The continuous refill, short-burst, hand-powered water toy
Thesis (S.B.)--Massachusetts Institute of Technology, Dept. of Mechanical Engineering, 2010.Cataloged from PDF version of thesis.Includes bibliographical references (p. 21).The water "blaster" has been one of the most popular toys because it is fun, safe, and easy to operate. The water guns have appeared in many design variations, ranging from the simple squirt gun to motorized water guns. This research proposes a new approach to water gun design. The design consists of a small single burst chamber, and an air bulb is used to pressurize the chamber between shots, creating the sense of multi-shot, large, pressurized tank. Similar to the currently popular Super Soaker, the water in this version is pressurized inside the water channel before each blast. This model also takes advantage of the pressure head between the channel and the reservoir, which is from a backpack, to have the water flow passively into the channel. The user then can pressurize the channel by hand with an air bulb and blast continuously. We implemented a prototype to demonstrate the concept.by Long Lam.S.B
A sunny future: expert elicitation of China's solar photovoltaic technologies
China has emerged as the global manufacturing center for solar photovoltaic (PV) products. Chinese firms have entered all stages of the supply chain, producing most of the installed solar modules around the world. Meanwhile, production costs are at record lows. The decisions that Chinese solar producers make today will influence the path for the solar industry and its role towards de-carbonization of global energy systems in the years to come. However, to date, there have been no assessments of the future costs and efficiency of solar PV systems produced by the Chinese PV industry. We perform an expert elicitation to assess the technological and non-technological factors that led to the success of China's silicon PV industry as well as likely future costs and performance. Experts evaluated key metrics such as efficiency, costs, and commercial viability of 17 silicon and non-silicon solar PV technologies by 2030. Silicon-based technologies will continue to be the mainstream product for large-scale electricity generation application in the near future, with module efficiency reaching as high as 23% and production cost as low as 34/MWh, allowing solar PV to be competitive with traditional energy resources like coal. The industry's future developments may be affected by overinvestment, overcapacity, and singular short-term focus.info:eu-repo/semantics/publishedVersio
China's wind electricity and cost of carbon mitigation are more expensive than anticipated
The success of China's transition to a low-carbon energy system will be key to achieve the global level of emissions reductions needed to avoid large negative consequences from climate change. China is undergoing an impressive build up of renewable capacity, in particular wind. Using data from the Clean Mechanism Development project database between 2004 and 2012, this study shows that while China made progress in bringing down the levelized cost of wind electricity and cost of carbon mitigation (CCM), serious grid-connection issues and high wind curtailment rates resulted in a levelized cost of wind electricity that is one-half to two times higher than expected, and a CCM that is four to six times higher. Sharp drop in electricity demand, utilization rate, and coal prices in recent years may lead to even higher results.info:eu-repo/semantics/publishedVersio
China's wind industry: leading in deployment, lagging in innovation
China's massive carbon emissions and air pollution concerns have led its government to embrace clean energy innovation as a means of transitioning to a more sustainable energy system. We address the question of whether China's wind industry has become an important source of clean energy technology innovation. We find that in terms of wind capacity expansion, China has delivered enormous progress, increasing its wind capacity from virtually no wind capacity in the early 2000s to 140 GW by 2015. However, in terms of innovation and cost competitiveness, the outcomes were more limited: Chinese wind turbine manufacturers have secured few international patents and achieved moderate learning rates compared to the global industry's historical learning rate. Leading China-based indigenous producers are likely to remain important global players for the foreseeable future, but further progress in reducing the cost of capital equipment may slow relative to the recent past. However, opportunities in lowering curtailment rates and improving turbine quality can reduce China's overall levelized cost of electricity for wind.info:eu-repo/semantics/publishedVersio
The 3-3-1 model with S_4 flavor symmetry
We construct a 3-3-1 model based on family symmetry S_4 responsible for the
neutrino and quark masses. The tribimaximal neutrino mixing and the diagonal
quark mixing have been obtained. The new lepton charge \mathcal{L} related to
the ordinary lepton charge L and a SU(3) charge by L=2/\sqrt{3} T_8+\mathcal{L}
and the lepton parity P_l=(-)^L known as a residual symmetry of L have been
introduced which provide insights in this kind of model. The expected vacuum
alignments resulting in potential minimization can origin from appropriate
violation terms of S_4 and \mathcal{L}. The smallness of seesaw contributions
can be explained from the existence of such terms too. If P_l is not broken by
the vacuum values of the scalar fields, there is no mixing between the exotic
and the ordinary quarks at the tree level.Comment: 20 pages, revised versio

Molecular testing for the clinical diagnosis of fibrolamellar carcinoma.
Fibrolamellar carcinoma has a distinctive morphology and immunophenotype, including cytokeratin 7 and CD68 co-expression. Despite the distinct findings, accurate diagnosis of fibrolamellar carcinoma continues to be a challenge. Recently, fibrolamellar carcinomas were found to harbor a characteristic somatic gene fusion, DNAJB1-PRKACA. A break-apart fluorescence in situ hybridization (FISH) assay was designed to detect this fusion event and to examine its diagnostic performance in a large, multicenter, multinational study. Cases initially classified as fibrolamellar carcinoma based on histological features were reviewed from 124 patients. Upon central review, 104 of the 124 cases were classified histologically as typical of fibrolamellar carcinoma, 12 cases as 'possible fibrolamellar carcinoma' and 8 cases as 'unlikely to be fibrolamellar carcinoma'. PRKACA FISH was positive for rearrangement in 102 of 103 (99%) typical fibrolamellar carcinomas, 9 of 12 'possible fibrolamellar carcinomas' and 0 of 8 cases 'unlikely to be fibrolamellar carcinomas'. Within the morphologically typical group of fibrolamellar carcinomas, two tumors with unusual FISH patterns were also identified. Both cases had the fusion gene DNAJB1-PRKACA, but one also had amplification of the fusion gene and one had heterozygous deletion of the normal PRKACA locus. In addition, 88 conventional hepatocellular carcinomas were evaluated with PRKACA FISH and all were negative. These findings demonstrate that FISH for the PRKACA rearrangement is a clinically useful tool to confirm the diagnosis of fibrolamellar carcinoma, with high sensitivity and specificity. A diagnosis of fibrolamellar carcinoma is more accurate when based on morphology plus confirmatory testing than when based on morphology alone
Etiology and Clinical Characterization of Respiratory Virus Infections in Adult Patients Attending an Emergency Department in Beijing
BACKGROUND: Acute respiratory tract infections (ARTIs) represent a serious global health burden. To date, few reports have addressed the prevalence of respiratory viruses (RVs) in adults with ARTIs attending an emergency department (ED). Therefore, the potential impact of respiratory virus infections on such patients remains unknown. METHODOLOGY/PRINCIPAL FINDINGS: To determine the epidemiological and clinical profiles of common and recently discovered respiratory viruses in adults with ARTIs attending an ED in Beijing, a 1-year consecutive study was conducted from May, 2010, to April, 2011. Nose and throat swab samples from 416 ARTI patients were checked for 13 respiratory viruses using multiple reverse transcription polymerase chain reaction(RT-PCR) assays for common respiratory viruses, including influenza viruses (Flu) A, B, and adenoviruses (ADVs), picornaviruses (PICs), respiratory syncytial virus (RSV), parainfluenza viruses (PIVs) 1-3, combined with real-time RT-PCR for human metapneumovirus (HMPV) and human coronaviruses (HCoVs, -OC43, -229E, -NL63, and -HKU1). Viral pathogens were detected in 52.88% (220/416) of patient samples, and 7.21% (30/416) of patients tested positive for more than one virus. PICs (17.79%) were the dominant agents detected, followed by FluA (16.11%), HCoVs (11.78%), and ADV (11.30%). HMPV, PIVs, and FluB were also detected (<3%), but not RSV. The total prevalence and the dominant virus infections detected differed significantly between ours and a previous report. Co-infection rates were high for HCoV-229E (12/39, 30.76%), PIC (22/74, 29.73%), ADV (12/47, 25.53%) and FluA (15/67, 22.39%). Different patterns of clinical symptoms were associated with different respiratory viruses. CONCLUSIONS: The pattern of RV involvement in adults with ARTIs attending an ED in China differs from that previously reported. The high prevalence of viruses (PIC, FluA, HCoVs and ADV) reported here strongly highlight the need for the development of safe and effective therapeutic approaches for these viruses
Metabolic therapy with PEG-arginase induces a sustained complete remission in immunotherapy-resistant melanoma
Background
Metastatic melanoma is an aggressive skin cancer with a poor prognosis. Current treatment strategies for high-stage melanoma are based around the use of immunotherapy with immune checkpoint inhibitors such as anti-PDL1 or anti-CTLA4 antibodies to stimulate anti-cancer T cell responses, yet a number of patients will relapse and die of disease. Here, we report the first sustained complete remission in a patient with metastatic melanoma who failed two immunotherapy strategies, by targeting tumour arginine metabolism.
Case presentation
A 65-year-old patient with metastatic melanoma who progressed through two immunotherapy strategies with immune checkpoint inhibitor antibodies was enrolled in a phase I study (NCT02285101) and treated with 2 mg/kg intravenously, weekly pegylated recombinant arginase (BCT-100). The patient experienced no toxicities > grade 2 and entered a complete remission which is sustained for over 30 months. RNA-sequencing identified a number of transcriptomic pathway alterations compared to control samples. The tumour had absent expression of the recycling enzymes argininosuccinate synthetase (ASS) and ornithine transcarbamylase (OTC) indicating a state of arginine auxotrophy, which was reconfirmed by immunohistochemistry, and validation in a larger cohort of melanoma tumour samples.
Conclusions
Targeting arginine metabolism with therapeutic arginase in arginine auxotrophic melanoma can be an effective salvage for the treatment of patients who fail immunotherapy
