    Nanoscale temperature measurements using non-equilibrium Brownian dynamics of a levitated nanosphere

    Einstein realised that the fluctuations of a Brownian particle can be used to ascertain properties of its environment. A large number of experiments have since exploited the Brownian motion of colloidal particles for studies of dissipative processes, providing insight into soft matter physics, and leading to applications from energy harvesting to medical imaging. Here we use optically levitated nanospheres that are heated to investigate the non-equilibrium properties of the gas surrounding them. Analysing the sphere's Brownian motion allows us to determine the temperature of the centre-of-mass motion of the sphere, its surface temperature and the heated gas temperature in two spatial dimensions. We observe asymmetric heating of the sphere and gas, with temperatures reaching the melting point of the material.     Auditory distraction during reading: A Bayesian meta-analysis of a continuing controversy

    Everyday reading occurs in different settings, such as on the train to work, in a busy cafeteria, or at home, while listening to music. In these situations, readers are exposed to external auditory stimulation from nearby noise, speech, or music that may distract them from their task and reduce their comprehension. Although many studies have investigated auditory distraction effects during reading, the results have proved to be inconsistent and sometimes even contradictory. Additionally, the broader theoretical implications of the findings have not always been explicitly considered. In the present study, we report a Bayesian meta-analysis of 65 studies on auditory distraction effects during reading and use meta-regression models to test predictions derived from existing theories. The results showed that background noise, speech, and music all have a small, but reliably detrimental effect on reading performance. The degree of disruption in reading comprehension did not generally differ between adults and children. Intelligible speech and lyrical music resulted in the biggest distraction. While this last result is consistent with theories of semantic distraction, there was also reliable distraction by noise. It is argued that new theoretical models are needed that can account for distraction by both background speech and noise

    Communicating climate change: conduits, content, and consensus

    Climate change has been the subject of increasing efforts by scientists to understand its causes and implications; it has been of growing interest to policymakers, international bodies, and a variety of nongovernment organizations; and it has attracted varied amounts of attention from traditional and, increasingly, online media. These developments have been aligned with shifts in the nature of climate change communication, with changes in how researchers study it and how a variety of actors try to influence it. This article situates the theory and practice of climate change communication within developments that have taken place since we first reviewed the field in 2009. These include the rise of new social media conduits for communication, research, and practice aimed at fine tuning communication content, and the rise to prominence of scientific consensus as part of that content. We focus in particular on continuing tensions between a focus on the part of communicators to inform the public and more dialogic strategies of public engagement. We also consider the tension between efforts to promote consensus and certainty in climate science and approaches that attempt to engage with uncertainty more fully. We explore the lessons to be learnt from climate communication since 2009, highlighting how the field remains haunted by the deficit model of science communication. Finally, we point to more fruitful future directions for climate change communication, including more participatory models that acknowledge, rather than ignore, residual uncertainties in climate science in order to stimulate debate and deliberation

    Electroporation increases antitumoral efficacy of the bcl-2 antisense G3139 and chemotherapy in a human melanoma xenograft

    <p>Abstract</p> <p>Background</p> <p>Nucleic acids designed to modulate the expression of target proteins remain a promising therapeutic strategy in several diseases, including cancer. However, clinical success is limited by the lack of efficient intracellular delivery. In this study we evaluated whether electroporation could increase the delivery of antisense oligodeoxynucleotides against bcl-2 (G3139) as well as the efficacy of combination chemotherapy in human melanoma xenografts.</p> <p>Methods</p> <p>Melanoma-bearing nude mice were treated i.v. with G3139 and/or cisplatin (DDP) followed by the application of trains of electric pulses to tumors. Western blot, immunohistochemistry and real-time PCR were performed to analyze protein and mRNA expression. The effect of electroporation on muscles was determined by histology, while tumor apoptosis and the proliferation index were analyzed by immunohistochemistry. Antisense oligodeoxynucleotides tumor accumulation was measured by FACS and confocal microscopy.</p> <p>Results</p> <p>The G3139/Electroporation combined therapy produced a significant inhibition of tumor growth (TWI, more than 50%) accompanied by a marked tumor re-growth delay (TRD, about 20 days). The efficacy of this treatment was due to the higher G3139 uptake in tumor cells which led to a marked down-regulation of bcl-2 protein expression. Moreover, the G3139/EP combination treatment resulted in an enhanced apoptotic index and a decreased proliferation rate of tumors. Finally, an increased tumor response was observed after treatment with the triple combination G3139/DDP/EP, showing a TWI of about 75% and TRD of 30 days.</p> <p>Conclusions</p> <p>These results demonstrate that electroporation is an effective strategy to improve the delivery of antisense oligodeoxynucleotides within tumor cells <it>in vivo </it>and it may be instrumental in optimizing the response of melanoma to chemotherapy. The high response rate observed in this study suggest to apply this strategy for the treatment of melanoma patients.</p

    Effects of Anacetrapib in Patients with Atherosclerotic Vascular Disease

    BACKGROUND: Patients with atherosclerotic vascular disease remain at high risk for cardiovascular events despite effective statin-based treatment of low-density lipoprotein (LDL) cholesterol levels. The inhibition of cholesteryl ester transfer protein (CETP) by anacetrapib reduces LDL cholesterol levels and increases high-density lipoprotein (HDL) cholesterol levels. However, trials of other CETP inhibitors have shown neutral or adverse effects on cardiovascular outcomes. METHODS: We conducted a randomized, double-blind, placebo-controlled trial involving 30,449 adults with atherosclerotic vascular disease who were receiving intensive atorvastatin therapy and who had a mean LDL cholesterol level of 61 mg per deciliter (1.58 mmol per liter), a mean non-HDL cholesterol level of 92 mg per deciliter (2.38 mmol per liter), and a mean HDL cholesterol level of 40 mg per deciliter (1.03 mmol per liter). The patients were assigned to receive either 100 mg of anacetrapib once daily (15,225 patients) or matching placebo (15,224 patients). The primary outcome was the first major coronary event, a composite of coronary death, myocardial infarction, or coronary revascularization. RESULTS: During the median follow-up period of 4.1 years, the primary outcome occurred in significantly fewer patients in the anacetrapib group than in the placebo group (1640 of 15,225 patients [10.8%] vs. 1803 of 15,224 patients [11.8%]; rate ratio, 0.91; 95% confidence interval, 0.85 to 0.97; P=0.004). The relative difference in risk was similar across multiple prespecified subgroups. At the trial midpoint, the mean level of HDL cholesterol was higher by 43 mg per deciliter (1.12 mmol per liter) in the anacetrapib group than in the placebo group (a relative difference of 104%), and the mean level of non-HDL cholesterol was lower by 17 mg per deciliter (0.44 mmol per liter), a relative difference of -18%. There were no significant between-group differences in the risk of death, cancer, or other serious adverse events. CONCLUSIONS: Among patients with atherosclerotic vascular disease who were receiving intensive statin therapy, the use of anacetrapib resulted in a lower incidence of major coronary events than the use of placebo.     Ultrazvuk i vibracije sa stanovišta profesionalnog oštećenja

    Changes in wakefulness during exposure to infrasound and whole body vibrations were analysed by means of EEG, pulse rate and blood pressure recordings. Both infrasound at 6 Hz 11.5 dB(lin), and whole body vibrations at 3 Hz 110 dB, in laboratory exposures of 15-20 minutes, were correlated to significant reductions in wakefulness.Opisuju se današnje spoznaje o štetnim učincima ultrazvuka i niskofrekventnih vibracija. Istraživanja su provedena na nekoliko parametara i posebice s obzirom na budnost. U ispitanika koji su bili izvrgnuti ultrazvuku od 6 Hz i 115 dB i vibraciji cijelog tijela od 3 Hz i 110 dB tijekom 15-20 minuta mjeren je ritam pulsa, krvni tlak i sniman je EEG. Utvrđeno je da postoji značajna korelacija između stupnja ekspozicije i smanjenja budnosti. Ova zapažanja upućuju na to da niskofrekventne vibracije i ultrazvuk, što se javljaju na nekim radnim mjestima (kamioni, vlakovi, brodovi, helikopteri i sl.) mogu štetno utjecati na budnost osoba u kojih je visoki stupanj budnosti bitan za rad. U takvim prilikama bilo bi korisno uvoditi promjene u okolini (temperatura, svjetlo, radno vrijeme itd.) čime bi se povećao stupanj budnosti, ukoliko se ne mogu ukloniti izvori vibracija i ultrazvuka
