    Optimization of 3-D Wavelet Decomposition on Multiprocessors

    In this work we discuss various ideas for the optimization of 3-D wavelet/subband decomposition on shared memory MIMD computers. We theoretically evaluate the characteristics of these approaches and verify the results on parallel computers.     Balancing the Waiting Times in a Simple Traffic Intersection Model

    We propose a novel dynamical model of a simple traffic intersection, where the state variables represent the queue lengths and the mean waiting times in the queues. Including the mean waiting times in the model allows for a more fair traffic control, where the waiting times of the individual vehicles in the various streets of the intersection are taken into account to some degree. The model is linearized and its parameters are estimated using real traffic data measured during one day in Prague. For the balancing of the waiting times, two different controllers are considered: a linear quadratic regulator and a nonlinear model predictive controller. The controllers are evaluated in simulations where real traffic data is used for the incoming flows

V druhé části je uvedeno základní rozdělení nástrojů a popis nástrojových materiálů. Poslední část je zaměřena na porovnání sortimentu vybraných producentů nástrojů v konstrukční oblasti.This thesis is focused on high-performance turning tools. The first part deals with the characterization of turning, including basic kinematic relations. In the second part the division of tools and a the description of tool materials are introduced. The last part is centred on the assortment comparison of selected producers of tools in the field of construction.

    The evolution of LOL, the secondary metabolite gene cluster for insecticidal loline alkaloids in fungal endophytes of grasses.

    LOL is a novel secondary metabolite gene cluster associated with the production of loline alkaloids (saturated 1-aminopyrrolizidine alkaloids with an oxygen bridge) exclusively in closely related grass-endophyte species in the genera Epichloë and Neotyphodium. In this study I characterize the LOL cluster in E. festucae, including the presentation of sequence corresponding to 10 individual lol genes as well as defining the boundaries of the cluster and evaluation of the genomic DNA region flanking LOL in E. festucae. In addition to characterizing the LOL cluster in E. festucae, I present LOL sequence from two additional species, Neotyphodium coenophialum and Neotyphodium sp. PauTG-1. Together with two recently published LOL clusters from N. uncinatum, these data allow for a powerful phylogenetic comparison of five clusters from four closely related species. There is a high degree of microsynteny (conserved gene order and orientation) among the five LOL clusters, allowing us to predict potential transcriptional co-regulatory binding motifs in lol promoter regions. The relatedness of LOL clusters is especially interesting in light of the history of interspecific hybridizations that generated the asexual, Neotyphodium lineages. In fact, three of the clusters appear to have been introduced to different Neotyphodium species by the same ancestral Epichloë species, for which present day isolates are no longer able to produce lolines. To address the evolutionary origins of the cluster we have investigated the phylogenetic relationships of particular lol ORFs to their paralogous primary metabolism genes (and gene families) from endophytes, other fungi and even other kingdoms. I present extensive evidence that at least two individual lol genes have evolved from primary metabolism genes within the fungal ancestors of endophytes, rather than being introduced via horizontal gene transfer. I also present complementation studies in Neurospora crassa exploring the functional divergence of one lol gene from its primary metabolism paralog. While it is clear that these insecticidal compounds should convey a selective advantage to the fungus and its host, thus explaining preservation of the trait, this analysis provides an exploration into the evolutionary origin and maintenance of the genes that comprise the LOL and the cluster itself

    Podstatou této diplomové práce je nalezení vlivu průměru trysky na kvalitu tisku 3D tiskárny. V první části práce je stručně charakterizována technologie Rapid Prototyping a popsán projekt RepRap. Druhá část popisuje stavbu experimentální FDM 3D tiskárny. V následující kapitole jsou popsány technologické parametry tisku a jejich vliv na tištěný výrobek. Experimentální část práce je zaměřena na zjištění vlivu průměru výtlačné trysky na mechanické vlastnosti, kvalitu ploch a přesnost tvarů výsledného výrobku a následně doporučení provozních parametrů.The matter of this thesis is to find the influence of the diameter of the nozzle on the 3D printer´s print quality. The first part briefly characterizes the Rapid Prototyping technology and describes the RepRap project. The second part describes the construction of the experimental FMD 3D printer. The following section deals with technical printing parametres and their influence on the printed product. The experimental part of the thesis is focused on the impact of the discharge nozzle diameter on the mechanical properties, surface quality and shape accuracy of the final product and on the following recommendations of operational parametres.

    Comparison of the welding productivity MAG and FCAW

    Tato práce je zaměřena na obloukové svařování metodami MAG a FCAW. První část obsahuje stručný popis těchto technologií. Důraz je kladen na princip obloukového svařování v aktivním ochranném plynu a na přídavné materiály, zejména na plné a plněné svařovací dráty. Druhá, experimentální část, je zaměřena na porovnání metod MAG (135) a FCAW (136 a 138) z hlediska produktivity, nákladnosti a v neposlední řadě i kvality koutových svarů zhotovených v poloze svařování PF. Na základě vypočtených a naměřených hodnot získaných z experimentu byla vybrána nejvhodnější metoda pro danou polohu svařování.This thesis is focused on arc welding with MAG and FCAW methods. The first part contains a brief description of these technologies. Emphasis is laid on the principle of arc welding in active shielding gas and on additional materials, especially on solid and flux-cored tubular wires. The second, experimental part, centres on the comparison of MAG (135) and FCAW (136 and 138) methods in terms of the productivity, costs and, last but not least, the quality of fillet welds made in PF welding position. Based on the calculated and measured figures obtained from the experiment, the most suitable method for the given welding position was chosen.

    Evaluation of 3D Printing Use for Multinational Armed Forces Logistic Processes in Crisis Situations

    3D printing technology could be an option for the delivery of spare parts in a multinational army supply process, especially in crisis situations, when specific spare parts need to be available as soon as possible. The article deals with the selection of fused filament fabrication as an appropriate method for that purpose by comparing its advantages and disadvantages, as well as the best material (Nylon CF15 Carbon) based on important mechanical properties using the comparative and Saaty\u27s methods. Several samples were made to be exposed to UV radiation in a climate chamber (Q-LAB Q-SUN-Xe3 equipped with a DAYLIGHT-Q filter) and tested according to the ASTM D2565 and ASTM D4459 standards. To compare the mechanical properties of the samples exposed to UV radiation with those of the unexposed ones, a tensile test was done according to the CSN EN ISO 527-2 standard. Finally, the specific spare part was designed and 3D printed, and the delivery times of the standard supply period were compared with those of the 3D printing technology
