    Consumer Preferences for Amount and Type of Fat in Ground Beef

    Scientists and beef industry participants are investigating ways to improve the healthiness of beef. We report results of a nationwide mail survey developed to determine consumers’ preferences for fat content in ground beef and identify how consumers would most like to improve the healthiness of beef. The results from a choice-based conjoint experiment indicate that consumers place significant value on reducing saturated fat and the Omega 6:3 ratio in ground beef, but were relatively unconcerned about conjugated linoleic acid. The relatively new method of best-worst scaling was used to further identify which methods consumers most preferred producers use to improve fat content in beef. The results indicate consumers preferred feeding cattle a grass-fed diet as opposed to supplementing cattle feed with fishmeal or flaxseed to improve the fatty acid content in beef.     Identification of an Active Site-bound Nitrile Hydratase Intermediate through Single Turnover Stopped-flow Spectroscopy

    Stopped-flow kinetic data were obtained for the iron-type nitrile hydratase from Rhodococcus equi TG328-2 (ReNHase) using methacrylonitrile as the substrate. Multiple turnover experiments suggest a three-step kinetic model that allows for the reversible binding of substrate, the presence of an intermediate, and the formation of product. Microscopic rate constants determined from these data are in good agreement with steady state data confirming that the stopped-flow method used was appropriate for the reaction. Single turnover stopped-flow experiments were used to identify catalytic intermediates. These data were globally fit confirming a three-step kinetic model. Independent absorption spectra acquired between 0.005 and 0.5 s of the reaction reveal a significant increase in absorbance at 375, 460, and 550 nm along with the hypsochromic shift of an Fe3+←S ligand-to-metal charge transfer band from 700 to 650 nm. The observed UV-visible absorption bands for the Fe3+-nitrile intermediate species are similar to low spin Fe3+-enzyme and model complexes bound by NO or N3−. These data provide spectroscopic evidence for the direct coordination of the nitrile substrate to the nitrile hydratase active site low spin Fe3+ center

    The development of a new sport-specific classification of coping and a meta-analysis of the relationship between different coping strategies and moderators on sporting outcomes

    There is an ever growing coping and sports performance literature, with researchers using many different methods to assess performance and different classifications of coping. As such, it makes it difficult to compare studies and therefore identify how coping is related to performance. Furthermore, there are no quantitative syntheses of the results from these studies. A quantitative synthesis would facilitate a more comprehensive understanding of how coping is associated with athletic performance. In order to accurately compare studies, our first aim was to develop a new coping classification that would make this possible. Firstly, we reviewed the strengths and limitations of the different coping classifications and then identified the commonalities and differences between such classifications. We opted for a three-factor classification of coping, because the evidence suggests that a three-factor classification provides a superior model fit to two-factor approaches. Our new classification of coping was based on an existing model from the developmental literature, which received an excellent model fit. We made some adaptations, however, as our classification was intended for an athletic population. As such, we classified coping as mastery (i.e., controlling the situation and eliminating the stressor), internal regulation (i.e., managing internal stress responses), or goal withdrawal (i.e., ceasing efforts towards goal attainment). Undertaking a meta-analysis, our second aim was to identify which coping strategies correlated with sports performance and whether this relationship varied according to moderator variables. Articles were sourced from online electronic databases and manual journal searches. PRISMA guidelines were used to search, select, and synthesize relevant studies. Random effects meta-analyses were performed to identify associations between coping classification and sport performance. Q, I2, and R2 values assessed heterogeneity. Eighteen published investigations, including 3900 participants and incorporating fifty-nine correlations, indicated an overall positive effect for mastery coping, a negligible negative effect for internal regulation coping, and a negative effect for goal withdrawal strategies. The findings of this meta-analysis could be used by sports practitioners to help them deliver effective coping interventions. In order to maximize performance, practitioners could encourage the use of mastery coping, but advise their athletes not to use goal withdrawal strategies

    Sec6 mutations and the Drosophila exocyst complex

    To allow a detailed analysis of exocyst function in multicellular organisms, we have generated sec6 mutants in Drosophila. We have used these mutations to compare the phenotypes of sec6 and sec5 in the ovary and nervous system, and we find them to be similar. We also find that Sec5 is mislocalized in sec6 mutants. Additionally, we have generated an epitope-tagged Sec8 that localized with Sec5 on oocyte membranes and was mislocalized in sec5 and sec6 germ-line clones. This construct further revealed a genetic interaction of sec8 and sec5. These data, taken together, provide new information about the organization of the exocyst complex and suggest that Sec5, Sec6 and Sec8 act as a complex, each member dependent on the others for proper localization and function

    Is there a link between previous exposure to sport injury psychology education and UK sport injury rehabilitation professionals' attitudes and behaviour towards sport psychology?

    Objectives: The use of sport psychology strategies during sport injury rehabilitation can lead to several positive outcomes such as improved adherence and self-efficacy. The purpose of this study was to compare the sport psychology related attitudes and behaviours of UK sport injury rehabilitation professionals (SIRPs) who had studied the psychological aspects of sport injury to those who had not. Participants and design: Ninety-four SIRPs (54 physiotherapists and 40 sports therapists with a mean of 9.22 years' experience of working in sport) completed an online survey and were grouped according to their level of previous exposure to sport injury psychology education at an undergraduate/postgraduate level. Analyses were undertaken to establish whether there were any differences in sport psychology related attitude (MANOVA), usage (MANOVA), and referral behaviours (chi square) between the groups. Results: The MANOVA and chi square tests conducted revealed that those who had studied the psychological aspects of sport injury reported using significantly more sport psychology in their practice and making more referrals to sport psychologists. Conclusions: It was concluded that sport injury psychology education appears to be effective in increasing the sport psychology related behaviours (use of sport psychology and referral) of SIRPs and should be integrated into professional training

    How are rare species maintained?: Reproductive barriers between Layia jonesii, a rare serpentine endemic, and L. platyglossa

    Reproductive barriers are vital to generating new species as well as maintaining distinct species. Investigating reproductive barriers between closely related plant taxa helps us to understand how these barriers are maintained, particularly between rare and widespread relatives. Layia jonesii, a rare San Luis Obispo County serpentine endemic, and L. platyglossa, a common coastal species, co-occur on serpentine derived hillsides and are interfertile. At these locations, L. jonesii is isolated to dry soils near serpentine rock outcrops and L. platyglossa is located on slightly deeper grassland soils surrounding the rock outcrops. On hillsides where they co-occur, I observe two morphologically distinct species, therefore the two species must be maintaining reproductive barriers, yet mechanisms that maintain this isolation are unknown. I studied this system to investigate possible mechanisms contributing to the maintenance of reproductive barriers. I hypothesize prezygotic reproductive isolation in this system is due to (1) habitat isolation due to local adaptation to differential edaphic environments on the hillside, (2) flowering time differences, and (3) reduced seed set resulting from hybrid crosses. To investigate the local adaptation of L. jonesii and L. platyglossa, I reciprocally transplanted both species into the center of each species’ distribution. I also conducted a competition experiment to determine if L. jonesii is sensitive to resource competition beyond its natural distribution. To investigate flowering time differences, I tracked flowering time of both wild and reciprocally transplanted populations. I also performed controlled crosses to determine if heterospecific, or hybrid crosses, result in lowered seed set than conspecific crosses. The reciprocal transplants showed L. platyglossa is locally adapted to the grassland habitat. Local adaptation likely prevents L. playtyglossa from dispersing into the rock outcrop habitat. Results of the competition experiment revealed L. jonesii is sensitive to competition and this may contribute to its constrained distribution to shallow soils. Local adaptation and competition likely contribute to habitat isolation between the two species. I also documented stark differences in flowering time between the species which contributes to reproductive isolation by reducing pollen flow. Hybrid crosses also resulted in lowered seed set than conspecific crosses. These results suggest prezygotic barriers to reproduction likely maintain the majority of isolation between the two species. These results provide insight into mechanisms that maintain reproductive barriers between closely related taxa existing in similar habitats. The results also contribute to our understanding of how rare plants preserve genetic integrity near common and interfertile relatives
