    Profitability of organic and conventional dairy production with different dietary proportions of high-quality grass silage

    Profitability in milk production was calculated using diets with three different proportions of high-quality grass silage. The treatments consisted of the same feeds, but differed in the dietary proportion of forage: low (L), medium (M) and high (H), representing one conventional and two organic diets. The calculations were based on results in feed intake and milk production from a large dairy cow experiment. In the calculations, three different districts with different conditions for farming were used as models, and calculations were performed on two different herd sizes. In addition, current financial supports were both included and excluded in the calculations. The results showed only minor differences in profitability between using in average 60% or 70% of dry matter (DM) of silage in diets used in organic production. In conventional production, it was profitable to increase the average dietary proportion of silage from 50% to 60% of DM when the prices of concentrate and grains were as high as at the present. The results showed clear economic benefits of increasing the proportion of high-quality silage in conventional Scandinavian dairy cow diets up to levels similar to the standards of the organic production system

    Characteristics and Predictors for Students Classified with Emotional and Behavioral Disorder Who Have Also Experienced Maltreatment

    Though experiencing maltreatment (abuse or neglect) appears to be common in students with the special education label of emotional and behavioral disorders (EBD), little research has been devoted to this topic by EBD educators. This paper uses archived file drawer data from 1992 that focuses on 149 students newly classified with EBD for whom a wide range of enrollment variables was collected, and who were subsequently followed up on an average of 8 years later to assess their educational outcomes. At enrollment, experiences of maltreatment were determined to have occurred in 57.7% of these participants. The group who experienced maltreatment was predicted at enrollment only by the family stress of having at least one natural parent with a history of psychiatric illness, although the concordance was not strong (52.3%). When the children who experienced maltreatment were next divided into two longitudinal groups according to educational outcomes (52.3% successful), the enrollment variables of the presence of anxiety and/or depressive disorder and younger age predicted the successful outcome group with good concordance (76.0%). Professional and programmatic implications for educators of students identified with EBD who have also experienced maltreatment are discussed, along with practical recommendations for serving this population

    Diagnostic and prognostic value of bone biomarkers in progressive knee osteoarthritis: a 6-year follow-up study in middle-aged subjects

    SummaryObjectiveTo determine the value of bone markers in early-stage progressive knee osteoarthritis (OA), a population-based cohort of middle-aged subjects with chronic knee complaints was followed over 6 years (two consecutive two 3-year periods).MethodsTibiofemoral (TF) and patellofemoral (PF) radiographs were graded in 128 subjects (mean age at baseline 45 ± 6.2 years) in 2002, 2005 and 2008. Bone formation was assessed by the serum concentration of procollagen type I amino-terminal propeptide (sPINP); bone resorption by the level of the C-terminal cross-linked telopeptides of type I collagen (sCTx-I); and bone mineralization by the values of osteocalcin (sOC) by electrochemiluminescence immunoassay. A novel marker of bone resorption, urinary osteocalcin midfragments (uMidOC), was assayed using enzyme linked immunosorbent assay (ELISA).ResultsSeveral diagnostic associations were found between the bone markers (PINP, OC, MidOC) and progressive OA expressed by TF osteophytosis. The increasing output of MidOC demonstrated several-fold higher risk for progressive TF osteophytosis [odds ratio (OR) 5.32; 95% confidence interval (CI) 1.41–20.06, P = 0.014] than other bone markers. The values of PINP had prognostic value for subsequent more severely expressed knee OA progression [r(s) = 0.460, P = 0.005].ConclusionsBone metabolism is activated in early-stage knee OA. OA progression was preceded by the enhanced bone formation (by PINP) and accompanied by the activation of bone formation (by PINP), non-collagenous bone resorption (by MidOC), as well as by changes in mineralization (by OC). All three bone markers had diagnostic value, and one of them, PINP, had also a predictive value for knee OA progression, especially for progressive osteophytosis

    LUXOR: An FPGA Logic Cell Architecture for Efficient Compressor Tree Implementations

    We propose two tiers of modifications to FPGA logic cell architecture to deliver a variety of performance and utilization benefits with only minor area overheads. In the irst tier, we augment existing commercial logic cell datapaths with a 6-input XOR gate in order to improve the expressiveness of each element, while maintaining backward compatibility. This new architecture is vendor-agnostic, and we refer to it as LUXOR. We also consider a secondary tier of vendor-speciic modifications to both Xilinx and Intel FPGAs, which we refer to as X-LUXOR+ and I-LUXOR+ respectively. We demonstrate that compressor tree synthesis using generalized parallel counters (GPCs) is further improved with the proposed modifications. Using both the Intel adaptive logic module and the Xilinx slice at the 65nm technology node for a comparative study, it is shown that the silicon area overhead is less than 0.5% for LUXOR and 5-6% for LUXOR+, while the delay increments are 1-6% and 3-9% respectively. We demonstrate that LUXOR can deliver an average reduction of 13-19% in logic utilization on micro-benchmarks from a variety of domains.BNN benchmarks benefit the most with an average reduction of 37-47% in logic utilization, which is due to the highly-efficient mapping of the XnorPopcount operation on our proposed LUXOR+ logic cells.Comment: In Proceedings of the 2020 ACM/SIGDA International Symposium on Field-Programmable Gate Arrays (FPGA'20), February 23-25, 2020, Seaside, CA, US

    The cell cycle checkpoint system MAST(L)-ENSA/ARPP19-PP2A is targeted by cAMP/PKA and cGMP/PKG in anucleate human platelets

    The cell cycle is controlled by microtubule-associated serine/threonine kinase-like (MASTL), which phosphorylates the cAMP-regulated phosphoproteins 19 (ARPP19) at S62 and 19e/α-endosulfine (ENSA) at S67and converts them into protein phosphatase 2A (PP2A) inhibitors. Based on initial proteomic data, we hypothesized that the MASTL-ENSA/ARPP19-PP2A pathway, unknown until now in platelets, is regulated and functional in these anucleate cells. We detected ENSA, ARPP19 and various PP2A subunits (including seven different PP2A B-subunits) in proteomic studies of human platelets. ENSA-S109/ARPP19–S104 were efficiently phosphorylated in platelets treated with cAMP- (iloprost) and cGMP-elevating (NO donors/riociguat) agents. ENSA-S67/ARPP19-S62 phosphorylations increased following PP2A inhibition by okadaic acid (OA) in intact and lysed platelets indicating the presence of MASTL or a related protein kinase in human platelets. These data were validated with recombinant ENSA/ARPP19 and phospho-mutants using recombinant MASTL, protein kinase A and G. Both ARPP19 phosphorylation sites S62/S104 were dephosphorylated by platelet PP2A, but only S62-phosphorylated ARPP19 acted as PP2A inhibitor. Low-dose OA treatment of platelets caused PP2A inhibition, diminished thrombin-stimulated platelet aggregation and increased phosphorylation of distinct sites of VASP, Akt, p38 and ERK1/2 MAP kinases. In summary, our data establish the entire MASTL(like)–ENSA/ARPP19–PP2A pathway in human platelets and important interactions with the PKA, MAPK and PI3K/Akt systems.     Private Law and the European Constitutionalisation of Values

    According to the CFREU, the EU is founded on the general values such as values of human dignity, freedom, equality and solidarity. In addition, the TEU refers to a more political set of foundational values, ie respect for human dignity, freedom, democracy, equality, the rule of law and respect for human rights. These references could be understood as purely ornamental, or as irrelevant in any case for private law. Indeed, it is true that the Court of Justice so far has never made any references to these values in private law cases. Still, the Court already has shown boldness before in the context of the interpretation and review of secondary EU law in private law cases, when it discovered general principles of EU law and general principles of civil law. Therefore, it should not be excluded that the Court may be tempted one day to follow the example of the German constitutional court that famously understands its national constitution as expressing an objective system of constitutional values. This paper explores what such an understanding of private law as an instrument for furthering common European values would entail and examines whether such an ethical reading of European private law would be desirable. It argues that the promotion by the EU of a set of official values through its laws is not compatible we the respect we owe each other in a society characterised by reasonable pluralism. In addition, it points to further difficulties, both of a moral and a practical nature, of the idea of advancing ethical values through private law. It concludes that although it is very well thinkable that the values to which the TEU and the Charter refer will one day be interpreted as an objective value system with (indirect) horizontal effects, the Court of Justice nevertheless should refrain from going down that road

    S4S8-RPA phosphorylation as an indicator of cancer progression in oral squamous cell carcinomas.

    Oral cancers are easily accessible compared to many other cancers. Nevertheless, oral cancer is often diagnosed late, resulting in a poor prognosis. Most oral cancers are squamous cell carcinomas that predominantly develop from cell hyperplasias and dysplasias. DNA damage is induced in these tissues directly or indirectly in response to oncogene-induced deregulation of cellular proliferation. Consequently, a DNA Damage response (DDR) and a cell cycle checkpoint is activated. As dysplasia transitions to cancer, proteins involved in DNA damage and checkpoint signaling are mutated or silenced decreasing cell death while increasing genomic instability and allowing continued tumor progression. Hyperphosphorylation of Replication Protein A (RPA), including phosphorylation of Ser4 and Ser8 of RPA2, is a well-known indicator of DNA damage and checkpoint activation. In this study, we utilize S4S8-RPA phosphorylation as a marker for cancer development and progression in oral squamous cell carcinomas (OSCC). S4S8-RPA phosphorylation was observed to be low in normal cells, high in dysplasias, moderate in early grade tumors, and low in late stage tumors, essentially supporting the model of the DDR as an early barrier to tumorigenesis in certain types of cancers. In contrast, overall RPA expression was not correlative to DDR activation or tumor progression. Utilizing S4S8-RPA phosphorylation to indicate competent DDR activation in the future may have clinical significance in OSCC treatment decisions, by predicting the susceptibility of cancer cells to first-line platinum-based therapies for locally advanced, metastatic and recurrent OSCC
