    Building Sustainable Success in Art Galleries: An Exploratory Study of Adaptive Strategies

    Historically, art galleries have operated in a cottage industry that placed a premium on building unique relations with artists and clients. Recent economic and technological trends threaten the viability of this model. Art galleries now face a very challenging environment, demanding careful attention to strategy formulation and execution. However, more questions than answers can be found in the literature about management practices, art gallery performance, and factors affecting gallery success. This paper reports findings on these issues from an exploratory survey of Colorado art galleries. Results suggest that art gallery owners and mangers are not overly concerned about external pressures influencing their organizations. This attitude could adversely affect efforts to build sustainable success and value through adaptive strategies

    Environmental Application of Cyclodextrin Metal-Organic Frameworks in an Undergraduate Teaching Laboratory

    Reported herein is a multidisciplinary laboratory experiment for advanced undergraduate students that includes elements of material synthesis, in the synthesis of cyclodextrin-containing metalā€“organic frameworks (CD-MOFs), and environmental chemistry, in the use of these MOFs for pollutant removal. This multiday laboratory experiment starts with the synthesis of cyclodextrin-containing metalā€“organic frameworks (CD-MOFs) using vapor diffusion crystal growth procedures, followed by the use of the CD-MOFs for a pollutant removal application. Specifically, the CD-MOFs were used for the removal of a methylene blue dye (a common mimic of aromatic pollutants) from an organic solution, with the monitoring of the success of the removal procedures using UVā€“vis spectroscopy. This experiment was implemented as part of a larger multiday unit, and undergraduate students were particularly engaged with and excited by the CD-MOF synthesis and methylene blue removal experimental modules. As a result, the decision was made to make these two components a stand-alone multidisciplinary laboratory experiment, the results of which are reported herein

    A strategic alliance (SA) is a mutually beneficial long-term formal relationship formed between two or more parties to pursue a set of agreed upon goals or to meet a critical business need while remaining independent organizations.  It is a synergistic arrangement whereby two or more organizations agree to cooperate in the carrying out of a business activity where each brings different strengths and capabilities to the arrangement.  The social structure of alliances has been considered previously (Gulati 1995, et al.), so instead of discussing the social structure relative to alliance partners, this paper looks at the relationship between the dyad alliance entity and its customer(s).  This newer aspect is particularly important when there are differences in trust and culture to consider (Das & Teng 1998) between alliance partners.  Other considerations include authority, governance and structure, conflict, and the make-up of the strategic alliance, its partners, and the customer(s).&nbsp

    Investigation of the Luco dei Marsi DSGSD revealing the first evidence of a basal shear zone in the central Apennine belt (Italy)

    Deep-seated gravitational slope deformations (DSGSDs) show a wide range of geomorphological characteristics and kinematic behaviours. In many cases, deforming rock masses move on a continuous surface or a thick basal shear zone (BSZ) overlying the stable bedrock. The nature of this boundary is a significant issue in scientific debates since examples of BSZs have been observed or inferred in some DSGSDs worldwide. In the central Apennines, although several cases of DSGSDs have been described in recent decades, no evidence of BSZs has been documented thus far. This work presents the first case of a BSZ found in the region at the bottom of a large-scale gravitational deformation that affects the Mesozoic-Cenozoic carbonate ridge overhanging the Luco dei Marsi village (Abruzzi region). The BSZ consists of several metres-thick, cataclastic breccia developed within middle-Upper Cretaceous biodetritic limestone. The breccia level is exposed for approximately 200 m with a subhorizontal geometry and shows severe rock damage and weathering. The DSGSD hosting the BSZ affects an NNW-SSE-oriented and wide Miocene anticline whose eastern limb is dismembered by Pliocene-Quaternary normal faults delimiting the edge of a large Quaternary intermontane basin (the Fucino Basin). Field survey, aerial photointerpretation, and remote sensing (DInSAR technique) analyses outline an active gravity-driven process. This is characterized by several kinds of geomorphological features, including downhill- and uphill-facing scarps, ridge-top depressions, gravitational grabens and trenches in the upper and middle parts of the ridge, and bulging at the toe of the slope. These features, which can be distinguished from tectonic elements due to their shape and extension, are an indication of a high degree of internal deformation and a compound sagging geometry for the Luco dei Marsi DSGSD. The short-term activity of the process was revealed by DInSAR time series covering almost thirty years of satellite datasets, including ERS1/2, ENVISAT, COSMO-SkyMed, and SENTINEL 1 constellations. Strain rates on the order of a few mm/yr were inferred, with a marked difference between different sectors of the DSGSD area. The long-term (y > 102) lifespan of the DSGSD was framed into a multiple-step conceptual model summarizing the Early Pleistocene-Holocene geological evolution of the area. The model results outline the control exercised by extensional tectonics on DSGSD development, as progressive displacements along normal faults in the latest Pleistocene were the cause of lateral unconfinement at the toe of the slope. This work further contributes to the increasing knowledge on DSGSDs in the central Apennines and the understanding of the relationship between deformation features induced by slope morphogenesis, such as the BSZ, and Quaternary tectonics within the mountain belt

    Digging the New York City Skyline: Soil Fungal Communities in Green Roofs and City Parks

    In urban environments, green roofs provide a number of benefits, including decreased urban heat island effects and reduced energy costs for buildings. However, little research has been done on the non-plant biota associated with green roofs, which likely affect their functionality. For the current study, we evaluated whether or not green roofs planted with two native plant communities in New York City functioned as habitats for soil fungal communities, and compared fungal communities in green roof growing media to soil microbial composition in five city parks, including Central Park and the High Line. Ten replicate roofs were sampled one year after planting; three of these roofs were more intensively sampled and compared to nearby city parks. Using Illumina sequencing of the fungal ITS region we found that green roofs supported a diverse fungal community, with numerous taxa belonging to fungal groups capable of surviving in disturbed and polluted habitats. Across roofs, there was significant biogeographical clustering of fungal communities, indicating that community assembly of roof microbes across the greater New York City area is locally variable. Green roof fungal communities were compositionally distinct from city parks and only 54% of the green roof taxa were also found in the park soils. Phospholipid fatty acid analysis revealed that park soils had greater microbial biomass and higher bacterial to fungal ratios than green roof substrates. City park soils were also more enriched with heavy metals, had lower pH, and lower quantities of total bases (Ca, K, and Mg) compared to green roof substrates. While fungal communities were compositionally distinct across green roofs, they did not differentiate by plant community. Together, these results suggest that fungi living in the growing medium of green roofs may be an underestimated component of these biotic systems functioning to support some of the valued ecological services of green roofs

    The Opdc missense mutation of Pax2 has a milder than loss-of-function phenotype

    Renal-coloboma syndrome, also known as papillorenal syndrome, is an autosomal dominant human disorder in which optic disc coloboma is associated with kidney abnormalities. Mutations in the paired domain transcription factor PAX2 have been found to be the underlying cause of this disease. Disease severity varies between patients, and in some cases, renal hypoplasia has been found in the absence of any retinal defects. Here we report an N-ethyl-N-nitrosourea-induced mouse mutation, Opdc, which is an isoleucinetothreonine missense mutation, I40T, in the first Ī±-helix of the Pax2 paired domain. The mutant protein binds target DNA sequences less strongly than the wild-type protein and acts poorly to transactivate target promoters in culture. The phenotypic consequence of this mutation on the development of the eye and ear is similar to that reported for null alleles of Pax2. However, in homozygotes, cerebellar development is normal on a genetic background in which loss of Pax2 results in failure of cerebellar formation. Moreover, there is a genetic background effect on the heterozygous phenotype such that on some strain backgrounds, kidney development is unaffected. Opdc is the first hypomorphic mutation reported for Pax2 that differs in phenotype from loss-of-function mutations. These results suggest that PAX2 is a strong candidate gene for cases in which human patients have optic disc coloboma not associated with renal dysplasia

    WT1 gene expression as a prognostic marker in advanced serous epithelial ovarian carcinoma: an immunohistochemical study

    BACKGROUND: WT1 is a tumor suppressor gene responsible for Wilms' tumor. WT1 reactivity is limited to ovarian serous carcinomas. Recent studies have shown that WT1 plays an important role in the progression of disease and indicates a poorer prognosis of human malignancies such as acute myeloid leukemia and breast cancer. The aims of this study were to determine the survival and recurrence-free survival of women with advanced serous epithelial ovarian carcinoma in relation to WT1 gene expression. METHODS: The study accrued women over an 18-year period, from 1987ā€“2004. During the study period, 163 patients were diagnosed with advanced serous epithelial ovarian carcinoma and had undergone complete post-operative chemotherapy, but the final study group comprised 99 patients. The records of these women were reviewed and the paraffin-embedded tissue of these women stained with WT1 immunostaining. Survival analysis was performed using Kaplan-Meier and Cox regression methods. RESULTS: Fifty patients showed WT1 staining and forty-nine did not. Five-year survival of non-staining and staining groups were 39.4% and 10.7% (p < 0.00005); five-year recurrence-free survival of these groups were 29.8% and ā‰¤ 7.5% (p < 0.00005), respectively. For survival the HR of WT1 staining, adjusted for residual tumor and chemotherapy response, was 1.98 (95% CI 1.28ā€“3.79), and for recurrence-free survival the HR was 3.36 (95% CI 1.60ā€“7.03). The HR for recurrence-free survival was not confounded by any other variables. CONCLUSION: This study suggests that expression of WT1 gene may be indicative of an unfavorable prognosis in patients with advanced serous epithelial ovarian carcinoma

    PPARĪ² activation inhibits melanoma cell proliferation involving repression of the Wilmsā€™ tumour suppressor WT1

    Peroxisome proliferator-activated receptors (PPARs) are ligand-activated transcription factors that strongly influence molecular signalling in normal and cancer cells. Although increasing evidence suggests a role of PPARs in skin carcinogenesis, only expression of PPARĪ³ has been investigated in human melanoma tissues. Activation of PPARĪ± has been shown to inhibit the metastatic potential, whereas stimulation of PPARĪ³ decreased melanoma cell proliferation. We show here that the third member of the PPAR family, PPARĪ²/Ī“ is expressed in human melanoma samples. Specific pharmacological activation of PPARĪ² using GW0742 or GW501516 in low concentrations inhibits proliferation of human and murine melanoma cells. Inhibition of proliferation is accompanied by decreased expression of the Wilmsā€™ tumour suppressor 1 (WT1), which is implicated in melanoma proliferation. We demonstrate that PPARĪ² directly represses WT1 as (1) PPARĪ² activation represses WT1 promoter activity; (2) in chromatin immunoprecipitation and electrophoretic mobility shift assays, we identified a binding element for PPARĪ² in the WT1 promoter; (3) deletion of this binding element abolishes repression by PPARĪ² and (4) the WT1 downstream molecules nestin and zyxin are down-regulated upon PPARĪ² activation. Our findings elucidate a novel mechanism of signalling by ligands of PPARĪ², which leads to suppression of melanoma cell growth through direct repression of WT1
