    Shotgun proteomics of Aspergillus niger microsomes upon D-xylose induction

    Protein secretion plays an eminent role in cell maintenance and adaptation to the extracellular environment of microorganisms. Although protein secretion is an extremely efficient process in filamentous fungi, the mechanisms underlying protein secretion have remained largely uncharacterized in these organisms. In this study, we analyzed the effects of the d-xylose induction of cellulase and hemicellulase enzyme secretion on the protein composition of secretory organelles in Aspergillus niger. We aimed to systematically identify the components involved in the secretion of these enzymes via mass spectrometry of enriched subcellular microsomal fractions. Under each condition, fractions enriched for secretory organelles were processed for tandem mass spectrometry, resulting in the identification of peptides that originate from 1,081 proteins, 254 of which-many of them hypothetical proteins-were predicted to play direct roles in the secretory pathway. d-Xylose induction led to an increase in specific small GTPases known to be associated with polarized growth, exocytosis, and endocytosis. Moreover, the endoplasmic-reticulum-associated degradation (ERAD) components Cdc48 and all 14 of the 20S proteasomal subunits were recruited to the secretory organelles. In conclusion, induction of extracellular enzymes results in specific changes in the secretory subproteome of A. niger, and the most prominent change found in this study was the recruitment of the 20S proteasomal subunits to the secretory organelle

    Anomalous tag diffusion in the asymmetric exclusion model with particles of arbitrary sizes

    Anomalous behavior of correlation functions of tagged particles are studied in generalizations of the one dimensional asymmetric exclusion problem. In these generalized models the range of the hard-core interactions are changed and the restriction of relative ordering of the particles is partially brocken. The models probing these effects are those of biased diffusion of particles having size S=0,1,2,..., or an effective negative "size" S=-1,-2,..., in units of lattice space. Our numerical simulations show that irrespective of the range of the hard-core potential, as long some relative ordering of particles are kept, we find suitable sliding-tag correlation functions whose fluctuations growth with time anomalously slow (t1/3t^{{1/3}}), when compared with the normal diffusive behavior (t1/2t^{{1/2}}). These results indicate that the critical behavior of these stochastic models are in the Kardar-Parisi-Zhang (KPZ) universality class. Moreover a previous Bethe-ansatz calculation of the dynamical critical exponent zz, for size S0S \geq 0 particles is extended to the case S<0S<0 and the KPZ result z=3/2z=3/2 is predicted for all values of SZS \in {Z}.Comment: 4 pages, 3 figure

    Lack of mutations of exon 2 of the MEN1 gene in endocrine and nonendocrine sporadic tumors

    In addition to the mutations that underlie most cases of the multiple endocrine neoplasia type 1 (MEN1) syndrome, somatic mutations of the MEN1 gene have also been described in sporadic tumors like gastrinomas, insulinomas and bronchial carcinoid neoplasm. We examined exon 2 of this gene, where most of the mutations have been described, in 148 endocrine and nonendocrine sporadic tumors. DNA was obtained by phenol/chloroform extraction and ethanol precipitation from 92 formalin-fixed, paraffin-embedded samples, and from 40 fresh tumor tissue samples. We used 5 pairs of primers to encompass the complete coding sequence of exon 2 of the MEN1 gene that was screened by the polymerase chain reaction-single-stranded conformation polymorphism (PCR-SSCP) technique in 78 sporadic thyroid cancers: 28 follicular adenomas, 35 papillary carcinomas, 14 follicular carcinomas, and 1 anaplastic thyroid carcinoma. We also examined 46 adrenal lesions (3 hyperplasias, 3 adenomas and 35 adrenocortical carcinomas, 2 pheochromocytomas, 2 ganglioneuroblastomas, and 1 lymphoma) and 24 breast cancers (6 noninvasive, 16 infiltrating ductal, and 2 invasive lobular tumors). The PCR product of 5 tumors suspected to present band shifts by SSCP was cloned. Direct sense and antisense sequencing did not identify mutations. These results suggest that the MEN1 gene is not important in breast, thyroid or adrenal sporadic tumorigenesis. Because the frequency of mutations varies significantly among tumor subgroups and allelic deletions are frequently observed at 11q13 in thyroid and adrenal cancers, another tumor suppressor gene residing in this region is likely to be involved in the tumorigenesis of these neoplasms.86186

    Multidisciplinary Experience In The Selection Of Patients For Tubal Sterilization.

    Results of the use of a special protocol for evaluation of patients requiring tubal ligation is presented after applied by a multidisciplinary group. The authors conclude that the use of defined parameters of age, parity, marital union duration, number of children alive and the presence of maternal clinical pathology are useful to identify patients with smaller chances of regret after surgery.27% of reproductive-age women in Brazil have chosen surgical sterilization as their contraceptive method. Most of these women who have undergone tubal sterilization opted for cesarean surgery. However, given the young ages of many of these women, many regret having been sterilized. This paper summarizes the experience of a multidisciplinary group in evaluating women who apply for surgical sterilization at the Department of Tocogynecology, Faculdade de Ciencas Medicas, Universidade Estadual de Campinas in Sao Paulo. Detailed descriptions are presented of the medical and social characteristics of cases seen between June 1988 and July 1989. The authors conclude that the use of the defined parameters of age, parity, marital union duration, number of living children, and the presence of maternal clinical pathology are useful in identifying the patients who are least likely to regret undergoing surgical sterilization.113171572

    Gravitational field around a time-like current-carrying screwed cosmic string in scalar-tensor theories

    In this paper we obtain the space-time generated by a time-like current-carrying superconducting screwed cosmic string(TCSCS). This gravitational field is obtained in a modified scalar-tensor theory in the sense that torsion is taken into account. We show that this solution is comptible with a torsion field generated by the scalar field ϕ\phi . The analysis of gravitational effects of a TCSCS shows up that the torsion effects that appear in the physical frame of Jordan-Fierz can be described in a geometric form given by contorsion term plus a symmetric part which contains the scalar gradient. As an important application of this solution, we consider the linear perturbation method developed by Zel'dovich, investigate the accretion of cold dark matter due to the formation of wakes when a TCSCS moves with speed vv and discuss the role played by torsion.     Quintessence and Gravitational Waves

    We investigate some aspects of quintessence models with a non-minimally coupled scalar field and in particular we show that it can behave as a component of matter with 3P/ρ0-3 \lesssim P/\rho \lesssim 0. We study the properties of gravitational waves in this class of models and discuss their energy spectrum and the cosmic microwave background anisotropies they induce. We also show that gravitational waves are damped by the anisotropic stress of the radiation and that their energy spectrum may help to distinguish between inverse power law potential and supergravity motivated potential.     The kk-essence scalar field in the context of Supernova Ia Observations

    A kk-essence scalar field model having (non canonical) Lagrangian of the form L=V(ϕ)F(X)L=-V(\phi)F(X) where X=1/2gμνμϕνϕX=1/2g^{\mu\nu}\nabla_{\mu}\phi\nabla_{\nu}\phi with constant V(ϕ)V(\phi) is shown to be consistent with luminosity distance-redshift data observed for type Ia Supernova. For constant V(ϕ)V(\phi), F(X)F(X) satisfies a scaling relation which is used to set up a differential equation involving the Hubble parameter HH, the scale factor aa and the kk-essence field ϕ\phi. HH and aa are extracted from SNe Ia data and using the differential equation the time dependence of the field ϕ\phi is found to be: ϕ(t)λ0+λ1t+λ2t2\phi(t) \sim \lambda_0 + \lambda_1 t + \lambda_2 t^2. The constants λi\lambda_i have been determined. The time dependence is similar to that of the quintessence scalar field (having canonical kinetic energy) responsible for homogeneous inflation. Furthermore, the scaling relation and the obtained time dependence of the field ϕ\phi is used to determine the XX-dependence of the function F(X)F(X).Comment: 8 pages, 5 figures, Late
