    Evaluation of a lateral flow device for in-field detection of Banana Xanthomonas Wilt and its application in tracking the systemicity of Xanthomonas campestris pv. musacearum

    Early detection of Banana Xanthomonas Wilt (BXW) in the field and immediate destruction of infected plants or plant tissue are key control methods to prevent the introduction and spread of BXW. This requires rapid, cost effective and an on-site diagnostic tool to detect the bacterium, Xanthomonas campestris pv musacearum (Xcm). Polymerase chain reaction (PCR) detection technique for BXW is efficient but requires expensive equipment and knowledgeable expertise; this limits PCR application to the laboratory. This study therefore was carried out to evaluate the enzyme - linked immunosorbent assay (ELISA) tool configured as a lateral flow device (LFD) for detection of Xcm. Studies on the systemicity of Xcm in banana were carried out using the BXW - LFD in a field trial of 300 banana plants of Pisang Awak inoculated with the Xcm at Kiifu Forest, Mukono District, Uganda. Pseudo-stem samples from symptomatic and asymptomatic suckers were collected and tested with the LFD and the results compared with conventional PCR using the GspDm BXW primers.     Community mobilization: a key to effective control of banana Xanthomonas wilt

    Banana xanthomonas wilt (BXW) remains a major threat to banana, an important food and income crop for 12 million poor small-holder farmers in Uganda. Although, BXW has been controlled to some extent in parts of South-western Uganda, it is still a big problem in banana growing areas of Central and Eastern Uganda. We hypothesized that differential success in BXW control is mainly due to approaches used in the BXW control. This paper therefore, evaluates stakeholder mobilization approaches used in promoting technologies for BXW control in Uganda between 2006 and 2009. Results showed that farmer field schools host communities had more farmers (33%) that had low or no BXW infection (<10 infected plants) as compared to smaller proportions (23.5%) of farmers from communities that were using community action or that were mobilised using the traditional approach (22.9%) to control BXW. There was higher BXW prevalence in communities that were using community action (68.8%) or were mobilised traditionally (66.3%) than in those that hosted farmer field schools (43.4%). Consequently, there was higher (53%) banana production recovery on farms that hosted farmer field schools than those that used other institutional approaches (22%). BXW was better controlled by farmers mobilised using farmers field schools than those mobilized through community or traditional approaches

    Genetically engineered East African highland bananas – proximate analysis and effect of cooking on the enhanced provitamin A levels

    Micronutrient deficiency is a major challenge in the developing world. This is mainly attributed to over-reliance on starchy staples foods such as cassava, rice and banana among others, which are deficient in micronutrients such as vitamin A and iron. Strategies put in place to provide a solution to micronutrient deficiencies such as dietary supplementation of vitamin A and food fortification have not been successful in the developing world due to high costs and unreliable supply chains (food and medical). Biofortification of the easily accessible staple foods could help reduce this problem associated with micronutrient deficiency. On this account, the Biofortification project in Uganda under the National Banana Program developed transgenic East African Highland Bananas (EAHBs) (M9 and Nakitembe) with enhanced levels of provitamin A (PVA) using the Fe’i banana-derived phytoene synthase 2a (MtPsy2a) gene. To determine the nutritional quality of the transgenic bananas, an analysis of the proximate composition of the biofortified East African Highland Bananas was carried out. The effect of cooking on retention of provitamin A carotenoids (pVAC), was assessed using two cooking methods; boiling and steaming (most common methods of preparing cooking banana meals in Uganda). It was observed that there were no significant (P≤0.05) differences in moisture content (P=0.4287), carbohydrate (P=0.3966), crude fat (P=0.4051), crude fiber (P=0.3214), protein (P=0.0858) and ash content P=0.1336) between transgenic and non-transformed bananas. It was found that steaming, as a cooking method allowed for retention of more provitamin A carotenoids compared to boiling. Comparison of the cultivars on their retention of provitamin A carotenoids, results indicated that Nakitembe was superior to M9. Genetic engineering of bananas by biofortification has no effect on major food components in EAHBs (M9 and Nakitembe) and, therefore, genetically modified M9 and Nakitembe are substantially equivalent to the non-transgenic controls and the biofortified bananas can provide the necessary nutrients even after cooking. This&nbsp; data will inform subsequent steps for the commercialization of biofortified EAHBs

    Management of Banana Xanthomonas Wilt: Evidence from impact of adoption of cultural control practices in Uganda

    Banana Xanthomonas Wilt (BXW) is an important emerging and non-curable infectious plant pathogen in sub-Saharan Africa that can cause up to 100% yield loss, negatively impacting sustainable access to food and income to more than 100 million banana farmers. This study disentangles adopters into partial and full adopters to investigate the factors that are relevant to sustain the adoption process of BXW control practices and quantifies the impact of adopting the practices. Data from a randomly selected sample of 1200 banana farmers in Uganda where the disease is endemic was used. A multinomial logit model was used to determine the factors affecting adoption of control practices and augmented inverse probability weighting was employed to estimate the impacts of adoption on banana productivity and sales. Results show that training a woman farmer and having diverse sources of information about BXW control practices increased adoption of the control practices and reduced the disease incidences. Farmers who adopted all the recommended control practices achieved significantly the highest values of banana production and sales. We conclude that improving information access through farmers’ preferred communication channels, having women-inclusive trainings, and a combination of cultural practices are effective ways for sustaining adoption of the control practices

    Farmers' awareness and application of banana Xanthomonas wilt control options: The case of Uganda and Kenya

    Banana is a key staple and cash crop in east and central Africa. However, the recent outbreak of Banana Xanthomonas wilt (BXW) has threatened farmers whose livelihoods depend on the crop. Since 2002, stakeholders embarked on campaigns to sensitize farmers on the disease and its management. This study evaluated the effectiveness of these efforts by examining farmers' awareness of the disease, control options and their application in Uganda and Kenya. Data on BXW distribution and incidence, farmers' awareness of symptoms, mechanisms of spread and control options of the disease and banana production were collected through household surveys conducted in 2010. The results indicated that most farmers (> 90%) were aware of the disease and its symptoms and more than 50% were aware of the recommended control measures. Farmers obtained information on the disease from multiple sources but farmer-to-farmer interaction was the main source suggesting the key role rural social networks play in managing the disease. Not all affected farmers were able to apply control measures due to cost of intervention, lack of labour and inputs. The study recommends strengthening of linkages among stakeholders for better coordination of efforts and participatory development and promotion of farmer-oriented interventions that address farmer constraints

    Canopy management, leaf fall and litter quality of dominant tree species in the banana agroforestry system in Uganda

    Small-scale farmers in the banana-coffee agro-zone of Central Uganda plant andmaintain trees to provide a range of benefits. However, the impact of trees on soil fertility and crop yields is small. On many farms, trees exist in infinite numbers, compositions, with no proper spacing, sequencing and canopy management  recommendations. The objectives of this study were to: quantify and compare leaf fall and leaf litter biomass for the dominant tree species subjected to different pruning regimes and during different seasons and determine the nutrient  concentrations in leaf litter materials in order to assess their potential to improve soil fertility. A survey was conducted on 30 randomly selected farms to examine and  document the tree species. Four tree species were dominant; Ficus natalensis (15.3%), Albizia coriaria (10.2%), Artocarpus  heterophyllus (10.0%) and Mangifera indica (9.2%). Experiments such as canopy pruning, collecting leaf fall and weights of pruned leaf biomass were established for the four dominant trees on five randomly selected farms. A total of 120 trees were subjected to three pruning regimes (0%, 25% and 50%). Litter traps (1 m × 1 m) were placed 2 m from the tree trunk to collect falling leaves. Leaf biomass was collected from pruned branches and eighed. Leaf samples were collected from pruned leaves and a composite sample analysed for carbon and nutrient concentrations. Results showed that 40 tree and shrub species belonging to 21 families existed on farms. Leaf fall was highest in the dry season and A. Heterophyllus registered the highest amount. Leaf biomass from pruned trees was heaviest for M. indica (42.4 ±3 kg) in the wet season and at the 50% pruning regime (45.7± 2.1kg). Total  nutrient concentrations were in the order A. coriaria&gt;F. natalensis&gt;A.heterophyllus&gt;M.indica. We concluded that F. natalensis and A. Coriaria should be integrated more into the agroforestry system to improve soil fertility. A.heterophyllus and M.indica should be planted on farm boundaries.     Genome Sequencing of Xanthomonas vasicola Pathovar vasculorum Reveals Variation in Plasmids and Genes Encoding Lipopolysaccharide Synthesis, Type-IV Pilus and Type-III Secretion Effectors

    This is the final version. Available on open access from MDPI via the DOI in this recordXanthomonas vasicola pathovar vasculorum (Xvv) is the bacterial agent causing gumming disease in sugarcane. Here, we compare complete genome sequences for five isolates of Xvv originating from sugarcane and one from maize. This identified two distinct types of lipopolysaccharide synthesis gene clusters among Xvv isolates: one is similar to that of Xanthomonas axonopodis pathovar citri (Xac) and is probably the ancestral type, while the other is similar to those of the sugarcane-inhabiting species, Xanthomonas sacchari. Four of six Xvv isolates harboured sequences similar to the Xac plasmid, pXAC47, and showed a distinct Type-IV pilus (T4P) sequence type, whereas the T4P locus of the other two isolates resembled that of the closely related banana pathogen, Xanthomonas campestris pathovar musacearum (Xcm). The Xvv isolate from maize has lost a gene encoding a homologue of the virulence effector, xopAF, which was present in all five of the sugarcane isolates, while xopL contained a premature stop codon in four out of six isolates. These findings shed new light on evolutionary events since the divergence of Xvv and Xcm, as well as further elucidating the relationships between the two closely related pathogens.This study was supported in part by the National Agriculture Research Organisation, Uganda, under the MSI/World Bank grant 2009
