    Serum urate, menopause, and postmenopausal hormone use: from eminence to evidence-based medicine

    The relationship between serum urate, menopause, and aging has not been clearly defined by scientific evidence. In the present issue of Arthritis Research and Therapy, Hak and Choi present a cross-sectional analysis to clarify the effect of menopause and hormone replacement therapy on serum urate in women within the Third National Health and Nutritional Examination Survey. Menopause increased serum urate and hormone replacement therapy significantly decreased serum urate, although the overall level of change was small. The implications of these urate changes on gout and cardiovascular disease outcomes require further study

    Molecular Dynamics of Film Formation of Metal Tetrasulfonated Phthalocyanine and Poly Amidoamine Dendrimers

    We performed molecular dynamics computer simulations to elucidate the behavior and properties of the metal tetrasulfonated phthalocyanine molecule and the poly(amidoamine) dendrimers in self-assembly depositions, respectively, on poly(allylamine hydrochloride) polymer and on film formed by metal tetrasulfonated phthalocyanine with poly(allylamine hydrochloride). Important physical properties of phthalocyanines were obtained such as the kinetic energy and temperature in situ. By the semiempirical model, we also obtained the UV-Vis absorption spectrum of the film formed by cobalt tetrasulfonated phthalocyanine deposited on poly(allylamine hydrochloride). We performed a study with poly(amidoamine) dendrimers on their deposition time on metal tetrasulfonated phthalocyanine, poly(allylamine hydrochloride) film, and we show the relationship of deposition time with the electrical charge and molecular mass of phthalocyanines. The deposition times of the dendrimers, as a function of their mass, were also elucidated

    Resonant nonlinear absorption in Zn-phthalocyanines

    In this work, we investigate the nonlinear absorption dynamics of Zn phthalocyanine in dimethyl sulfoxide (DMSO). We used single pulse and pulse train Z-scan techniques to determine the dynamics and absorption cross-sections of singlet and triplet states at 532 nm. The excited singlet state absorption cross-section was determined to be 3.2 times higher than the ground state one, giving rise to reverse saturable absorption. We also observed that reverse saturable absorption occurs from the triplet state, after its population by intersystem crossing, whose characteristic time was determined to be 8.9 ns. The triplet state absorption cross-section determined is 2.6 times higher than the ground state one. In addition, we used the white light continuum Z-scan to evaluate the singlet excited state spectrum from 450 to 710 nm. The results show two well-defined regions, one above 600 nm, where reverse saturable absorption is predominant. Below 600 nm, we detected a strong saturable absorption. A three-energy-level diagram was used to explain the experimental results, leading to the excited state absorption cross-section determination from 450 nm up to 710 nm.FAPESPCNPQAFOSR (FA9550- 07-1-0374

    Expanding the repertoire of miRNAs and miRNA-offset RNAs expressed in multiple myeloma by small RNA deep sequencing

    Microarray analysis of the multiple myeloma (MM) miRNome has unraveled the differential expression of miRNAs in cytogenetic subgroups, their involvement in the tumor biology and their effectiveness in prognostic models. Herein, the small RNA transcriptional landscape in MM has been investigated exploiting the possibilities offered by small RNA-seq, including accurate quantification of known mature species, discovery and characterization of isomiRs, and miRNA-offset RNAs (moRNAs). Matched small RNA-seq and miRNA GeneChip \uae microarray expression profiles were obtained in a representative panel of 30 primary MM tumors, fully characterized for genomic aberrations and mutations. RNA-seq and microarray gave concordant estimations of known species. Enhanced analysis of RNA-seq data with the miR&moRe pipeline led to the characterization of 655 known and 17 new mature miRNAs and of 74 moRNAs expressed in the considered cohort, 5 of which (moR-150-3p, moR-24-2-5p, moR-421-5p, moR-21-5p, and moR-6724-5p) at high level. Ectopic expression of miR-135a-3p in t(4;14) patients, upregulation of moR-150-3p and moR-21-5p in t(14;16)/t(14;20) samples, and of moR-6724-1-5p in patients overexpressing CCND1 were uncovered and validated by qRT-PCR. Overall, RNA-seq offered a more complete overview of small non-coding RNA in MM tumors, indicating specific moRNAs that demand further investigations to explore their role in MM biology

    Analytical method is applied for description of the small angle Bhabha scattering at LEP1. Inclusive event selection for asymmetrical wide-narrow circular detectors is considered. The QED correction to the Born cross-section is calculated with leading and next-to-leading accuracy in the second order of perturbation theory and with leading one --- in the third order. All contributions in the second order due to photonic radiative corrections and pair production are calculated starting from essential Feynman diagrams. The third order correction is computed by means of electron structure function. Numerical results illustrate the analytical calculsations.Comment: 33 pages, LaTeX, submitted to ZhET

    A definition of flare in low back pain (LBP): A multiphase process involving perspectives of individuals with LBP and expert consensus

    Low back pain (LBP) varies over time. Consumers, clinicians and researchers use various terms to describe fluctuations of LBP symptoms. Although "flare" is commonly used to describe symptom fluctuation, there is no consensus on how it is defined. This study aimed to obtain consensus for a LBP flare definition using a mixed-method approach. Step 1 involved derivation of a preliminary candidate flare definition based on thematic analysis of consumers' views in consultation with an expert consumer writer. In Step 2, a workshop was conducted to incorporate perspectives of LBP experts into the preliminary flare definition, which resulted in two alternative LBP flare definitions. Step 3 refined the definition using a two-round Delphi consensus process with experts in musculoskeletal conditions. The definition favoured by experts was further tested with individuals with LBP in Step 4, using the definition in three scenarios. This multiphase study produced a LBP flare definition that distinguishes it from other LBP fluctuations, represents views of consumers, involves expert consensus, and is understandable by consumers in clinical and research contexts: "A flare-up is a worsening of your condition that lasts from hours to weeks that is difficult to tolerate and generally impacts your usual activities and/or emotions".     Association of Functional Polymorphism rs2231142 (Q141K) in the ABCG2 Gene With Serum Uric Acid and Gout in 4 US Populations

    A loss-of-function mutation (Q141K, rs2231142) in the ATP-binding cassette, subfamily G, member 2 gene (ABCG2) has been shown to be associated with serum uric acid levels and gout in Asians, Europeans, and European and African Americans; however, less is known about these associations in other populations. Rs2231142 was genotyped in 22,734 European Americans, 9,720 African Americans, 3,849 Mexican Americans, and 3,550 American Indians in the Population Architecture using Genomics and Epidemiology (PAGE) Study (2008–2012). Rs2231142 was significantly associated with serum uric acid levels (P = 2.37 × 10−67, P = 3.98 × 10−5, P = 6.97 × 10−9, and P = 5.33 × 10−4 in European Americans, African Americans, Mexican Americans, and American Indians, respectively) and gout (P = 2.83 × 10−10, P = 0.01, and P = 0.01 in European Americans, African Americans, and Mexican Americans, respectively). Overall, the T allele was associated with a 0.24-mg/dL increase in serum uric acid level (P = 1.37 × 10−80) and a 1.75-fold increase in the odds of gout (P = 1.09 × 10−12). The association between rs2231142 and serum uric acid was significantly stronger in men, postmenopausal women, and hormone therapy users compared with their counterparts. The association with gout was also significantly stronger in men than in women. These results highlight a possible role of sex hormones in the regulation of ABCG2 urate transporter and its potential implications for the prevention, diagnosis, and treatment of hyperuricemia and gout
