    Bright ligand-activatable fluorescent protein for high-quality multicolor live-cell super-resolution microscopy

    We introduce UnaG as a green-to-dark photoswitching fluorescent protein capable of high-quality super-resolution imaging with photon numbers equivalent to the brightest photoswitchable red protein. UnaG only fluoresces upon binding of a fluorogenic metabolite, bilirubin, enabling UV-free reversible photoswitching with easily controllable kinetics and low background under Epi illumination. The on- and off-switching rates are controlled by the concentration of the ligand and the excitation light intensity, respectively, where the dissolved oxygen also promotes the off-switching. The photo-oxidation reaction mechanism of bilirubin in UnaG suggests that the lack of ligand-protein covalent bond allows the oxidized ligand to detach from the protein, emptying the binding cavity for rebinding to a fresh ligand molecule. We demonstrate super-resolution single-molecule localization imaging of various subcellular structures genetically encoded with UnaG, which enables facile labeling and simultaneous multicolor imaging of live cells. UnaG has the promise of becoming a default protein for high-performance super-resolution imaging.     The deglaciation of coastal areas of southeast Greenland

    Large marine-terminating glaciers around the margins of the Greenland Ice Sheet have retreated, accelerated and thinned over the last two decades. Relatively little is known about the longer term behaviour of the Greenland Ice Sheet, yet this information is valuable for assessing the significance of modern changes. We address this by reporting 11 new beryllium-10 (10Be) exposure ages from previously uninvestigated coastal areas across southeast Greenland. The new ages are combined with existing data from the region to assess the timing of glacier retreat after the Last Glacial Maximum. The results show that deglaciation occurred first in the north of the region (~68°N) and progressed southwards. This north–south progression is attributed to the influence of the warm Irminger Current on the ice margin. Areas in the south of the region were isolated from the warm waters by the shallow bathymetry of the continental shelf. This demonstrates that oceanographic forcing paced the deglaciation of southeast Greenland through the Younger Dryas and early Holocene. In most areas of southeast Greenland bedrock ages are systematically older than their counterpart boulder samples; this offset is likely the result of inherited 10Be content in bedrock surfaces. This suggests that subglacial erosion during the last glacial cycle was insufficient to completely remove pre-existing 10Be content. Alternatively, this pattern may be the signature of a substantial retreat and advance cycle prior to final Holocene deglaciation

    Weaker land–climate feedbacks from nutrient uptake during photosynthesis-inactive periods

    Terrestrial carbon–climate feedbacks depend on two large and opposing fluxes—soil organic matter decomposition and photosynthesis—that are tightly regulated by nutrients . Earth system models (ESMs) participating in the Coupled Model Intercomparison Project Phase 5 represented nutrient dynamics poorly , rendering predictions of twenty-first century carbon–climate feedbacks highly uncertain. Here, we use a new land model to quantify the effects of observed plant nutrient uptake mechanisms missing in most other ESMs. In particular, we estimate the global role of root nutrient competition with microbes and abiotic processes during periods without photosynthesis. Nitrogen and phosphorus uptake during these periods account for 45 and 43%, respectively, of annual uptake, with large latitudinal variation. Globally, night-time nutrient uptake dominates this signal. Simulations show that ignoring this plant uptake, as is done when applying an instantaneous relative demand approach, leads to large positive biases in annual nitrogen leaching (96%) and N O emissions (44%). This N O emission bias has a GWP equivalent of ~2.4 PgCO yr , which is substantial compared to the current terrestrial CO sink. Such large biases will lead to predictions of overly open terrestrial nutrient cycles and lower carbon sequestration capacity.     Socio-demographic and health-related factors associated with cognitive impairment in the elderly in Taiwan

    <p>Abstract</p> <p>Background</p> <p>Cognitive impairment is an age-related condition as the rate of cognitive decline rapidly increases with aging. It is especially important to better understand factors involving in cognitive decline for the countries where the older population is growing rapidly. The aim of this study was to examine the association between socio-demographic and health-related factors and cognitive impairment in the elderly in Taiwan.</p> <p>Methods</p> <p>We analysed data from 2119 persons aged 65 years and over who participated in the 2005 National Health Interview Survey. Cognitive impairment was defined as having the score of the Mini Mental State Examination lower than 24. The χ<sup>2 </sup>test and multiple logistic regression models were used to evaluate the association between cognitive impairment and variables of socio-demography, chronic diseases, geriatric conditions, lifestyle, and dietary factors.</p> <p>Results</p> <p>The prevalence of cognitive impairment was 22.2%. Results of multivariate analysis indicated that low education, being single, low social support, lower lipid level, history of stroke, physical inactivity, non-coffee drinking and poor physical function were associated with a higher risk of cognitive impairment.</p> <p>Conclusion</p> <p>Most of the characteristics in relation to cognitive impairment identified in our analysis are potentially modifiable. These results suggest that improving lifestyle behaviours such as regular exercise and increased social participation could help prevent or decrease the risk of cognitive impairment. Further investigations using longitudinal data are needed to clarify our findings.</p

    Long-term and realistic global change manipulations had low impact on diversity of soil biota in temperate heathland

    In a dry heathland ecosystem we manipulated temperature (warming), precipitation (drought) and atmospheric concentration of CO(2) in a full-factorial experiment in order to investigate changes in below-ground biodiversity as a result of future climate change. We investigated the responses in community diversity of nematodes, enchytraeids, collembolans and oribatid mites at two and eight years of manipulations. We used a structural equation modelling (SEM) approach analyzing the three manipulations, soil moisture and temperature, and seven soil biological and chemical variables. The analysis revealed a persistent and positive effect of elevated CO(2) on litter C:N ratio. After two years of treatment, the fungi to bacteria ratio was increased by warming, and the diversities within oribatid mites, collembolans and nematode groups were all affected by elevated CO(2) mediated through increased litter C:N ratio. After eight years of treatment, however, the CO(2)-increased litter C:N ratio did not influence the diversity in any of the four fauna groups. The number of significant correlations between treatments, food source quality, and soil biota diversities was reduced from six to three after two and eight years, respectively. These results suggest a remarkable resilience within the soil biota against global climate change treatments in the long term

    Veterans walk to beat back pain: study rationale, design and protocol of a randomized trial of a pedometer-based Internet mediated intervention for patients with chronic low back pain

    <p>Abstract</p> <p>Background</p> <p>Chronic back pain is a significant problem worldwide and may be especially prevalent among patients receiving care in the U.S. Department of Veterans Affairs healthcare system. Back pain affects adults at all ages and is associated with disability, lost workplace productivity, functional limitations and social isolation. Exercise is one of the most effective strategies for managing chronic back pain. Yet, there are few clinical programs that use low cost approaches to help patients with chronic back pain initiate and maintain an exercise program.</p> <p>Methods/Design</p> <p>We describe the design and rationale of a randomized controlled trial to assess the efficacy of a pedometer-based Internet mediated intervention for patients with chronic back pain. The intervention uses an enhanced pedometer, website and e-community to assist these patients with initiating and maintaining a regular walking program with the primary aim of reducing pain-related disability and functional interference. The study specific aims are: 1) To determine whether a pedometer-based Internet-mediated intervention reduces pain-related functional interference among patients with chronic back pain in the short term and over a 12-month timeframe. 2) To assess the effect of the intervention on walking (measured by step counts), quality of life, pain intensity, pain related fear and self-efficacy for exercise. 3) To identify factors associated with a sustained increase in walking over a 12-month timeframe among patients randomized to the intervention.</p> <p>Discussion</p> <p>Exercise is an integral part of managing chronic back pain but to be effective requires that patients actively participate in the management process. This intervention is designed to increase activity levels, improve functional status and make exercise programs more accessible for a broad range of patients with chronic back pain.</p> <p>Trial Registration Number</p> <p>NCT00694018</p
