    Is it possible to estimate aerosol optical depth from historic colour paintings?

    The idea of estimating stratospheric aerosol optical thickness from the twilight colours in historic paintings – particularly under conditions of volcanically enhanced stratospheric aerosol loading – is very tantalizing because it would provide information on the stratospheric aerosol loading over a period of several centuries. This idea has in fact been applied in a few studies in order to provide quantitative estimates of the aerosol optical depth after some of the major volcanic eruptions that occurred during the past 500 years. In this study we critically review this approach and come to the conclusion that the uncertainties in the estimated aerosol optical depths are so large that the values have to be considered questionable. We show that several auxiliary parameters – which are typically poorly known for historic eruptions – can have a similar effect on the red–green colour ratio as a change in optical depth typically associated with eruptions such as, for example, Tambora in 1815 or Krakatoa in 1883. Among the effects considered here, uncertainties in the aerosol particle size distribution have the largest impact on the colour ratios and hence the aerosol optical depth estimate. For solar zenith angles exceeding 80∘, uncertainties in the stratospheric ozone amount can also have a significant impact on the colour ratios. In addition, for solar zenith angles exceeding 90∘ the colour ratios exhibit a dramatic dependence on solar zenith angle, rendering the estimation of aerosol optical depth highly challenging. A quantitative determination of the aerosol optical depth may be possible for individual paintings for which all relevant parameters are sufficiently well constrained in order to reduce the related errors.</p

    Using Canonical Correlation Analysis to Discover Genetic Regulatory Variants

    Background: Discovering genetic associations between genetic markers and gene expression levels can provide insight into gene regulation and, potentially, mechanisms of disease. Such analyses typically involve a linkage or association analysis in which expression data are used as phenotypes. This approach leads to a large number of multiple comparisons and may therefore lack power. We assess the potential of applying canonical correlation analysis to partitioned genomewide data as a method for discovering regulatory variants. Methodology/Principal Findings: Simulations suggest that canonical correlation analysis has higher power than standard pairwise univariate regression to detect single nucleotide polymorphisms when the expression trait has low heritability. The increase in power is even greater under the recessive model. We demonstrate this approach using the Childhood Asthma Management Program data. Conclusions/Significance: Our approach reduces multiple comparisons and may provide insight into the complex relationships between genotype and gene expression

    Profile instabilities of the millisecond pulsar PSR J1022+1001

    We present evidence that the integrated profiles of some millisecond pulsars exhibit severe changes that are inconsistent with the moding phenomenon as known from slowly rotating pulsars. We study these profile instabilities in particular for PSR J1022+1001 and show that they occur smoothly, exhibiting longer time constants than those associated with moding. In addition, the profile changes of this pulsar seem to be associated with a relatively narrow-band variation of the pulse shape. Only parts of the integrated profile participate in this process which suggests that the origin of this phenomenon is intrinsic to the pulsar magnetosphere and unrelated to the interstellar medium. A polarization study rules out profile changes due to geometrical effects produced by any sort of precession. However, changes are observed in the circularly polarized radiation component. In total we identify four recycled pulsars which also exhibit instabilities in the total power or polarization profiles due to an unknown phenomenon (PSRs J1022+1001, J1730-2304, B1821-24, J2145-0750). The consequences for high precision pulsar timing are discussed in view of the standard assumption that the integrated profiles of millisecond pulsars are stable. As a result we present a new method to determine pulse times-of-arrival that involves an adjustment of relative component amplitudes of the template profile. Applying this method to PSR J1022+1001, we obtain an improved timing solution with a proper motion measurement of -17 \pm 2 mas/yr in ecliptic longitude.     Non-cross-linked biological mesh in complex abdominal wall hernia: a cohort study

    Purpose: Complex abdominal wall hernia repair (CAWHR) is a challenging procedure. Mesh prosthesis is indicated, but the use of synthetic mesh in a contaminated area may add to overall morbidity. Biological meshes may provide a solution, but little is known about long-term results. The aim of our study was to evaluate clinical efficacy and patient satisfaction following Strattice™ (PADM) placement. Methods: In this cohort study, all patients operated for CAWHR with PADM in three large community hospitals in Germany were included. Patients underwent abdominal examination, an ultrasound was performed, and patients completed quality-of-life questionnaires. The study was registered in ClinicalTrials.gov under Identifier NCT02168231. Results: Twenty-seven patients were assessed (14 male, age 67.5 years, follow-up 42.4 months). The most frequent postoperative complication was wound infection (39.1%). In no case, the PADM had to be removed. Four patients had passed away. During outpatient clinic visit, six out of 23 patients (26.1%) had a recurrence of hernia, one patient had undergone reoperation. Five patients (21.7%) had bulging of the abdominal wall. Quality-of-life questionnaires revealed that patients judged their scar with a median 3.5 out of 10 points (0 = best) and judged their restrictions during daily activities with a median of 0 out of 10

    Spin-wave Scattering in the Effective Lagrangian Perspective

    Nonrelativistic systems exhibiting collective magnetic behavior are analyzed in the framework of effective Lagrangians. The method, formulating the dynamics in terms of Goldstone bosons, allows to investigate the consequences of spontaneous symmetry breaking from a unified point of view. Low energy theorems concerning spin-wave scattering in ferro- and antiferromagnets are established, emphasizing the simplicity of actual calculations. The present work includes approximate symmetries and discusses the modification of the low energy structure imposed by an external magnetic and an anisotropy field, respectively. Throughout the paper, analogies between condensed matter physics and Lorentz-invariant theories are pointed out, demonstrating the universal feature of the effective Lagrangian technique.Comment: Published versio

    On the Analysis of Genome-Wide Association Studies in Family-Based Designs: A Universal, Robust Analysis Approach and an Application to Four Genome-Wide Association Studies

    For genome-wide association studies in family-based designs, we propose a new, universally applicable approach. The new test statistic exploits all available information about the association, while, by virtue of its design, it maintains the same robustness against population admixture as traditional family-based approaches that are based exclusively on the within-family information. The approach is suitable for the analysis of almost any trait type, e.g. binary, continuous, time-to-onset, multivariate, etc., and combinations of those. We use simulation studies to verify all theoretically derived properties of the approach, estimate its power, and compare it with other standard approaches. We illustrate the practical implications of the new analysis method by an application to a lung-function phenotype, forced expiratory volume in one second (FEV1) in 4 genome-wide association studies

    Idiopathic giant abdominal lymph cyst: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Giant lymph cysts are a relatively frequent complication after surgical procedures in the abdomen, often after kidney transplantation, but there are also cases after pelvic surgery such as lymphadenectomy and others. In the recent literature, there have been no reported cases of idiopathic giant lymphocyst.</p> <p>Case presentation</p> <p>We present the case of a 76-year-old Caucasian man who had a lymph cyst he had known of for more than 15 years. Laparoscopic treatment was necessary because of hydronephrosis of the left kidney.</p> <p>Conclusion</p> <p>This case shows that laparoscopic drainage and partial resection of the lymph cyst is a safe and effective treatment.</p

    Randomised controlled comparison of the Health Survey Short Form (SF-12) and the Graded Chronic Pain Scale (GCPS) in telephone interviews versus self-administered questionnaires. Are the results equivalent?

    <p>Abstract</p> <p>Background</p> <p>The most commonly used survey methods are self-administered questionnaires, telephone interviews, and a mixture of both. But until now evidence out of randomised controlled trials as to whether patient responses differ depending on the survey mode is lacking. Therefore this study assessed whether patient responses to surveys depend on the mode of survey administration. The comparison was between mailed, self-administered questionnaires and telephone interviews.</p> <p>Methods</p> <p>A four-armed, randomised controlled two-period change-over design. Each patient responded to the same survey twice, once in written form and once by telephone interview, separated by at least a fortnight. The study was conducted in 2003/2004 in Germany. 1087 patients taking part in the German Acupuncture Trials (GERAC cohort study), who agreed to participate in a survey after completing acupuncture treatment from an acupuncture-certified family physician for headache, were randomised. Of these, 823 (664 women) from the ages of 18 to 83 (mean 51.7) completed both parts of the study. The main outcome measure was the comparison of the scores on the 12-Item Short-Form Health Survey (SF-12) and the Graded Chronic Pain Scale (GCPS) questionnaire for the two survey modes.</p> <p>Results</p> <p>Computer-aided telephone interviews (CATI) resulted in significantly fewer missing data (0.5%) than did mailed questionnaires (2.8%; p < 0.001). The analysis of equivalence revealed a difference between the survey modes only for the SF-12 mental scales. On average, reported mental status score was 3.5 score points (2.9 to 4.0) lower on the self-administered questionnaire compared to the telephone interview. The order of administration affected results. Patients who responded to the telephone interview first reported better mental health in the subsequent paper questionnaire (mean difference 2.8 score points) compared to those who responded to the paper questionnaire first (mean difference 4.1 score points).</p> <p>Conclusion</p> <p>Despite the comparatively high cost of telephone interviews, they offer clear advantages over mailed self-administered questionnaires as regards completeness of data. Only items concerning mental status were dependent on the survey mode and sequence of administration. Items on physical status were not affected. Normative data for standardized telephone questionnaires could contribute to a better comparability with the results of the corresponding standardized paper questionnaires.</p

    Constrained mixture estimation for analysis and robust classification of clinical time series

    Motivation: Personalized medicine based on molecular aspects of diseases, such as gene expression profiling, has become increasingly popular. However, one faces multiple challenges when analyzing clinical gene expression data; most of the well-known theoretical issues such as high dimension of feature spaces versus few examples, noise and missing data apply. Special care is needed when designing classification procedures that support personalized diagnosis and choice of treatment. Here, we particularly focus on classification of interferon-β (IFNβ) treatment response in Multiple Sclerosis (MS) patients which has attracted substantial attention in the recent past. Half of the patients remain unaffected by IFNβ treatment, which is still the standard. For them the treatment should be timely ceased to mitigate the side effects

    Post-stroke inhibition of induced NADPH oxidase type 4 prevents oxidative stress and neurodegeneration

    Ischemic stroke is the second leading cause of death worldwide. Only one moderately effective therapy exists, albeit with contraindications that exclude 90% of the patients. This medical need contrasts with a high failure rate of more than 1,000 pre-clinical drug candidates for stroke therapies. Thus, there is a need for translatable mechanisms of neuroprotection and more rigid thresholds of relevance in pre-clinical stroke models. One such candidate mechanism is oxidative stress. However, antioxidant approaches have failed in clinical trials, and the significant sources of oxidative stress in stroke are unknown. We here identify NADPH oxidase type 4 (NOX4) as a major source of oxidative stress and an effective therapeutic target in acute stroke. Upon ischemia, NOX4 was induced in human and mouse brain. Mice deficient in NOX4 (Nox4(-/-)) of either sex, but not those deficient for NOX1 or NOX2, were largely protected from oxidative stress, blood-brain-barrier leakage, and neuronal apoptosis, after both transient and permanent cerebral ischemia. This effect was independent of age, as elderly mice were equally protected. Restoration of oxidative stress reversed the stroke-protective phenotype in Nox4(-/-) mice. Application of the only validated low-molecular-weight pharmacological NADPH oxidase inhibitor, VAS2870, several hours after ischemia was as protective as deleting NOX4. The extent of neuroprotection was exceptional, resulting in significantly improved long-term neurological functions and reduced mortality. NOX4 therefore represents a major source of oxidative stress and novel class of drug target for stroke therapy
