    Conformational Studies of Glucose Transporter 1 (GLUT1) as an Anticancer Drug Target

    Glucose transporter 1 (GLUT1) is a facilitative glucose transporter overexpressed in various types of tumors; thus, it has been considered as an important target for cancer therapy. GLUT1 works through conformational switching from an outward-open (OOP) to an inward-open (IOP) conformation passing through an occluded conformation. It is critical to determine which conformation is preferred by bound ligands because the success of structure-based drug design depends on the appropriate starting conformation of the target protein. To find out the most favorable GLUT 1 conformation for ligand binding, we ran systemic molecular docking studies for different conformations of GLUT1 using known GLUT1 inhibitors. Our data revealed that the IOP is the preferred conformation and that residues Phe291, Phe379, Glu380, Trp388, and Trp412 may play critical roles in ligand binding to GLUT1. Our data suggests that conformational differences in these five amino acids in the different conformers of GLUT1 may be used to design ligands that inhibit GLUT1

    Teaching and Learning the Ninth Principle of the ALA Code of Ethics

    In 2021 the American Library Association (ALA) Council approved a ninth principle to be added to the ALA Code of Ethics. The principle reads, “We affirm the inherent dignity and rights of every person. We work to recognize and dismantle systemic and individual biases; to confront inequity and oppression; to enhance diversity and inclusion; and to advance racial and social justice in our libraries, communities, profession, and associations through awareness, advocacy, education, collaboration, services, and allocation of resources and spaces.” This principle is in keeping with the Association for Library and Information Science Education (ALISE) Ethical Guidelines for Library and Information Science Educators: “As LIS educators, we respect and uphold academic freedom and protect the freedom to learn and to teach. We resist censorship and actively promote access to diverse points of view.” In both instances the primary thrust is equity, diversity, inclusion, decolonization, and justice. This panel addresses the ALA Code of Ethics, with special attention to the ninth principle. The panelists will address the core meaning of the principle to determine what it portends for education for library and information professionals. The principle is designed to guide action though praxis. The panelists will examine the degree to which that objective is met by the principle, as well as a set of questions related to the principle: How praxis can address systemic inequity and oppression; how diversity and inclusion manifests itself in praxis; how the advancement of racial and social justice through education can be introduced in the workplace; and how the insertion of these goals can be made into the institutions in which graduates work. The matter of the pressures in which educators work related to racial and social justice work will also be brought up. Throughout the investigation of these matters, the overarching concern of the panelists will be the insertion of the matters into the educational milieu. The panelists will bring to the fore their extensive knowledge and experience in their examination. They will not only provide analyses of the elements of the ninth principle, but will raise questions about the implementation of the principle into the education of professionals. The panel presenters and their talk titles are: (1) John Budd, The Ninth Principle and Global Ethics: The Case for Global Ethics; (2) Suliman Hawamdeh and Michele A. L. Villagran, Multiculturalism and the Role of Information Ethics in Dealing with Academic Integrity Issues; and, (3) Bharat Mehra, “White Pricks” (a.k.a. Inoculations Against Racism) to Decenter Shades of White Privilege in a Professional Association’s Leadership Networks of LIS Educators. Facilitated by John Burgess, delegate for Information Ethics SIG convenor Toni Samek, the panelists will engage the audience in a discussion of the meaning of the ninth principle, but will also have the opportunity to compare it with the ALISE guideline and to offer suggestions regarding the implementation of the tenets of the principle into education (including which kinds of curricular elements are best suited to the insertion of the ALA Code of Ethics into instruction and discussion)

    The prevalence of burnout and depression in medical doctors working in the Cape Town Metropolitan Municipality community healthcare clinics and district hospitals of the Provincial Government of the Western Cape: a cross-sectional study

    Aim: This study investigated burnout and depression in medical doctors in the context of work-related conditions and the role of resilience as a modifiable factor.Method: A cross-sectional, observational study was conducted on consenting medical doctors (n = 132) working at Cape Town Metropolitan Municipality primary healthcare facilities of the Provincial Government of the Western Cape. Data were collected from doctors at 27 facilities by means of a self-administered questionnaire battery, containing socio-demographic information, the Beck Depression Inventory (BDI), the Maslach Burnout Inventory (MBI) and the Connor-Davidson Resilience Scale (CD-RISC).Results: Of 132 doctors included in the analysis, 76% experienced burnout, as indicated by high scores in either the emotional exhaustion or depersonalisation subscales. In addition, 27% of doctors had cut-off scores on the BDI indicating moderate depression, while 3% were identified to have severe depression. The number of hours, work load, working conditions and system-related frustrations were ranked as the most important contributing factors to burnout. More experienced doctors and those with higher resilience scores had lower levels of burnout, as evident by their lower scores in the emotional exhaustion and depersonalisation domains of the MBI.Conclusion: Both burnout and depression are prevalent problems in doctors working at district level and in communities.     Emergency care provision for, and psychological distress in, survivors of domestic violence

    Background This study aimed (i) to ascertain the number of treatment referrals and information about protection orders given to survivors of domestic violence presenting for emergency trauma care, as reported at the one-month visit, (ii) to obtain a profile of violent incidents and injuries, and (iii) to assess self-esteem and posttraumatic and depressive symptomatology in the aftermath of injury. Methods A survey of 62 participants presenting in the acute aftermath of domestic violence (as defined by the Domestic Violence Act of 1998) was conducted over 12 weeks at the Trauma and Resuscitation Unit of a Level One trauma centre in an urban public hospital in South Africa.1 Following informed consent, face-to-face structured interviews were conducted during admission and a month later. The following instruments were administered at baseline: a Demographic and Injury Questionnaire, the Beck Depression and Rosenberg Self-Esteem Inventories, and the Davidson Trauma Scale. A psychosocial questionnaire was administered at the one-month follow-up. Results Fifty-eight per cent of the participants were female and 42% were male. Seventy-four per cent of the perpetrators were male. Ninety-five per cent of the participants said that no health professional had informed them about where or how they could find help. Although all were seriously injured, 76% of the participants said only the researcher had asked about their experience. Sixty-six per cent of the cases of domestic violence were related to intimate partner violence. Overall, subjects displayed high levels of depressive and post-traumatic stress symptomatology that had neither been treated nor adequately referred. Conclusion Even though domestic violence poses significant health threats and costs to the health system, it appears to be a neglected area of South African health care. Health professionals should at least be able to identify and intervene within the "open window" period when psychosocial opportunities are pivotal.     Infections acquired in utero or during the birth process are a significant cause of fetal and neonatal mortality and an important contributor to early and later childhood morbidity. The original concept of the TORCH perinatal infections was to group five infections with similar presentations, including rash and ocular findings (1). This TORCH complex encompasses the infections caused by Toxoplasma gondii, rubella virus, cytomegalovirus, herpes simplex virus both types 1 and 2 and other infective organisms. This review will concentrate on the traditional TORCH infections with Syphilis, Parvovirus B 19, Group B Streptococci infection and Varicella discussed under 'Other Infections(8).     In-beam fast-timing measurements in 103,105,107Cd

    Fast-timing measurements were performed recently in the region of the medium-mass 103,105,107Cd isotopes, produced in fusion evaporation reactions. Emitted gamma-rays were detected by eight HPGe and five LaBr3:Ce detectors working in coincidence. Results on new and re-evaluated half-lives are discussed within a systematic of transition rates. The 7/21+7/2_1^+ states in 103,105,107Cd are interpreted as arising from a single-particle excitation. The half-life analysis of the 11/2111/2_1^- states in 103,105,107Cd shows no change in the single-particle transition strength as a function of the neutron number

    Distinct RNA profiles in subpopulations of extracellular vesicles: apoptotic bodies, microvesicles and exosomes

    Introduction: In recent years, there has been an exponential increase in the number of studies aiming to understand the biology of exosomes, as well as other extracellular vesicles. However, classification of membrane vesicles and the appropriate protocols for their isolation are still under intense discussion and investigation. When isolating vesicles, it is crucial to use systems that are able to separate them, to avoid cross-contamination. Method: EVs released from three different kinds of cell lines: HMC-1, TF-1 and BV-2 were isolated using two centrifugation-based protocols. In protocol 1, apoptotic bodies were collected at 2,000×g, followed by filtering the supernatant through 0.8 µm pores and pelleting of microvesicles at 12,200×g. In protocol 2, apoptotic bodies and microvesicles were collected together at 16,500×g, followed by filtering of the supernatant through 0.2 µm pores and pelleting of exosomes at 120,000×g. Extracellular vesicles were analyzed by transmission electron microscopy, flow cytometry and the RNA profiles were investigated using a Bioanalyzer®. Results: RNA profiles showed that ribosomal RNA was primary detectable in apoptotic bodies and smaller RNAs without prominent ribosomal RNA peaks in exosomes. In contrast, microvesicles contained little or no RNA except for microvesicles collected from TF-1 cell cultures. The different vesicle pellets showed highly different distribution of size, shape and electron density with typical apoptotic body, microvesicle and exosome characteristics when analyzed by transmission electron microscopy. Flow cytometry revealed the presence of CD63 and CD81 in all vesicles investigated, as well as CD9 except in the TF-1-derived vesicles, as these cells do not express CD9. Conclusions: Our results demonstrate that centrifugation-based protocols are simple and fast systems to distinguish subpopulations of extracellular vesicles. Different vesicles show different RNA profiles and morphological characteristics, but they are indistinguishable using CD63-coated beads for flow cytometry analysis
