    Naltrexone Reverses Ethanol-Induced Rat Hippocampal and Serum Oxidative Damage

    Naltrexone, an antagonist of �-opioid receptors, is clinically used as adjuvant therapy of alcohol dishabituation. The aim of the present work was to test the effect of 1 mg/kg body weight of naltrexone to revert oxidative stress-related biochemical alterations, in the hippocampus and serum of chronic alcoholic adult rats. Malondialdehyde concentration was increased and glutathione peroxidase activity was decreased in hippocampus and serum of alcohol-treated rats. Naltrexone treatment restored these alterations. The in vitro antioxidant ability of Ntx could not justify these effects considering the doses used.     Propuesta de WebQuest como herramienta didáctica para la adiquisición de competencias

    [SPA]Los nuevos estudios de Grado y Posgrado presentan unas competencias que debe adquirir el alumno, con el resultado de un aprendizaje tras el alcance de unos objetivos. Necesitamos nuevas herramientas para la valoración y evaluación de estas competencias, así como nuevas alternativas metodológicas que nos lo facilite. Los docentes tenemos para ello en Internet, un potencial extraordinario, si sabemos utilizarlo adecuadamente. Presentamos, como propuesta, la utilización de la WebQues, como recurso educativo digital en el proceso de enseñanza-aprendizaje. Objetivo: Proporcionar estrategias al alumno para la adquisición de competencias, así como facilitar al docente la evaluación del resultado del aprendizaje.. Metodología: Revisión bibliográfica sobre diseño y aplicabilidad de la WebQuest, Ley de Convergencia de Educación Superior y EEES. Resultados. La WQ es una actividad orientada hacia la investigación con información procedente de la red. Consta de: Introducción, Tarea, Proceso, Evaluación y Conclusión, con sus correspondientes ventajas para el alumno y docente, relacionado con la adquisición de competencias de una materia. Conclusiones: Permite al alumno crear, analizar , transformar y valorar información de la red, recomendada por el docente, y participar en el diseño y presentación de resultados. Constituye un elemento importante para la evaluación del alumno. [ENG]The new Grade and Master studies show some abilities that must be obtained by the student, with the result of a learning after achieving the aims. We need new tools, and new methodological alternatives, to assess these abilities. Professors have in Internet an extraordinary potential if we know how to use it properly. We show in our proposal, the use of WebQuest like a digital education resource in the learning-teaching process. Objetives: Giving to the student different ways to develop the abilities, and make it easier to the teacher how to evaluate the result of the learning process. Methodology: bibliographic revision about the Convergence of Higher Education Law and the European Higher Education Area, and design and use of the WebQuest. Results: WQ is an activity directed to the research with information from the net. It has: Introduction, tasks, Process, Assess and Conclusion, with its advantages for the student and teacher, related to the process of development abilities. Conclusions: It allows the student to create, analize, transform and evaluate recommended information from the net, and participate in the design and presentation of results.     Differential hippocampal response to chronic alcohol consumption of young adult and mature adult rats

    AIMS Early ethanol consumption could be a risk factor for young brain integrity and its maturation, and also for the development of addictive behaviors in adulthood. Neuronal nitric oxide synthase (nNOS) expressing neurons are specifically located in the subgranular layer (SGL) of dentate gyrus and may be relevant for hippocampal neurogenesis. The focus of this work is aimed to determine local changes in the nNOS-like immunoreactive (nNOS-LIR) cell populations of the SGL after chronic ethanol exposure in young adult and mature adult rats. METHODS We used the nicotinamide adenine dinucleotide phosphate (NADPH)-diaphorase (NADPH-d) reaction as a qualitative marker of nNOS enzyme activity. We also analyzed the nNOS-LIR cell density by the nNOS immunocytochemistry in order to compare these two methods of labeling. Dorsal striatum (CPu) was also analyzed in order to compare two neural areas with high nNOS-LIR cell density. RESULTS The young adult group showed less hippocampal NADPH-d(+) cell density than the mature adult group. Interestingly, the NADPH-d(+) cell density was increased in the SGL of the young adult ethanol-treated group, whereas it decreased in the mature adult ethanol-treated group, when compared with their respective controls. No change was observed in any of the groups for the hippocampal nNOS-LIR cell density and no differences could be established in CPu for nNOS-LIR and NADPH-d(+) cell densities in any of the groups studied. CONCLUSION The NADPH-d expression is affected by chronic ethanol exposure in opposite ways between both age groups studied. Further studies are needed to evaluate the relative importance of these findings, especially when considering human subject

    Role of hippocampal NF-κB and GluN2B in the memory acquisition impairment of experiences gathered prior to cocaine administration in rats

    Cocaine can induce severe neurobehavioral changes, among others, the ones involved in learning and memory processes. It is known that during drug consumption, cocaine-associated memory and learning processes take place. However, much less is known about the effects of this drug upon the mechanisms involved in forgetting.The present report focuses on the mechanisms by which cocaine affects memory consolidation of experiences acquired prior to drug administration. We also study the involvement of hippocampus in these processes, with special interest on the role of Nuclear factor kappa B (NF-κB), N-methyl-D-aspartate glutamate receptor 2B (GluN2B), and their relationship with other proteins, such as cyclic AMP response element binding protein (CREB). For this purpose, we developed a rat experimental model of chronic cocaine administration in which spatial memory and the expression or activity of several proteins in the hippocampus were assessed after 36 days of drug administration. We report an impairment in memory acquisition of experiences gathered prior to cocaine administration, associated to an increase in GluN2B expression in the hippocampus. We also demonstrate a decrease in NF-κB activity, as well as in the expression of the active form of CREB, confirming the role of these transcription factors in the cocaine-induced memory impairment

    This study presents a new zenith looking narrow-band radiometer based system (ZEN), conceived for dust aerosol optical depth (AOD) monitoring.     Light conditions and heterogeneity in illumination affect growth and survival of Octopus vulgaris paralarvae reared in the hatchery

    In order to succeed in common octopus (Octopus vulgaris) aquaculture, it is necessary to overcome the massive paralarval mortalities, which are likely related to zootechnical and/or nutritional aspects. Among the zootechnical aspects, the effect of light could be vital for the development of paralarvae given their sophisticated visual system. O. vulgaris paralarvae display vertical behavior in the wild, being in deeper waters during the day and close to the surface at night. For this reason, the present study traces a collection of light assays in captivity, to test ecologically-driven hypotheses on the growth and survival of octopus paralarvae. The first assay tested a factorial combination of light colors (blue vs white) and intensity (low ~13 lx (36 W/m2), medium ~87 lx (151 W/m2) and high ~265 lx (422 W/m2)). The results pointed to a significant negative impact of low light intensities (13 and 87 lx) on paralarval growth. The second assay was designed to contrast light colors (blue vs white), different levels of intensities (300 lx (478 W/m2) vs 600 lx (1077 W/m2)) and partial coverage of the tanks using a shade mesh, named as “shadow zone” (half-covered vs non-covered tanks). In half-covered tanks, survival was significantly improved, with the best results coming from half-covered tanks with blue light and 600 lx, but no differences in dry weight were observed. The third experiment tested a factor called “light source position” with two levels: “Control” when the light incidence angle was 0° with respect to the normal direction (i.e. perpendicular to the water surface), and “Oblique” when the light incidence angle was 60° with respect to the normal direction. In this experiment, survival significantly improved under oblique light but no differences in dry weight were detected. The last experiment was an unifactorial design combining “light source position” and “tank partial coverage” with three treatments: i) control light with uncovered tanks, ii) oblique light with uncovered tanks, iii) control light with semi-covered tanks. Oblique light with uncovered tanks was associated with a higher survival rate but without statistical significance, probably due to data variability. It can be concluded that light intensity tended to affect paralarval dry weight, whereas the existence of a shadow zone or oblique light are more related with an improvement of survival rate

    Cocaine promotes oxidative stress and microglial-macrophage activation in rat cerebellum

    Different mechanisms have been suggested for cocaine neurotoxicity, including oxidative stress alterations. Nuclear factor kappa B (NF-κB), considered a sensor of oxidative stress and inflammation, is involved in drug toxicity and addiction. NF-κB is a key mediator for immune responses that induces microglial/macrophage activation under inflammatory processes and neuronal injury/degeneration. Although cerebellum is commonly associated to motor control, muscular tone, and balance. Its relation with addiction is getting relevance, being associated to compulsive and perseverative behaviors. Some reports indicate that cerebellar microglial activation induced by cannabis or ethanol, promote cerebellar alterations and these alterations could be associated to addictive-related behaviors. After considering the effects of some drugs on cerebellum, the aim of the present work analyzes pro-inflammatory changes after cocaine exposure. Rats received daily 15 mg/kg cocaine i.p., for 18 days. Reduced and oxidized forms of glutathione (GSH) and oxidized glutathione (GSSG), glutathione peroxidase (GPx) activity and glutamate were determined in cerebellar homogenates. NF-κB activity, CD68, and GFAP expression were determined. Cerebellar GPx activity and GSH/GSSG ratio are significantly decreased after cocaine exposure. A significant increase of glutamate concentration is also observed. Interestingly, increased NF-κB activity is also accompanied by an increased expression of the lysosomal mononuclear phagocytic marker ED1 without GFAP alterations. Current trends in addiction biology are focusing on the role of cerebellum on addictive behaviors. Cocaine-induced cerebellar changes described herein fit with previosus data showing cerebellar alterations on addict subjects and support the proposed role of cerebelum in addiction

    Advanced trajectory generator for two carts with RGB-D sensor on circular rail

    This paper presents a motorised circular rail that generates the motion of two carts with an RGB-D sensor each. The objective of both carts' trajectory generation is to track a person's physical rehabilitation exercises from two points of view and his/her emotional state from one of these viewpoints. The person is moving freely his/her position and posture within the circle drawn by the motorised rail. More specifically, this paper describes the calculation of trajectories for safe motion of the two carts on the motorised circular rail in detail.
