Probing a Complex of Cytochromecand Cardiolipin by Magnetic Circular Dichroism Spectroscopy: Implications for the Initial Events in Apoptosis
Oxidation of cardiolipin (CL) by its complex with cytochrome c (cyt c) plays a crucial role in triggering apoptosis. Through a combination of magnetic circular dichroism spectroscopy and potentiometric titrations, we show that both the ferric and ferrous forms of the heme group of a CL:cyt c complex exist as multiple conformers at a physiologically relevant pH of 7.4. For the ferric state, these conformers are His/Lys- and His/OHβ-ligated. The ferrous state is predominantly high-spin and, most likely, His/β. Interconversion of the ferric and ferrous conformers is described by a single midpoint potential of -80 Β± 9 mV vs SHE. These results suggest that CL oxidation in mitochondria could occur by the reaction of molecular oxygen with the ferrous CL:cyt c complex in addition to the well-described reaction of peroxides with the ferric form
Magnetostimulated Chandges of Microhardness in Potassium Acid Phthalate Crystals
A decrease in microhardness along the (010) cleavage in potassium acid
phthalate single crystals by 15--18% after the application of a permanent
magnetic field was revealed for the first time. It is shown that the effect
revealed is of the volume character. The role of interlayer water in the
processes stimulated by a magnetic field is studied., Interlayer water plays
does not cause the observed changes it only plays the part of an indicator of
these changes in potassium acid phthalate crystals in a magnetic field. It is
established that microhardness in the (100) plane of the crystal in an applied
a magnetic field first increases by 12--15% and then remains constant in time
within the accuracy of the experiment. The possibility of varying the crystal
structure of potassium acid phthalate crystals by applying magnetic fields
inducing rearrangement in the system of hydrogen bonds or in the defect

Complex therapy of urogenital candidosis in women of fertil age in accordance with vaginal microbiocenosis
The results of complex examination (clinic-laboratory, microbiologic and electron-microscopic indices of vaginal microbiocenosis in 52 women with urogenital candidosis of recurrent form) were shown in the present study. The sensitivity of vaginal microflora to intraconazol depending on its compositional kind was also determined. The received data of the investigation indicate the sufficiently high efficacy of intraconazol in the correction of mycrocenosis of vagina. Reducing of treatment duration, increasing of the remission period, fast relief of clinical symptoms of reaction were marked. Combination of high therapeutic efficacy, easy use method of application without side effects, with good tolerance of preparation allow to recommend the using of intraconazol as a choice method in the treatment of urogenital candidosis in women of fertile age

[The possible role of the inner mitochondrial membrane in regulating oxidative phosphorylation in cells in vivo]
International audienceIt has been shown for the first time that the outer mitochondrial membrane has a low permeability for ADP and can control its diffusion into cells in vivo. Respiration of saponin-skinned cardiac and skeletal muscle fibers is maximally stimulated by millimolar concentrations of external ADP. The apparent Km values for ADP are equal to 297 +/- 35 and 334 +/- 54 microM, respectively. After complete extraction of myosin with 0.8 M KCl, which fully preserves the intact structure of the mitochondria, the apparent Km values for exogenously added ADP does not change. However, disruption of the outer mitochondrial membrane by osmotic shock (treatment with 40 mOsM KCl) causes a reduction of the apparent Km value down to 32.3 +/- 5.0 microM of ADP. The apparent Km for ADP in isolated heart mitochondria is 17.6 +/- 1.0 microM. It is concluded that there exists an intracellular factor in the cells in vivo which controls the outer mitochondrial membrane and notably decreases its permeability for ADP. After isolation of mitochondria this factor is lost. When mitochondrial creatine kinase is activated, weak intracellular fluxes of ADP passing through the outer mitochondrial membrane in the skinned fibers are amplified manifold due to the tight functional coupling between mitochondrial creatine kinase and the oxidative phosphorylation system. This coupling is considered to be the central mechanism in the control of cell respiration
