Platelet Carbonic Anhydrase II, a Forgotten Enzyme, May Be Responsible for Aspirin Resistance
Background. Thromboembolic events constitute a major health problem, despite the steadily expanding arsenal of antiplatelet drugs. Hence, there is still a need to optimize the antiplatelet therapy. Objectives. The aim of our study was to verify a hypothesis that there are no differences in platelet proteome between two groups of healthy people representing different acetylsalicylic acid (aspirin) responses as assessed by the liquid chromatography/mass spectrometry (LC/MS) technique. Patients/Methods. A total of 61 healthy volunteers were recruited for the study. Physical examination and blood collection were followed by platelet-rich plasma aggregation assays and platelet separation for proteomic LC/MS analysis. Arachidonic acid- (AA-) induced aggregation (in the presence of aspirin) allowed to divide study participants into two groups aspirin-resistant (AR) and aspirin-sensitive (AS) ones. Subsequently, platelet proteome was compared in groups using the LC/MS analysis. Results. The LC/MS analysis of platelet proteome between groups revealed that out of all identified proteins, the only discriminatory protein, affecting aspirin responsiveness, is platelet carbonic anhydrase II (CA II). Conclusions. CA II is a platelet function modulator and should be taken into consideration as a cardiovascular event risk factor or therapeutic target

Breast cancer family history and allele-specific DNA methylation in the Legacy Girls Study
Family history, a well-established risk factor for breast cancer, can have both genetic and environmental contributions. Shared environment in families as well as epigenetic changes that also may be influenced by shared genetics and environment may also explain familial clustering of cancers. Epigenetic regulation, such as DNA methylation, can change the activity of a DNA segment without a change in the sequence; environmental exposures experienced across the life course can induce such changes. However, genetic-epigenetic interactions, detected as methylation quantitative trait loci (mQTLs; a.k.a. meQTLs) and haplotype-dependent allele-specific methylation (hap-ASM), can also contribute to inter-individual differences in DNA methylation patterns. To identify differentially methylated regions (DMRs) associated with breast cancer susceptibility, we examined differences in white blood cell DNA methylation in 29 candidate genes in 426 girls (ages 6-13 years) from the LEGACY Girls Study, 239 with and 187 without a breast cancer family history (BCFH). We measured methylation by targeted massively parallel bisulfite sequencing (bis-seq) and observed BCFH DMRs in two genes: ESR1 (Δ4.9%, P = 0.003) and SEC16B (Δ3.6%, P = 0.026), each of which has been previously implicated in breast cancer susceptibility and pubertal development. These DMRs showed high inter-individual variability in methylation, suggesting the presence of mQTLs/hap-ASM. Using single nucleotide polymorphisms data in the bis-seq amplicon, we found strong hap-ASM in SEC16B (with allele specific-differences ranging from 42% to 74%). These findings suggest that differential methylation in genes relevant to breast cancer susceptibility may be present early in life, and that inherited genetic factors underlie some of these epigenetic differences
"Pi of the Sky" - all-sky, real-time search for fast optical transients
An apparatus to search for optical flashes in the sky is described. It has
been optimized for gamma ray bursts (GRB) optical counterparts. It consists of
2x16 cameras covering all the sky. The sky is monitored continuously and the
data are analysed on-line. It has self-triggering capability and can react to
external triggers with negative delay. The prototype with two cameras has been
installed at Las Campanas (Chile) and is operational from July 2004. The paper
presents general idea and describes the apparatus in detail. Performance of the
prototype is briefly reviewed and perspectives for the future are outlined
High-Pressure EPR Study of Spin Molecular Bistability in
The spin transitions in were studied by EPR under hydrostatic pressure in the temperature range 80-332 K. The structural changes in the complex anion induced by the pressure above 350 MPa (when LS-2 phase appears) are observed, which are manifested in abrupt changes in factor and linewidth of the low-spin phase LS. It is also manifested in the change in the shape of EPR spectrum. The high-spin low-spin transition shows hysteresis whose width is constant up to about 265 K and then rapidly increases. The spin phase diagram studied shows a large similarity to the phase diagram of , which means that characteristics of the transition depend mainly on complex anion, not on cation. The hysteresis indicates on a cooperative character of the spin transition and on grouping of the complexes in domains
Device for compressibility and thermal expansion study up to 3 GPa
We present the realization of a project of a device utilizing a standard high-pressure chamber adapted to compressibility and thermal expansion measurements of solids at hydrostatic pressures up to 3 GPa. The device has been tested on the triglycine sulphate crystal (TGS)
Influence of Hydrostatic Pressure on the Zero-Field Splitting in NiSnCl·6HO
The dependence of the crystal-field splitting parameter D in NiSnCl·6HO on hydrostatic pressure was determined by the electron paramagnetic resonance method, for the pressures up to 500 MPa. The derivative (∂D/∂p) as well as the temperatures and pressures at which the D-parameter reaches zero, were determined. The molecular mechanism responsible for the temperature and pressure changes of the D-parameter is similar to that one observed in the isomorphic NiSiF·6HO
