    High Accuracy Determination of Rheological Properties of Drilling Fluids Using the Marsh Funnel

    Efficient and safe drilling operations require precise determination of rheological properties in drilling fluids, encompassing dynamic viscosity for Newtonian fluids, and apparent viscosity, plastic viscosity, and yield point for non-Newtonian fluids. Conventional viscometers like vibrating wire, ZNN-D6, and Fann-35 offer high accuracy but are limited by cost and complexity in small-scale industries and labs. To address this, our research presents a novel mathematical model based on the Herschel-Bulkley model, aiming to accurately characterise drilling fluids' rheological properties using the Marsh funnel as an alternative device -- an economical, operator-friendly, and power-independent equipment. Drawing inspiration from seminal works by Li et al. (2020), Sedaghat (2017), and Guria et al. (2013), this innovative framework establishes a universal inverse linear relationship between a fluid's flow factor and final discharge time. For any fluid, it utilises its density and flow factor (or final discharge time) to determine all its rheological properties. Specifically, it evaluates dynamic viscosity for Newtonian fluids, apparent viscosity, plastic viscosity, and yield point for weighted non-Newtonian fluids, and apparent viscosity for non-weighted non-Newtonian fluids, with average systematic errors (against Fann-35 measurements) of 0.39%, 3.52%, 2.17%, 18.38%, and 5.84%, respectively, surpassing the precision of alternative mathematical models found in the aforementioned literature. Furthermore, while our framework's precision in plastic viscosity and yield point assessment of non-weighted non-Newtonian fluids slightly lags behind the framework of Li et al. (2020), it outperforms the model of Sedaghat (2017). In conclusion, despite minor limitations, our proposed mathematical model holds huge promise for drilling fluid rheology in petroleum, drilling, and related industries.Comment: 57 pages, 1 figure, and 10 tables.     Study on thyroid function test in pregnancy in a tertiary care hospital

    Thyroid disorder is a very common endocrine problem encountered by pregnant women. Maternal thyroid dysfunction is associated with adverse outcome both in mother and fetus. The aim of the study: to find out the prevalence of various thyroid disorders in pregnant women attending antenatal clinic. Materials and methods. This prospective cross-sectional study was carried out in the Department of Obstetrics and Gynecology, F.M. Medical College & Hospital, Balasore, Odisha from June 2020 to May 2021. 220 women with uncomplicated singleton pregnancy were included. Serum Thyroid-stimulating hormone (TSH), free T4 (FT4) and free T3 (FT3) were estimated by using electro-chemiluminescence immunoassay technique. Results. Out of 220 pregnant women screened for thyroid dysfunction, 68 were found to have thyroid disorders. 27.3 % of pregnant women had subclinical hypothyroidism, 1.4 % had overt hypothyroidism, 1.8 % had subclinical hyperthyroidism and 0.5 % had overt hyperthyroidism. Prevalence of subclinical hypothyroidism was 6.36 % when the upper reference limit of TSH level taken as 4 mIU/L. Prevalence of thyroid disorder among pregnant women in the age groups 18–25 years, 26–30 years and 31–40 years were 28.9 %, 32.1 % and 38.9 % respectively. There were 35.5 %, 28 % and 26 % pregnant women with thyroid disorders in the first, second and third trimester respectively. Prevalence of both subclinical and overt hypothyroidism were more in multigravida compared to primigravida. Conclusion. Our study revealed high prevalence of thyroid disorders in pregnant women and maternal subclinical hypothyroidism was the most common pattern

    Japakusumadi Yoga is an Ayurvedic formulation, indicated for contraception in Ayurveda. This study determines acute toxicity of ‘Japakusumadi Yoga’ an oral formulation in wister albino rats. Single dose acute toxicity was assessed by employing OECD guidelines 425 using AOT software. Test formulations was administered to overnight fasted animals and 14 days observation of dosed (up down as per requirement) rats was done for general appearance, cage side behaviour including increased or decreasing motor activity, convulsions, straub’s reaction, catatonia, muscle spasm, spasticity, ophisthotonus, hyperesthesia, muscle relaxation, anaesthesia, arching and rolling, lacrimation, salivation, diarrhoea, writhing movement, mode of respiration and changes in skin colour etc., with mortality and autopsy finding in case of dead animal. Based on the observation made and recorded it can be concluded that the test drug is without any toxic potential even at the dose of 2000 mg/kg in animals equivalent to 22.4g for human being

    Effect of varieties and storage on the quality parameters of nectarine (Prunus persica)-based intermediate moisture food (IMF) products

    Fruits play an important role in maintaining a healthy life. Nectarine is a hybrid fruit of peach and plum, wherein efforts were made to develop intermediate moisture food products (jam and jelly) from nectarine varieties (May Fire, Snow Queen, and Silver King). The study aimed to determine the effect of storage on the nutritional (TSS, pH, acidity, ascorbic acid, and sugars) and sensory parameters (color, taste, flavor, texture, and overall acceptability) of jam and jelly at different storage intervals. Storage had a nonsignificant effect on the total soluble solids, with reported mean values of 69.670 Brix, while the pH content of jam varied significantly from 2.90-2.20 during 6 months of storage. The values for acidity and total sugars increased (P≤0.05) significantly from 1.92-2.03 percent and 57.04 to 56.93 percent, respectively. However, the ascorbic acid content decreased significantly from 4.64 - 1.66 mg/100 g. In the case of jelly, the total soluble solids and pH decreased from 67.78 – 67.440 Brix and 2.70 – 2.48, respectively, during storage for 6 months at ambient temperature. The ascorbic acid content decreased from 4.56-2.10 mg/100 g. Among cultivars, there was a nonsignificant difference in the nutritional parameters of jam, but in the case of jelly, different cultivars had a significant effect on TSS, pH, and ascorbic acid content. Organoleptically, the nectarine jam was rated as ‘liked very much’, while the jelly ‘liked slightly’, with good storage acceptance up to 6 months. Being nectarine as a superfood can be explored to develop speciality food products for vulnerable sections of society

    Effects of Gibberellic acid (GA3) on shelf life and physiochemical properties of mango (Mangifera indica L. var Bombay green)

    The present study investigated the effect of GA3 on the physicochemical properties and shelf life of mango (Mangifera indica L. var. Bombay green) from 31st May 2022 to 8th June 2022 at the Central Laboratory of GPCAR collage Gothgaun, Morang, Nepal. The study's goal was to find the right gibberellic acid concentration to use in mangoes that were collected at a mature stage to delay fruit ripening, preserve quality, and lengthen shelf life. The experiment was laid out in a Completely Randomized Design (CRD) with five treatments and four replications. Mature freshly harvested mango fruits of uniform size were dipped into an aqueous solution of gibberellic acid at 0 ppm (T1), 100 ppm (T2), 200 ppm (T3), 300 ppm (T4) and 400 ppm (T5) for 10 minutes. Data on physicochemical parameters (mango pulp pH, total soluble solids, titratable acidity, physiological weight loss, and shelf life) were statistically analyzed through biochemical analyses. Further, fruits treated with 400 ppm of GA3; resulted in the lowest physiological loss in weight (22.08%), the minimum pulp pH (5.02), and the minimum titratable acidity (0.14%) on the 8th day after storage. The highest total soluble solid (19.85°B) was recorded with GA3 @400ppm, while the lowest soluble solids (16.90°B) were recorded with control ppm on the 8th day after storage. Fruits treated with GA3 at 400 ppm had the longest shelf life (7.17 days), while fruits treated with GA at 300 ppm had the shortest shelf life (7.19 days). Therefore, the best results were obtained when gibberellic acid was applied at 400 ppm, which extended the shelf life and physiochemical traits of mango fruits

    Role of polyvinyl pyrrolidone as a capping agent in the synthesis of magnetite (Fe3O4) nanoparticles

    Uncapped magnetite (Fe3O4) and magnetite capped with polyvinyl pyrrolidone (PVP) (Fe3O4-PVP) have been synthesized by a rapid, cost effective aqueous precipitation method. The nanoparticles are characterized by powder X-ray diffraction, scanning electron microscopy, Fourier transform infrared spectroscopy and UV spectroscopy. The role of PVP as a capping agent in the synthesis of magnetite nanoparticles has been investigated. Thermal stability and surface charge of the nanoparticles have been characterized by thermogravimetric analysis, differential scanning calorimetry (DSC) and zeta potential measurements. Results suggest that PVP as a capping agent reduces the grain size, regularizes the shape, and increases the crystallinity of Fe3O4 nanoparticles. Thermal stability and surface charge of Fe3O4-PVP nanoparticles are also significantly higher as compared to the uncapped Fe3O4 nanoparticles. Magnetic characterization by vibrating sample magnetometry of both Fe3O4 and Fe3O4-PVP nanoparticles indicates the superparamagnetic behavior of the nanoparticles.

    Naphthoquinone-mediated inhibition of lysine acetyltransferase KAT3B/p300, basis for non-toxic inhibitor synthesis

    Hydroxynaphthoquinone-based inhibitors of the lysine acetyltransferase KAT3B (p300), such as plumbagin, are relatively toxic. Here, we report that free thiol reactivity and redox cycling properties greatly contribute to the toxicity of plumbagin. A reactive 3rd position in the naphthoquinone derivatives is essential for thiol reactivity and enhances redox cycling. Using this clue, we synthesized PTK1, harboring a methyl substitution at the 3rd position of plumbagin. This molecule loses its thiol reactivity completely and its redox cycling ability to a lesser extent. Mechanistically, non-competitive, reversible binding of the inhibitor to the lysine acetyltransferase (KAT) domain of p300 is largely responsible for the acetyltransferase inhibition. Remarkably, the modified inhibitor PTK1 was a nearly non-toxic inhibitor of p300. The present report elucidates the mechanism of acetyltransferase activity inhibition by 1,4-naphthoquinones, which involves redox cycling and nucleophilic adduct formation, and it suggests possible routes of synthesis of the non-toxic inhibitor
