A continuous isotropic-nematic liquid crystalline transition of F-actin solutions
The phase transition from the isotropic (I) to nematic (N) liquid crystalline
suspension of F-actin of average length m or above was studied by local
measurements of optical birefringence and protein concentration. Both
parameters were detected to be continuous in the transition region, suggesting
that the I-N transition is higher than 1st order. This finding is consistent
with a recent theory by Lammert, Rokhsar & Toner (PRL, 1993, 70:1650),
predicting that the I-N transition may become continuous due to suppression of
disclinations. Indeed, few line defects occur in the aligned phase of F-actin.
Individual filaments in solutions of a few mg/ml F-actin undergo fast
translational diffusion along the filament axis, whereas both lateral and

Hepatic consequences of COVID-19 infection. Lapping or biting?
The outbreak of coronavirus disease 2019 (COVID-19) starting last December in China placed emphasis on liver involvement during infection. This review discusses the underlying mechanisms linking COVID-19 to liver dysfunction, according to recent available information, while waiting further studies. The manifestations of liver damage are usually mild (moderately elevated serum aspartate aminotransferase activities), and generally asymptomatic. Few patients can still develop severe liver problems, and therapeutic options can be limited. Liver dysfunction may affect about one-third of the patients, with prevalence greater in men than women, and in elderly. Mechanisms of damage are complex and include direct cholangiocyte damage and other coexisting conditions such as the use of antiviral drugs, systemic inflammatory response, respiratory distress syndrome-induced hypoxia, sepsis, and multiple organ dysfunction. During new COVID-19 infections, liver injury may be observed. If liver involvement appears during COVID-19 infection, however, attention is required. This is particularly true if patients are older or have a pre-existing history of liver diseases. Pre-endoscopy SARS-CoV-2 testing strategy during COVID-19 pandemic: the care must go on
Background: In response to the COVID-19 pandemic, endoscopic societies initially recommended reduction of
endoscopic procedures. In particular non-urgent endoscopies should be postponed. However, this might lead to
unnecessary delay in diagnosing gastrointestinal conditions.
Methods: Retrospectively we analysed the gastrointestinal endoscopies performed at the Central Endoscopy Unit
of Saarland University Medical Center during seven weeks from 23 March to 10 May 2020 and present our real-world
single-centre experience with an individualized rtPCR-based pre-endoscopy SARS-CoV-2 testing strategy. We also
present our experience with this strategy in 2021.
Results: Altogether 359 gastrointestinal endoscopies were performed in the initial period. The testing strategy enabled us to conservatively handle endoscopy programme reduction (44% reduction as compared 2019) during the frst
wave of the COVID-19 pandemic. The results of COVID-19 rtPCR from nasopharyngeal swabs were available in 89%
of patients prior to endoscopies. Apart from six patients with known COVID-19, all other tested patients were negative. The frequencies of endoscopic therapies and clinically signifcant fndings did not difer between patients with
or without SARS-CoV-2 tests. In 2021 we were able to unrestrictedly perform all requested endoscopic procedures
(>5000 procedures) by applying the rtPCR-based pre-endoscopy SARS-CoV-2 testing strategy, regardless of next
waves of COVID-19. Only two out-patients (1893 out-patient procedures) were tested positive in the year 2021.
Conclusion: A structured pre-endoscopy SARS-CoV-2 testing strategy is feasible in the clinical routine of an endoscopy unit. rtPCR-based pre-endoscopy SARS-CoV-2 testing safely allowed unrestricted continuation of endoscopic
procedures even in the presence of high incidence rates of COVID-19. Given the low frequency of positive tests, the
absolute efect of pre-endoscopy testing on viral transmission may be low when FFP-2 masks are regularly used

TLR4 Deficiency Protects against Hepatic Fibrosis and Diethylnitrosamine-Induced Pre-Carcinogenic Liver Injury in Fibrotic Liver
Background
The development of hepatocellular carcinoma (HCC) is a common consequence of advanced liver fibrosis but the interactions between fibrogenesis and carcinogenesis are still poorly understood. Recently it has been shown that HCC promotion depends on Toll-like receptor (TLR) 4. Pre-cancerogenous events can be modelled in mice by the administration of a single dose of diethylnitrosamine (DEN), with HCC formation depending amongst others on interleukin (IL) 6 production. Mice lacking the hepatocanalicular phosphatidylcholine transporter ABCB4 develop liver fibrosis spontaneously, resemble patients with sclerosing cholangitis due to mutations of the orthologous human gene, and represent a valid model to study tumour formation in pre-injured cholestatic liver. The aim of this study was to investigate DEN-induced liver injury in TLR4-deficient mice with biliary fibrosis.
Methods
ABCB4-deficient mice on the FVB/NJ genetic background were crossed to two distinct genetic backgrounds (TLR4-sufficient C3H/HeN and TLR4-deficient C3H/HeJ) for more than 10 generations. The two congenic knockout and the two corresponding wild-type mouse lines were treated with a single dose of DEN for 48 hours. Phenotypic differences were assessed by measuring hepatic collagen contents, inflammatory markers (ALT, CRP, IL6) as well as hepatic apoptosis (TUNEL) and proliferation (Ki67) rates.
Results
Hepatic collagen accumulation is significantly reduced in ABCB4-/-:TLR4-/-double-deficient mice. After DEN challenge, apoptosis, proliferation and inflammatory markers are decreased in TLR4-deficient in comparison to TLR4-sufficient mice. When combining ABCB4 and TLR4 deficiency with DEN treatment, hepatic IL6 expression and proliferation rates are lowest in fibrotic livers from the double-deficient line. Consistent with these effects, selective digestive tract decontamination in ABCB4-/- mice also led to reduced tumor size and number after DEN.
Conclusion
This study demonstrates that liver injury upon DEN challenge depends on pre-existing fibrosis and genetic background. The generation of ABCB4-/: TLR4-/- double-deficient mice illustrates that TLR4-deficiency protects against hepatic injury in a preclinical mouse model of chronic liver disease
Generic theory of colloidal transport
We discuss the motion of colloidal particles relative to a two component
fluid consisting of solvent and solute. Particle motion can result from (i) net
body forces on the particle due to external fields such as gravity; (ii) slip
velocities on the particle surface due to surface dissipative phenomena. The
perturbations of the hydrodynamic flow field exhibits characteristic
differences in cases (i) and (ii) which reflect different patterns of momentum
flux corresponding to the existence of net forces, force dipoles or force
quadrupoles. In the absence of external fields, gradients of concentration or
pressure do not generate net forces on a colloidal particle. Such gradients can
nevertheless induce relative motion between particle and fluid. We present a
generic description of surface dissipative phenomena based on the linear
response of surface fluxes driven by conjugate surface forces. In this
framework we discuss different transport scenarios including self-propulsion
via surface slip that is induced by active processes on the particle surface.

Nonperturbative approach to the Hubbard model in C60 cluster
We propose a computational scheme for the Hubbard model in the C60 cluster in
which the interaction with the Fermi sea of charges added to the neutral
molecule is switched on sequentially. This is applied to the calculation of the
balance of charging energies, within a low-energy truncation of the space of
states which produces moderate errors for an intermediate range of the

Low phospholipid associated cholelithiasis: association with mutation in the MDR3/ABCB4 gene
Low phospholipid-associated cholelithiasis (LPAC) is characterized by the association of ABCB4 mutations and low biliary phospholipid concentration with symptomatic and recurring cholelithiasis. This syndrome is infrequent and corresponds to a peculiar small subgroup of patients with symptomatic gallstone disease. The patients with the LPAC syndrome present typically with the following main features: age less than 40 years at onset of symptoms, recurrence of biliary symptoms after cholecystectomy, intrahepatic hyperechoic foci or sludge or microlithiasis along the biliary tree. Defect in ABCB4 function causes the production of bile with low phospholipid content, increased lithogenicity and high detergent properties leading to bile duct luminal membrane injuries and resulting in cholestasis with increased serum gamma-glutamyltransferase (GGT) activity. Intrahepatic gallstones may be evidenced by ultrasonography (US), computing tomography (CT) abdominal scan or magnetic resonance cholangiopancreatography, intrahepatic hyperechogenic foci along the biliary tree may be evidenced by US, and hepatic bile composition (phospholipids) may be determined by duodenoscopy. In all cases where the ABCB4 genotyping confirms the diagnosis of LPAC syndrome in young adults, long-term curative or prophylactic therapy with ursodeoxycholic acid (UDCA) should be initiated early to prevent the occurrence or recurrence of the syndrome and its complications. Cholecystectomy is indicated in the case of symptomatic gallstones. Biliary drainage or partial hepatectomy may be indicated in the case of symptomatic intrahepatic bile duct dilatations filled with gallstones. Patients with end-stage liver disease may be candidates for liver transplantation
Off-diagonal Interactions, Hund's Rules and Pair-binding in Hubbard Molecules
We have studied the effect of including nearest-neighbor, electron-electron
interactions, in particular the off-diagonal (non density-density) terms, on
the spectra of truncated tetrahedral and icosahedral ``Hubbard molecules,''
focusing on the relevance of these systems to the physics of doped C.
Our perturbation theoretic and exact diagonalization results agree with
previous work in that the density-density term suppresses pair-binding.
However, we find that for the parameter values of interest for the
off-diagonal terms {\em enhance} pair-binding, though not enough to offset the
suppression due to the density-density term. We also find that the critical
interaction strengths for the Hund's rules violating level crossings in
C, C and C are quite insensitive to the
