A Model to Estimate Sediment Yield from Field-Sized Areas: Development of Model
A tool for evaluating sediment yield from field-sized areas is needed for planning management practices to control sediment yield. We developed a reasonably simple simulation model which incorporates fundamental principles of erosion, deposition, and sediment transport mechanics. The model summarizes the state-of-the-art in erosion and sediment yield modeling with appropriate simplifications required to couple the governing equations.
Limited testing showed that the procedures developed here give improved estimates over the Universal Soil Loss Equation. Specific components of the model were tested using experimental data from overland flow, erodible channel, and impoundment studies. These results suggest that the model produces reasonable estimates of erosion, sediment transport, and deposition under a variety of circumstances common to field-scale areas.
Alternative management practices such as conservation tillage, terracing, and contouring can be evaluated separately or in combination to determine their influence on sediment yield. Given a particular location with specified characteristics for climate, soils, topography, and crops, the model provides a means of evaluating alternative management practices to suit a particular farming operation
Adult attachment style across individuals and role-relationships: Avoidance is relationship-specific, but anxiety shows greater generalizability
A generalisability study examined the hypotheses that avoidant attachment, reflecting the representation of others, should be more relationship-specific (vary across relationships more than across individuals), while attachment anxiety, reflecting self-representation, should be more generalisable across a person’s relationships. College students responded to 6-item questionnaire measures of these variables for 5 relationships (mother, father, best same-gender friend, romantic partner or best opposite-gender friend, other close person), on 3 (N = 120) or 2 (N = 77) occasions separated by a few weeks. Results supported the hypotheses, with the person variance component being larger than the relationship-specific component for anxiety, and the opposite happening for avoidance. Anxiety therefore seems not to be as relationship-specific as previous research suggested. Possible reasons for discrepancies between the current and previous studies are discussed
Evolution of a functionally intact but antigenically distinct DENV fusion loop
A hallmark of dengue virus (DENV) pathogenesis is the potential for antibody-dependent enhancement, which is associated with deadly DENV secondary infection, complicates the identification of correlates of protection, and negatively impacts the safety and efficacy of DENV vaccines. Antibody-dependent enhancement is linked to antibodies targeting the fusion loop (FL) motif of the envelope protein, which is completely conserved in mosquito-borne flaviviruses and required for viral entry and fusion. In the current study, we utilized saturation mutagenesis and directed evolution to engineer a functional variant with a mutated FL (D2-FL), which is not neutralized by FL-targeting monoclonal antibodies. The FL mutations were combined with our previously evolved prM cleavage site to create a mature version of D2-FL (D2-FLM), which evades both prM- and FL-Abs but retains sensitivity to other type-specific and quaternary cross-reactive (CR) Abs. CR serum from heterotypic (DENV4)-infected non-human primates (NHP) showed lower neutralization titers against D2-FL and D2-FLM than isogenic wildtype DENV2 while similar neutralization titers were observed in serum from homotypic (DENV2)-infected NHP. We propose D2-FL and D2-FLM as valuable tools to delineate CR Ab subtypes in serum as well as an exciting platform for safer live-attenuated DENV vaccines suitable for naïve individuals and children
Master Sculptor at Work: Enteropathogenic Escherichia coli Infection Uniquely Modifies Mitochondrial Proteolysis during Its Control of Human Cell Death
Enteropathogenic Escherichia coli (EPEC) causes severe diarrheal disease
and is present globally. EPEC virulence requires a bacterial type III secretion system
to inject 20 effector proteins into human intestinal cells. Three effectors travel to
mitochondria and modulate apoptosis; however, the mechanisms by which effectors
control apoptosis from within mitochondria are unknown. To identify and quantify
global changes in mitochondrial proteolysis during infection, we applied the mitochondrial terminal proteomics technique mitochondrial stable isotope labeling by
amino acids in cell culture-terminal amine isotopic labeling of substrates (MS-TAILS).
MS-TAILS identified 1,695 amino N-terminal peptides from 1,060 unique proteins
and 390 N-terminal peptides from 215 mitochondrial proteins at a false discovery
rate of 0.01. Infection modified 230 cellular and 40 mitochondrial proteins, generating 27 cleaved mitochondrial neo-N termini, demonstrating altered proteolytic processing within mitochondria. To distinguish proteolytic events specific to EPEC from
those of canonical apoptosis, we compared mitochondrial changes during infection
with those reported from chemically induced apoptosis. During infection, fewer than
half of all mitochondrial cleavages were previously described for canonical apoptosis, and we identified nine mitochondrial proteolytic sites not previously reported,
including several in proteins with an annotated role in apoptosis, although none occurred at canonical Asp-Glu-Val-Asp (DEVD) sites associated with caspase cleavage.
The identification and quantification of novel neo-N termini evidences the involvement of noncaspase human or EPEC protease(s) resulting from mitochondrialtargeting effectors that modulate cell death upon infection. All proteomics data are

IMPORTANCE To our knowledge, this is the first study of the mitochondrial proteome or N-terminome during bacterial infection. Identified cleavage sites that had
not been previously reported in the mitochondrial N-terminome and that were not
generated in canonical apoptosis revealed a pathogen-specific strategy to control
human cell apoptosis. These data inform new mechanisms of virulence factors targeting mitochondria and apoptosis during infection and highlight how enteropathogenic Escherichia coli (EPEC) manipulates human cell death pathways during infection, including candidate substrates of an EPEC protease within mitochondria. Twenty five years after KLS: A celebration of non-equilibrium statistical mechanics
When Lenz proposed a simple model for phase transitions in magnetism, he
couldn't have imagined that the "Ising model" was to become a jewel in field of
equilibrium statistical mechanics. Its role spans the spectrum, from a good
pedagogical example to a universality class in critical phenomena. A quarter
century ago, Katz, Lebowitz and Spohn found a similar treasure. By introducing
a seemingly trivial modification to the Ising lattice gas, they took it into
the vast realms of non-equilibrium statistical mechanics. An abundant variety
of unexpected behavior emerged and caught many of us by surprise. We present a
brief review of some of the new insights garnered and some of the outstanding
puzzles, as well as speculate on the model's role in the future of

Mediation of detention trauma via perceived locus of control
Political detention in South Africa has been documented to contain conditions inherently detrimental to psychological health. Reports indicate high levels of stress reponses associated with conditions of solitary confinement, and physical and psychological abuse — particularly in the form of Post-Traumatic Stress Disorder. Amongst the moderating variables that may mediate between detention stress and post-detention trauma is perceived locus of control. In the present study the author aimed to determine post-detention sequelae and the moderating influence of perceived locus of control in this specific context. A Post-Traumatic Stress Disorder scale was combined with an Index of Well-being scale and correlated with a Detention Locus of Control scale to assess mediation significance of experienced ‘traumatization’. Results indicated a positive correlation with those who are internal in their perceived locus of control suffering reduced post-stress sequelae, compared to those who are more externally oriented. Implications and limitations of the study are discussed with specific reference to therapeutic intervention in the clinical context. © 1990, South African Psychological Association and the Psychological Institute of the Republic of South Africa. Specific oligomerization of the 5-HT1A receptor in the plasma membrane
In the present study we analyze the oligomerization of the 5-HT1A receptor within living cells at the sub-cellular level. Using a 2-excitation Förster Resonance Energy Transfer (FRET) method combined with spectral microscopy we are able to estimate the efficiency of energy transfer based on donor quenching as well as acceptor sensitization between CFP-and YFP-tagged 5-HT1A receptors at the plasma membrane. Through the analysis of the level of apparent FRET efficiency over the various relative amounts of donor and acceptor, as well as over a range of total surface expressions of the receptor, we verify the specific interaction of these receptors. Furthermore we study the role of acylation in this interaction through measurements of a palmitoylation-deficient 5-HT1A receptor mutant. Palmitoylation increases the tendency of a receptor to localize in lipid rich microdomains of the plasma membrane. This increases the effective surface density of the receptor and provides for a higher level of stochastic interaction
