derived from a metasomatized mantle source as a consequence of ‘slab break-off’ underneath the North African margin

    Biostratigraphie, géochimie et réponse des composantes microfauniques aux variations environnementales au passage Cénomanien-Turonien à Thénièt El Manchar (Monts de Bellezma, Batna, NE Algérie)

    The present study deals with the biostratigraphic and geochemical analysis of the Cenomanian-Turonian deposits of the Thénièt El Manchar district in the Bellezma-Batna Range, and their vertical fluctuations. A 180 m-thick profile is described in this area. This reference section encompasses the ‘Marnes de Smail’ Formation, subdivided into four lithostratigraphic units (IA, IB, IC, and ID respectively) and dated of the Cenomanian (ammonites and coral), and the base of the ‘Dolomies de l’Oued Skhoun’ Formation (unit IIA), dated of the Lower Turonian (planktonic foraminifers). Both qualitative and quantitative analysis of foraminifers (planktonic and/or benthonic), ostracodes and of geochemical indices, lead us to reconstitute the evolution of both palaeoenvironmental and palaeobathymetrical changes during this time interval. Subsequently units IA, IB, IC and the lower part of unit ID (all Cenomanian) are displaying agglutinated benthonic foraminifers and complete ostracode shells, as evidence of a shelfal environment and weak hydrodynamism. Within these deposits microfaunal assemblages display a low-to-medium species diversity and a relatively high specimen abundance, supporting evidence of normal trophic conditions and water oxygenation. The upper part of unit ID (topmost Cenomanian), and unit IIA (basal Turonian) are documenting an environmental setting into deeper conditions, supported by the standard event succession already recorded in northern Africa, namely: the abundance of planktonic foraminifers, the occurrence of ‘filaments’ and the sudden reduction of ostracofauna. Furthermore the total carbonate isotope fluctuations (δ13C and δ18O) are documenting isotopic anomalies related to palaeoenvironmental changes. The δ13C results, coupled with those of TOC, are evidencing a low primary productivity while the δ18O data are supporting a temperature rise as the main potential drive of the onset of OAE2. These paleoenvironmental assertions are consistent with the regional paleogeographic context and are highlighting tethysian features.[fr] La présente étude livre une analyse biostratigraphique et géochimique des dépôts cénomano-turoniens de la région Thénièt El Manchar, située dans les Monts de Bellezma-Batna, et leur évolution verticale. Une coupe (TM), épaisse de 180 m, est décrite dans ce secteur. Elle correspond à la Formation des Marnes de Smail, subdivisée en quatre unités litho-stratigraphiques (IA, IB, IC, ID), datée du Cénomanien grâce à la présence d’ammonites et de corail, et à la base de la Formation des Dolomies de l’Oued Skhoun (unité, IIA), datée de la base du Turonien au moyen de foraminifères planctoniques. L’analyse qualitative et quantitative des foraminifères (planctoniques et/ou benthiques), des ostracodes et des marqueurs géochimiques, permet de reconstituer l’évolution du paléo-environnement et les variations paléo-bathymétriques au cours de l’intervalle stratigraphique concerné. Ainsi, les unités IA-IB-IC et la partie inférieure de l’unité ID (Cénomanien) sont dominées par des associations de foraminifères benthiques agglutinés et des carapaces entières d’ostracodes, témoignant d’un milieu de plate-forme et d’un faible hydrodynamisme. Dans ces dépôts, les associations montrent une diversité spécifique faible à moyenne et une abondance plus ou moins élevée, témoignant de conditions trophiques et d’oxygénation considérées comme normales. Les derniers termes de l’unité ID (sommet du Cénomanien) et l’unité IIA (base du Turonien) marquent, quant à eux, une évolution vers des conditions plus profondes, comme le montre la succession standard des événements déjà reconnus en Afrique du nord, à savoir: l’abondance de foraminifères planctoniques, la présence de ‘filaments’ et une réduction drastique de la faune d’ostracodes. En outre, les courbes isotopiques du carbone (δ13C) et de l’oxygène (δ18O) des carbonates mettent en évidence des anomalies isotopiques relatives aux modifications paléo-environnementales. Les données du δ13C et celles du COT indiquent une productivité primaire faible. Les données du δ18O, quant à elles, indiquent une augmentation des paléo-températures, cause principale du déclenchement de l’EAO 2. Ces interprétations paléo-environnementales s’accordent avec les données connues du contexte paléogéographique régional et mettent en exergue les spécificités téthysiennes

    Transcriptional Repressor Gfi1 Integrates Cytokine-Receptor Signals Controlling B-Cell Differentiation

    Hematopoietic stem cell differentiation is specified by cytokines and transcription factors, but the mechanisms controlling instructive and permissive signalling networks are poorly understood. We provide evidence that CLP1-dependent IL7-receptor mediated B cell differentiation is critically controlled by the transcriptional repressor Gfi1. Gfi1-deficient progenitor B cells show global defects in IL7Rα-dependent signal cascades. Consequently, IL7-dependent trophic, proliferative and differentiation-inducing responses of progenitor B cells are perturbed. Gfi1 directly regulates expression levels of IL7Rα and indirectly controls STAT5 signalling via expression of SOCS3. Thus, Gfi1 selectively specifies IL7-dependent development of B cells from CLP1 progenitors, providing clues to the transcriptional networks integrating cytokine signals and lymphoid differentiation

    The Female Lower Genital Tract Is a Privileged Compartment with IL-10 Producing Dendritic Cells and Poor Th1 Immunity following Chlamydia trachomatis Infection

    While a primary genital tract infection with C. trachomatis stimulates partial-protection against re-infection, it may also result in severe inflammation and tissue destruction. Here we have dissected whether functional compartments exist in the genital tract that restrict Th1-mediated protective immunity. Apart from the Th1-subset, little is known about the role of other CD4+ T cell subsets in response to a genital tract chlamydial infection. Therefore, we investigated CD4+ T cell subset differentiation in the genital tract using RT-PCR for expression of critical transcription factors and cytokines in the upper (UGT) and lower genital tract (LGT) of female C57BL/6 mice in response to C. trachomatis serovar D infection. We found that the Th1 subset dominated the UGT, as IFN-γ and T-bet mRNA expression were high, while GATA-3 was low following genital infection with C. trachomatis serovar D. By contrast, IL-10 and GATA-3 mRNA dominated the LGT, suggesting the presence of Th2 cells. These functional compartments also attracted regulatory T cells (Tregs) differently as increased FoxP3 mRNA expression was seen primarily in the UGT. Although IL-17A mRNA was somewhat up-regulated in the LGT, no significant change in RORγ-t mRNA expression was observed, suggesting no involvement of Th17 cells. The dichotomy between the LGT and UGT was maintained during infection by IL-10 because in IL-10-deficient mice the distinction between the two compartments was completely lost and a dramatic shift to the predominance of Th1 cells in the LGT occurred. Unexpectedly, the major source of IL-10 was CD11c+ CD11b+ DC, probably creating an anti-inflammatory privileged site in the LGT

    Oxygen Tension Is a Determinant of the Matrix-Forming Phenotype of Cultured Human Meniscal Fibrochondrocytes

    BACKGROUND: Meniscal cartilage displays a poor repair capacity, especially when injury is located in the avascular region of the tissue. Cell-based tissue engineering strategies to generate functional meniscus substitutes is a promising approach to treat meniscus injuries. Meniscus fibrochondrocytes (MFC) can be used in this approach. However, MFC are unable to retain their phenotype when expanded in culture. In this study, we explored the effect of oxygen tension on MFC expansion and on their matrix-forming phenotype. METHODOLOGY/PRINCIPAL FINDINGS: MFC were isolated from human menisci followed by basic fibroblast growth factor (FGF-2) mediated cell expansion in monolayer culture under normoxia (21%O(2)) or hypoxia (3%O(2)). Normoxia and hypoxia expanded MFC were seeded on to a collagen scaffold. The MFC seeded scaffolds (constructs) were cultured in a serum free chondrogenic medium for 3 weeks under normoxia and hypoxia. Constructs containing normoxia-expanded MFC were subsequently cultured under normoxia while those formed from hypoxia-expanded MFC were subsequently cultured under hypoxia. After 3 weeks of in vitro culture, the constructs were assessed biochemically, histologically and for gene expression via real-time reverse transcription-PCR assays. The results showed that constructs under normoxia produced a matrix with enhanced mRNA ratio (3.5-fold higher; p<0.001) of collagen type II to I. This was confirmed by enhanced deposition of collagen II using immuno-histochemistry. Furthermore, the constructs under hypoxia produced a matrix with higher mRNA ratio of aggrecan to versican (3.5-fold, p<0.05). However, both constructs had the same capacity to produce a glycosaminoglycan (GAG) -specific extracellular matrix. CONCLUSIONS: Our data provide evidence that oxygen tension is a key player in determining the matrix phenotype of cultured MFC. These findings suggest that the use of normal and low oxygen tension during MFC expansion and subsequent neo-tissue formation cultures may be important in engineering different regions of the meniscus
