Valós idejű spektroszkópiai mérések és kemometriai módszerek alkalmazása gyógyszerkioldódás predikciójára: Development of real-time spectroscopic measurements and chemometric methods for predicting dissolution profiles of pharmaceutical tablets
More than a hundred million pharmaceutical tablets are produced annually, and pharmaceutical companies mainly use statistical methods to ensure the quality of them. This means that a small number of samples are taken from the batch, which are then analyzed by destructive methods, and the results let us draw conclusions of the whole batch. There is a huge necessity for reliable, fast, and non-destructive testing methods, which would allow us to analyze all the produced tablets and make it possible to eliminate the chance of letting faulty tablets on the market. NIR- and Raman-spectroscopy coupled with chemometric methods such as PLS regression, response surfaces and artificial neural networks (ANN) can be an appropriate tool for predicting drug release based on the tablets’ spectra. My goal was checking the robustness of previously designed models, building new expanded models, and comparing them to provide more precise results in a wider range of factors.

A világon évente több százmilliárd gyógyszertablettát gyártanak, melyek minőségének biztosítására a gyógyszergyártó cégek többsége statisztikai eszközöket alkalmaz. Ennek értelmében egy legyártott sarzsból kis számú mintát vesznek, amelyeket különböző roncsolásos analitikai módszerekkel vizsgálnak, majd ez alapján vonnak le következtetéseket az egész sarzsról. A gyártott tabletták minőségbiztosításának szempontjából nagy szükség lenne egy olyan megbízható, gyors és minden tablettát roncsolásmentesen átvizsgáló módszerre, amellyel elkerülhető lenne a nem megfelelő minőségű termékek piacra kerülése. Erre ad lehetőséget a Raman- és NIR-spektroszkópia, különböző kemometriai módszerekel kapcsolva, mint a PLS-regresszió, illesztett válaszfelületek és a mesterséges neurális hálók. Ezek segítségével lehetőség nyílik egy adott tabletta hatóanyagleadásának becslésére a felvett spektrumok alapján. Kutatásom során a célom már előzőleg felállított, az adott kísérlettérben bizonyítottan jól működő modellek robusztusságának ellenőrzése, kibővítése és összehasonlítása volt, hogy a változó paraméterek szélesebb tartományában biztosítsanak megbízható eredményt
Diffúziós anyagtranszport folyamatok és felületi jelenségek vizsgálata pásztázó elektrokémiai mikroszkópiás módszerrel = Investigation of diffusion processes and surface reactions using scanning electrochemical microscopy
Diffúziós folyamatokat és felületi jelenségeket vizsgáltunk elektrokémiai módszerekkel különböző közegekben. Alkalmaztuk az általuk kidolgozott, az 'elektrokémiai repülési idő' nyomonkövetésén alapuló diffúzió állandó mérés egy újszerű változatát. A módszer a pásztázó elektrokémiai mikroszkóp speciális képességén alapul. Segítségével egyszerű eljárást követve viszonylag pontos adat nyerhető anyagok különböző közegekben mutatkozó diffúziós koefficienséről. Vizes oldatokban, gélekben, szuszpenziókból képződött üledékekben, ion folyadékokban mértük különböző anyagok diffúziós koefficiensét. A kapott eredményeket összehasonlítottuk más elektrokémiai módszer alkalmazásával kapott adatokkal. Az új módszer előnyös voltát elektrokémiailag komplikált reakciót modellként választva igazoltuk. A munka során új elektrokémiai mikroszkópot építettünk, új mérőcellát, injektor készüléket állítottunk össze. A voltammertiás koncentrációmérés olyan esetekben ad megbízható eredményt, amikor a kalibráló oldatokban és az analizálandó közegben azonos a mérendő komponens diffúzió állandója. Ez a feltétel sok esetben, így például in vivo mérések során nehezen teljesíthető. Az ilyen esetben végzett koncentráció mérés megbízhatóságának növelését lehetővé tévő megoldások vizsgálatára kísérleteket végeztünk. Kidolgoztuk a diffúzió réteg feltöltődését lehetővé tevő periodikusan megszakított amperometriás módszert, ezzel egyes enzim elektródos mérések érzékenységét jelentősen sikerült megnövelnünk. | Diffusion transport processes and concentration profiles resulted by surface reactions were investigated using electrochemical methods. A new variety of the electrochemical time of flight method developed in our work has been used. It takes advantage on the special capability of the scanning electrochemical microscopy (SECM). This technically simple procedure can result in relatively accurate data about diffusion character of different species in different media. Measurements were done with different species in different media like aqueous solutions, gels, ionic liquids and sediments obtained from suspensions. The data obtained were compared with data measured with other electrochemical methods. The advantage of the new method has been verified selecting electrochemically complicated model reaction. In the work a new SECM, a new injector have been constructed, and new cell and electrodes were made and tested. The voltammetric concentration measurements are accurate only when the diffusion coefficient of the measured species is identical in the calibrating solutions and in samples. This is not always true. The diffusion coefficient in living tissues can considerable differ from that of in aqueous calibrating standards. In our work efforts were done for increasing the accuracy of voltammetric concentration measurements. A measuring method, the periodically interrupted amperometry (PIA) has been worked out. It allowing time for reloading the diffusion layer could increase dramatically the sensitivity of certain enzyme electrode measurements

Scanning electrochemical microscopy (SECM) in the sample generation-tip collection
mode was employed to gain new insights on the effect of aluminum on the anomalous
hydrogen evolution from anodically-polarized magnesium exposed to an aqueous saline
solution. Various experiments were designed to investigate the processes occurring on a
model Mg-Al galvanic pair in regards to the hydrogen evolution reaction occurring separately
at the anodic and cathodic sites. The data obtained in this work do not support the so-called
noble impurity theory for HER on anodically-polarized magnesium, whereas evidences were
obtained that the dark oxy-hydroxide layer formed on this metal may exert a combined
catalytic efficiency toward HER whereas imperfectly passivating the metal surface

dioxide film formed by anodic pretreatment over pure commercial titanium metal for
biomaterial application was investigated in situ using scanning electrochemical microscopy
(SECM). Quantitative information on the electron transfer rates (keff) at the titanium surface
was obtained using the feedback operation of SECM using ferrocene-methanol (FcMeOH) as
electrochemical mediator. An increase of keff values with the increase of the negative
polarization was detected, a feature that correlates well with the decrease of titanium oxide
resistance with increasing cathodic polarization observed using electrochemical impedance
spectroscopy (EIS). In addition, SECM operation in the redox competition mode proved that
hydrogen was absorbed in the surface oxide film leading to changes in conductivity and
electrochemical reactivit
Potentiometric scanning electrochemical microscopy for the local characterization of the electrochemical behaviour of magnesium-based materials
The applicability of scanning electrochemical microscopy for the local detection and quantification of relevant species participating in the corrosion of magnesium-based materials is presented. The iron-magnesium galvanic pair exposed to aqueous NaCl solution was adopted as model system for this purpose. Mg2+ ion concentration and pH profiles were investigated using ion selective electrodes, containing a liquid membrane and Sb/Sb2O3 as sensor elements, respectively. Oxygen consumption at the substrate related to the cathodic reaction was also monitored with the antimony-based electrode though operated amperometrically. Data show a major production of hydroxyl anions at the cathodic sites as result of the oxygen reduction half-reaction, whereas in the vicinity of the magnesium surface pH is greatly affected by the anodic dissolution process instead. The later produces the release of metal cations accompanied by hydrogen evolution
A novel scanning electrochemical microscopy strategy for the investigation of anomalous hydrogen evolution from AZ63 magnesium alloy
The evolution of hydrogen gas from corroding magnesium arises not exclusively from the
cathodic half-cell reaction due to the consumption of the electrons released by the dissolving
metal, but anodized magnesium generates significant amounts of H2 gas as well. In addition,
the increase of the anodic overpotential enhances the rate of hydrogen gas generation.
Therefore, spatially-resolved detection of the actual sites for hydrogen evolution related to
anodically-activated sites is attempted using scanning electrochemical microscopy (SECM).
This work describes a new experimental procedure for SECM based on a three-step (off-onoff)
anodization operation sequence that was designed to obtain new insights into the behavior
of magnesium and magnesium alloys when they are subjected to anodic polarization. Results
obtained with scanning electrochemical microscopy (SECM) and scanning vibrating electrode
