    Investigating the features of three Italian populist parties (lega nord, forza Italia, 5 star movement) in terms of leadership and party claims

    The General Italian elections in 2013 offered interesting outcomes, which were assessed as the continuation of populist success which started from the early 90s when the major Italian populist parties Lega Nord and Forza Italia appeared on the political field. Populism in general represents a multifaceted concept and the nature of it varies according to a number of dimensions. As Paul Taggart emphasizes, considering the different ways in which populism is studied1, it represents a “highly chameleonic” phenomenon.2 The scholarly attempts to define populism as a concept offered a diverse and at the same time contradictious interpretations, which often consider populism from the different perspectives3. Italy is one of the relevant cases for the discussion of populism. The mentioned phenomenon has historically been successful in different regional contexts and managed to flourish in Italy as well in the early 90s. This period left remarkable trace in the history of the mentioned country, because exactly the Second Italian Republic witnessed the emergence of the three influential populist movements such as Lega Nord, Forza Italia and 5 Star Movement, which played and are still playing a significant role in the modern Italian politics. Numbers of works are dedicated to the study of the circumstances which are vulnerable for populist emergence; however less attention is paid to the analysis of how particular populist parties reflect the idea of populism in general. Moreover, the peculiarities of Italian populist movements are not thoroughly explored, especially considering the General elections of 2013 and the new political actor, 5 Star Movement. The following thesis aims to understand the nature of the three Italian populist parties and to find how they correspond to the major features of the populism phenomenon. Based on the existing literature regarding populism, the author of the following work identified two general (leadership and political claims) and six specific (Strong appeal to people, Charismatic leadership, Active use of media, anti-elitist, anti-establishment, anti-party rhetoric) features of populism and according to the mentioned features analyzed three Italian populist parties. Considering the main findings of the research, Italian populist parties correspond to the majority of populist features, however still there are differences according the specific nature of the political organization.http://www.ester.ee/record=b4410582~S58*es

    Investigation of factors influencing the immunogenicity of hCG as a potential cancer vaccine

    Human hCG and its β‐subunit (hCGβ) are tumour autocrine growth factors whose presence in the serum of cancer patients has been linked to poorer prognosis. Previous studies have shown that vaccines, which target these molecules and/or the 37 amino acid C‐terminal hCGβ peptide (hCGβCTP), induce antibody responses in a majority of human recipients. Here we explored whether the immunogenicity of vaccines containing an hCGβ mutant (hCGβR68E, designed to eliminate cross‐reactivity with luteinizing hormone) or hCGβCTP could be enhanced by coupling the immunogen to different carriers (KLH or Hsp70) using different cross‐linkers (EDC or GAD) and formulated with different adjuvants (RIBI or Montanide ISA720). While there was little to choose between KLH and Hsp70 as carriers, their influence on the effectiveness of a vaccine containing the BAChCGβR68E mutant was less marked, presumably because being a foreign species, this mutant protein itself might provide T‐helper epitopes. The mutant provided a significantly better vaccine than the hCGβCTP peptide irrespective of the carrier used, how it was cross‐linked to the carrier or which adjuvant was used when hCG was the target. Nonetheless, for use in humans where hCG is a tolerated self‐protein, the need for a carrier is of fundamental importance. Highest antibody titres were obtained by linking the BAChCGβR68E to Hsp70 as a carrier by GAD and using RIBI as the adjuvant, which also resulted in antibodies with significantly higher affinity than those elicited by hCGβCTP peptide vaccine. This makes this mutant vaccine a promising candidate for therapeutic studies in hCGβ‐positive cancer patients

    Cardiosphere-derived cells suppress allogeneic lymphocytes by production of PGE2 acting via the EP4 receptor

    derived cells (CDCs) are a cardiac progenitor cell population, which have been shown to possess cardiac regenerative properties and can improve heart function in a variety of cardiac diseases. Studies in large animal models have predominantly focussed on using autologous cells for safety, however allogeneic cell banks would allow for a practical, cost-effective and efficient use in a clinical setting. The aim of this work was to determine the immunomodulatory status of these cells using CDCs and lymphocytes from 5 dogs. CDCs expressed MHC I but not MHC II molecules and in mixed lymphocyte reactions demonstrated a lack of lymphocyte proliferation in response to MHC-mismatched CDCs. Furthermore, MHC-mismatched CDCs suppressed lymphocyte proliferation and activation in response to Concanavalin A. Transwell experiments demonstrated that this was predominantly due to direct cell-cell contact in addition to soluble mediators whereby CDCs produced high levels of PGE2 under inflammatory conditions. This led to down-regulation of CD25 expression on lymphocytes via the EP4 receptor. Blocking prostaglandin synthesis restored both, proliferation and activation (measured via CD25 expression) of stimulated lymphocytes. We demonstrated for the first time in a large animal model that CDCs inhibit proliferation in allo-reactive lymphocytes and have potent immunosuppressive activity mediated via PGE2

    Role of cAMP in the promotion of colorectal cancer cell growth by Prostaglandin E2

    <p>Abstract</p> <p>Background</p> <p>Prostaglandin E2 (PGE2), a product of the cyclooxygenase (COX) reaction, stimulates the growth of colonic epithelial cells. It is inferred that the abrogation of prostaglandins' growth-promoting effects as a result of COX inhibition underlies the advantageous effects of non-steroidal anti-inflammatory drugs in colorectal carcinoma (CRC). Despite this appreciation, the underlying molecular mechanisms remain obscure since cell culture studies have yielded discrepant results regarding PGE2's mitogenicity.</p> <p>Methods</p> <p>We have employed several alternative approaches to score cell proliferation and apoptosis of 4 CRC cell lines exposed to PGE2 under various conditions. To investigate the role of cAMP in PGE2's functions, activation of the cAMP pathway was assessed at different levels (changes in cAMP levels and PKA activity) in cells subjected to specific manipulations including the use of specific inhibitors or prostanoid receptor-selective agonists/antagonists.</p> <p>Results</p> <p>Our data document that the dose-response curve to PGE2 is 'bell-shaped', with nano molar concentrations of PGE2 being more mitogenic than micro molar doses. Remarkably, mitogenicity inversely correlates with the ability of PGE2 doses to raise cAMP levels. Consistent with a major role for cAMP, cAMP raising agents and pertussis toxin revert the mitogenic response to PGE2. Accordingly, use of prostanoid receptor-selective agonists argues for the involvement of the EP3 receptor and serum deprivation of HT29 CRC cells specifically raises the levels of Gi-coupled EP3 splice variants.</p> <p>Conclusion</p> <p>The present data indicate that the mitogenic action of low PGE2 doses in CRC cells is mediated via Gi-proteins, most likely through the EP3 receptor subtype, and is superimposed by a second, cAMP-dependent anti-proliferative effect at higher PGE2 doses. We discuss how these findings contribute to rationalize conflictive literature data on the proliferative action of PGE2.</p
