    Michalski MA Portfolio: Finding My Path

    This portfolio is the culmination of my work in the English Teaching specialty Master\u27s program at Bowling Green State University. In addition to the works I produced for my classes and subsequently re-wrote for my portfolio, I\u27ve added an analytical reflection about my growth and evolution during my studies in the English MA program, and about overcoming my difficulties until I finally found the connection between English and historical interests that I sorely needed in order to heighten my enthusiasm and motivation. This reflection brings together the reasons I chose the works in my portfolio to represent my initial discomfort with stepping out of my comfort zone and how the last works helped me turn the corner toward being happy with my sometimes difficult transition. My portfolio includes several works in pedagogy and research with analysis. The pedagogy works include a college unit and lesson plan including literature and writing instruction, and a college syllabus for a literature class. The research and reflective works include a reflective/researched essay titled Why Foucault? that explores the connection between my historical training and background and the English teacher I\u27ve become, and an exploration of literary theory including historical and literary theorists I became familiar with in my literary theory studies. The last work is a research paper that explains scholarly views of teaching Standard American English while still valuing and allowing language diversity within the classroom

    GERMS KNOW NO COLOR: RACIAL SEGREGATION IN BALTIMORE DURING THE INFLUENZA PANDEMIC OF 1918-1919

    The influenza pandemic in 1918-1919 killed more people worldwide in a shorter period than any other known historical epidemic. Wartime conditions and gatherings of large groups of people aided disease transmission. Racial segregation in Baltimore and other cities created overcrowding, and poor living and economic conditions for black citizens. These conditions affected black health and proved conducive to increased viral transmission and subsequent influenza infection. Higher influenza infection rates coupled with lower resistance led to higher mortality rates from secondary complications like pneumonia. In my work, I examine the pandemic flu in Baltimore and especially racial segregation policies that ultimately affected black health and mortality during the event. Segregation caused overcrowding and poor living conditions. It also decreased the ability of blacks to receive medical or nursing care in the civilian and military sectors. Eugenic assumptions about African-American capabilities, in addition to the overwhelming demands of the pandemic emergency, allowed Baltimore’s white community to justify their disregard of the crisis in the African-American community

    The cryo-EM structure of a pannexin 1 reveals unique motifs for ion selection and inhibition

    Pannexins are large-pore forming channels responsible for ATP release under a variety of physiological and pathological conditions. Although predicted to share similar membrane topology with other large-pore forming proteins such as connexins, innexins, and LRRC8, pannexins have minimal sequence similarity to these protein families. Here, we present the cryo-EM structure of a frog pannexin 1 (Panx1) channel at 3.0 A. We find that Panx1 protomers harbor four transmembrane helices similar in arrangement to other large-pore forming proteins but assemble as a heptameric channel with a unique constriction formed by Trp74 in the first extracellular loop. Mutating Trp74 or the nearby Arg75 disrupt ion selectivity whereas altering residues in the hydrophobic groove formed by the two extracellular loops abrogates channel inhibition by carbenoxolone. Our structural and functional study establishes the extracellular loops as important structural motifs for ion selectivity and channel inhibition in Panx1

    Effectiveness of postoperative radiotherapy after radical cystectomy for locally advanced bladder cancer

    BACKGROUND: Local-regional failure (LF) for locally advanced bladder cancer (LABC) after radical cystectomy (RC) is common even with chemotherapy and is associated with high morbidity/mortality. Postoperative radiotherapy (PORT) can reduce LF and may enhance overall survival (OS) but has no defined role. We hypothesized that the addition of PORT would improve OS in LABC in a large nationwide oncology database. METHODS: We identified ≥ pT3pN0-3M0 LABC patients in the National Cancer Database diagnosed 2004-2014 who underwent RC ± PORT. OS was calculated using Kaplan-Meier and Cox proportional hazards regression modeling was used to identify predictors of OS. Propensity matching was performed to match RC patients who received PORT vs those who did not. RESULTS: 15,124 RC patients were identified with 512 (3.3%) receiving PORT. Median OS was 20.0 months (95% CI, 18.2-21.8) for PORT vs 20.8 months (95% CI, 20.3-21.3) for no PORT (P = 0.178). In multivariable analysis, PORT was independently associated with improved OS: hazard ratio 0.87 (95% CI, 0.78-0.97); P = 0.008. A one-to-three propensity match yielded 1,858 patients (24.9% receiving PORT and 75.1% without). In the propensity-matched cohort, median OS was 19.8 months (95% CI, 18.0-21.6) for PORT vs 16.9 months (95% CI, 15.6-18.1) for no PORT (P = 0.030). In the propensity-matched cohort of urothelial carcinoma patients (N = 1,460), PORT was associated with improved OS for pT4, pN+, and positive margins (P \u3c 0.01 all). CONCLUSION: In this observational cohort, PORT was associated with improved OS in LABC. While the data should be interpreted cautiously, these results lend support to the use of PORT in selected patients with LABC, regardless of histology. Prospective trials of PORT are warranted
