    Projection of the daily travel of an ageing population: The Paris and Montreal case, 1975-2020

    International audienceAgeing of the population, urban sprawl and car dependency will change travel patterns. The main objective of this paper is to give elements for a better understanding of the impact of changing demographics on the long term evolution of daily mobility using demographic-based models to forecast, for the elderly population, car-ownership, trip frequency, distance traveled, average trip distance. A second objective is to measure the impact of the long term tendencies observed on the appearance of new needs of travel demand such as a rapid increase of demand-responsive transport. The paper compares two agglomerations, both in a strong ageing process, but in quite different sociocultural contexts: a large European metropolis: Paris, and a medium sized north-american city: Montreal. Many common conclusions derived from the two different cases studies reinforce the possibility of generalizing the conclusions to other situations

    European food quality schemes in everyday food consumption: An exploration of sayings and doings through pragmatic regimes of engagement

    While European consumers generally support the principles underpinning Food Quality Schemes (FQS) sales of certified products remain modest. This phenomenon is known as ‘attitude-behaviour gap’ and considerable scholarly and policy efforts have been geared towards ‘filling’ or ‘bridging’ the gap. This study aims at casting new light on this ‘discrepancy’ between consumers’ sayings and doings through a study of everyday food practices connected to FQS. We used a qualitative, multi-method research design comprising extensive ethnographic fieldwork data, gathered from 41 households across seven European countries, including interviews, walk-along tours, and food diaries, in order to understand consumers’ perceptions of FQS in relation to their everyday food consumption practices. Building on convention theory and Th®evenot’s work, we showed that food practices can be understood through different ‘regimes of engagement’, namely different ways of thinking and behaving, following different logics corresponding to varying levels of knowledge and interest. We thus argue that the ‘attitude-behaviour gap’ should be reconceptualised as the co-existence of multiple regimes of engagement, namely a dynamic and always evolving process of adjustment through which consumers understand and engage with FQS in everyday food practices

    Effect of angiotensin-converting enzyme inhibitor and angiotensin receptor blocker initiation on organ support-free days in patients hospitalized with COVID-19

    IMPORTANCE Overactivation of the renin-angiotensin system (RAS) may contribute to poor clinical outcomes in patients with COVID-19. Objective To determine whether angiotensin-converting enzyme (ACE) inhibitor or angiotensin receptor blocker (ARB) initiation improves outcomes in patients hospitalized for COVID-19. DESIGN, SETTING, AND PARTICIPANTS In an ongoing, adaptive platform randomized clinical trial, 721 critically ill and 58 non–critically ill hospitalized adults were randomized to receive an RAS inhibitor or control between March 16, 2021, and February 25, 2022, at 69 sites in 7 countries (final follow-up on June 1, 2022). INTERVENTIONS Patients were randomized to receive open-label initiation of an ACE inhibitor (n = 257), ARB (n = 248), ARB in combination with DMX-200 (a chemokine receptor-2 inhibitor; n = 10), or no RAS inhibitor (control; n = 264) for up to 10 days. MAIN OUTCOMES AND MEASURES The primary outcome was organ support–free days, a composite of hospital survival and days alive without cardiovascular or respiratory organ support through 21 days. The primary analysis was a bayesian cumulative logistic model. Odds ratios (ORs) greater than 1 represent improved outcomes. RESULTS On February 25, 2022, enrollment was discontinued due to safety concerns. Among 679 critically ill patients with available primary outcome data, the median age was 56 years and 239 participants (35.2%) were women. Median (IQR) organ support–free days among critically ill patients was 10 (–1 to 16) in the ACE inhibitor group (n = 231), 8 (–1 to 17) in the ARB group (n = 217), and 12 (0 to 17) in the control group (n = 231) (median adjusted odds ratios of 0.77 [95% bayesian credible interval, 0.58-1.06] for improvement for ACE inhibitor and 0.76 [95% credible interval, 0.56-1.05] for ARB compared with control). The posterior probabilities that ACE inhibitors and ARBs worsened organ support–free days compared with control were 94.9% and 95.4%, respectively. Hospital survival occurred in 166 of 231 critically ill participants (71.9%) in the ACE inhibitor group, 152 of 217 (70.0%) in the ARB group, and 182 of 231 (78.8%) in the control group (posterior probabilities that ACE inhibitor and ARB worsened hospital survival compared with control were 95.3% and 98.1%, respectively). CONCLUSIONS AND RELEVANCE In this trial, among critically ill adults with COVID-19, initiation of an ACE inhibitor or ARB did not improve, and likely worsened, clinical outcomes.     Junctional Neurulation: A Unique Developmental Program Shaping a Discrete Region of the Spinal Cord Highly Susceptible to Neural Tube Defects

    International audienceIn higher vertebrates, the primordium of the nervous system, the neural tube, is shaped along the rostrocaudal axis through two consecutive, radically different processes referred to as primary and secondary neurulation. Failures in neurulation lead to severe anomalies of the nervous system, called neural tube defects (NTDs), which are among the most common congenital malformations in humans. Mechanisms causing NTDs in humans remain ill-defined. Of particular interest, the thoracolumbar region, which encompasses many NTD cases in the spine, corresponds to the junction between primary and secondary neurulations. Elucidating which developmental processes operate during neurulation in this region is therefore pivotal to unraveling the etiology of NTDs. Here, using the chick embryo as a model, we show that, at the junction, the neural tube is elaborated by a unique developmental program involving concerted movements of elevation and folding combined with local cell ingression and accretion. This process ensures the topological continuity between the primary and secondary neural tubes while supplying all neural progenitors of both the junctional and secondary neural tubes. Because it is distinct from the other neurulation events, we term this phenomenon junctional neurulation. Moreover, the planar-cell-polarity member, Prickle-1, is recruited specifically during junctional neurulation and its misexpression within a limited time period suffices to cause anomalies that phenocopy lower spine NTDs in human. Our study thus provides a molecular and cellular basis for understanding the causality of NTD prevalence in humans and ascribes to Prickle-1 a critical role in lower spinal cord formation

    Exploiting transcript profiling data to decipher rice – Magnaporthe grisea compatibility mechanisms

    International audienceWhereas the molecular events associated with disease resistance are largely studied, those occurring in susceptible plants are poorly known, especially in Monocots. We chose the rice/Magnaporthe grisea system to explore the molecular modifications that are induced in susceptible plant tissues following pathogen attack. A truly compatible interaction (between O. sativa cultivar Nipponbare and M. grisea isolate FR13) at time points where infection occurs without visible symptoms was studied. Changes in rice transcript levels at 3 and 4 days post inoculation were measured using Affymetrix chips. In addition to the up-regulation of a large number of defence-related genes, this analysis revealed new features indicating extensive reprogramming of host gene expression. Microarray expression data were confirmed for 35/38 genes using quantitative RT-PCR, thus demonstrating the robustness of these transcript profiling experiments. This comprehensive survey of global gene expression in response to colonization by a virulent fungal isolate constitutes an important resource that should help us to further our understanding of the rice blast disease process
