This feeling is never the main subject of this short stories, but he appears in the moral choices and judgments of the characters. This paper focuses above all on the contempt as one of the marks of pride in its noble, courteous and evangelical dimension.Le tableau des grandeurs et des folies, des exploits et des faiblesses humaines dans L’Heptaméron de Marguerite de Navarre ne saurait être complet sans l’évocation du mépris. Od dziennikarza wymaga się rzetelności oraz staranności w przygotowaniu materiałów prasowych. Celem artykułu jest ustalenie, jakie działania mają oni podejmować, aby dochować standardów przewidzianych obowiązującym prawem. Analizie podlegały przepisy dotyczące zarówno uprawnień dziennikarzy, jak i te mające chronić dobra osób będących przedmiotem zainteresowania pracowników środków masowego przekazu. Podjęto także temat technik oddziaływania na przedstawicieli mediów przez podmioty zainteresowane inicjowaniem lub tłumieniem krytyki prasowej.The journalist is required to be reliable and diligent in preparing the press materials. The aim of the paper is to determine the activities that should be undertaken by the journalists to meet the standards provided by applicable law. Regulations referring to the journalists’ rights as well as regulations intended to protect the rights of persons who are the subject of interest to mass media employees were analyzed. The subject of the techniques that are used by the entities interested in initiating or suppressing press criticism to impact media representatives is also undertaken

Wydawanie gazet bezpośrednio przez administrację terenową lub na jej zlecenie spotyka się jednak z krytyką. Podstawowym zarzutem jest nieuczciwa konkurencja wobec czasopism prywatnych. Zdaniem niektórych regionalnych izb obrachunkowych, nadzorujących jednostki samorządu terytorialnego, publikowanie płatnych reklam przez prasę samorządową jest łamaniem dyscypliny finansów publicznych. W opinii innych izb – to niezbywalne prawo prasy, także samorządowej. Na podstawie przeprowadzonych badań można stwierdzić, że sami dziennikarze mediów komercyjnych w większości uważają, że prasa samorządowa powinna być wydawana. Na celowość jej edycji wskazują też studenci dziennikarstwa i komunikacji społecznej, dostrzegając przy tym – podobnie jak dziennikarze – słabe strony prasy samorządowej, czyli np. przypadki nadmiernego akcentowania komunikacji perswazyjnej czy braku profesjonalizmu zespołów redakcyjnych. Ogólna ocena prasy samorządowej formułowana przez środowisko dziennikarskie jest pozytywna – przy świadomości, że prasa taka w obowiązującym stanie prawnym nie wypełnia funkcji kontrolnej wobec samorządów.The self-government press has become a permanent part of Polish media landscape. Publishing of newspapers directly by the local administration or by its order, however, meets with criticism. Their main criticism is unfair competition against private newspapers. According to some regional accounting chambers, supervising self-government units, publishing paid advertising by the selfgovernment press is the violation of public finance discipline. In the opinion of other chambers – it is the inalienable right of the press, including self-government press. Based on the surveys it can be stated that commercial media journalists mostly think that self-government press should be published. On the advisability of their publishing point out also students of journalism and communication, being aware – like the journalists – of weaknesses of self-government press, that is, e.g. the cases of overemphasizing persuasive communication or lack of professional editorial teams. Overall rating of the self-government press formulated by the journalism circle is positive – with the awareness that the press in the legal situation does not fulfill the control function to self-governments

The aim of this article is to analyse the prefaces to the French Renaissance apocalyptic epics of the years 1576-1606 (works by Jacques de Billy, Guillaume de Chevalier, Michel Quillian and Jude Serclier) in order to understand the literary and extra-literary issues addressed by their authors. This comparison will highlight in particular the ethos of the author, the problem of representation in a biblical poem and its eschatological dimension, the place of pagan fiction and encyclopaedic erudition, and the rhetorical strategies employed

Dziennikarze wobec działań public relations w województwie śląskim

Właściwie realizowaną aktywność w sferze media relations z całą pewnością zaliczyć trzeba do narzędzi umożliwiających efektywne zarządzanie komunikacją zewnętrzną. Biorąc pod uwagę obserwowane przez naukowców tendencje można przewidywać, że w Polsce, po zakończeniu badanego okresu, nastąpi dalszy wzrost znaczenia public relations, co wyniknie ze spotęgowanego zainteresowania kształtowaniem społecznego wizerunku przez podmioty gospodarcze, organizacje społeczne czy polityczne, jednostki administracyjne itd. Jednocześnie sfera media relations podlegać będzie stałym oraz coraz szybciej zachodzącym przemianom. Podobnie zresztą jak sytuacja środków masowego przekazu. W badanym okresie warunki wykonywania zawodu przez dziennikarzy w województwie śląskim były nieporównywalnie gorsze od komfortu świadczenia pracy, możliwości rozwoju kariery, a także dostępu do konkurencyjnych ofert zmiany miejsca zatrudnienia, jakie występowały w innych częściach kraju, a szczególnie w Warszawie. Prognozując kierunek zmian kondycji dziennikarstwa regionalnego należy założyć, że będą one uzależnione od potencjału ekonomicznego przedsiębiorstw medialnych oraz stosowanej przez nie polityki wobec jednostek odpowiedzialnych za działalność redakcyjną. Większe nakłady na motywowanie i wyposażenie dziennikarzy spowodują wzrost profesjonalizmu reporterów, ułatwią angażowanie wykwalifikowanych kadr dążących do specjalizacji tematycznej, poprawią odbiór prestiżu zawodu, zintegrują środowisko. Phenotype of dendritic cells generated in the presence of non-small cell lung cancer antigens - preliminary report.

Therapeutic outcomes of definitively treated non-small-cell lung cancer (NSCLC) are unacceptably poor. It has been proposed that the manipulation of dendritic cells (DCs) as a "natural" vaccine adjuvant may prove to be a particularly effective way to stimulate antitumor immunity. Presently, there is no standardized methodology for preparing vaccines and many questions concerning the optimal source and type of antigens as well as maturation state and activity of DCs are still unsolved. The study population comprised of ten patients with histologically confirmed NSCLC (mean age: 67.63 +/- 6.15 years). Resected small tumor pieces were placed in tissue culture dishes containing different growth factors in order to obtain pure cancer cells. Seven days after the operation, the PBMC were collected and monocytes were purified by the adherence to culture dishes. Monocytes were cultured in RPMI 1640 medium supplemented with 10% of autologous plasma in the presence of rhIL-4 and rhGM-CSF to generate immature autologous (DCs). TNF-alpha with or without tumor cells' lysate were added to maturation of DCs. After 7 days of culture, DCs were harvested and the expression of CD1a, CD83, CD80, CD86 and HLA-DR antigens were analyzed by flow cytometry. We discovered higher (p=0.07) percentage of semimature DCs in tumor cell lysate culture in comparison with TNF-alpha culture (21.22 +/- 16.82% versus 11.27 +/- 11.64%). The expression of co-stimulatory and maturation markers (CD86, CD83 and HLA-DR) was higher on DCs from the culture with tumor cell lysate compared with TNF-alpha culture as a control. Specimen of NSCLC's culture prepared in this way could generate differences in DCs phenotype, which may have an influence on the therapeutic and protective antitumor immunity of the vaccine. Our research seems to be the next step in the development of DC-based vaccine. We are going to continue the investigation to start the preparation of a pattern of immunological vaccine against lung cancer

Bliźnięta syjamskie – diagnoza prenatalna I trymestru ciąży. Analiza przypadku oraz przegląd literatury

Conjoined twins are a unique type of monozygotic twins. All monozygotic twins should be thoroughly evaluated for incomplete separation and, if incomplete separation is diagnosed, the extent of internal organ connection and the presence of additional developmental anomalies of the foetuses should be assessed. Common heart of foetuses is particularly difficult to diagnose and crucial for prognosis. We present an example of female thoracoomohalopagus twins with a common triventricular heart, connate livers, and joined hepatic vessels, diagnosed in week 12 of pregnancy. Due to the high complexity of foetal connection, separation was not possible and following interdisciplinary consultation, the pregnancy was aborted upon the patient’s request in week 16.Bliźnięta nierozdzielone stanowią unikalny typ bliźniąt monozygotycznych. Wszystkie bliźnięta monozygotyczne winny być wnikliwie ocenione w kontekście ich niecałkowitego rozdzielenia, a w sytuacji postawienia takiej diagnozy należy ustalić rozległość zespolenia narządów wewnętrznych oraz obecność dodatkowych anomalii rozwoju płodów. Szczególnie trudną diagnostycznie oraz kluczową w kategoriach rokowania jest sytuacja wystąpienia wspólnego serca płodów. Prezentujemy przypadek bliźniąt płci żeńskiej typu thoracoomphalopagus, ze wspólnym, trójkomorowym sercem, zrośniętymi wątrobami oraz połączonymi naczyniami wątrobowymi rozpoznany w 12 tygodniu ciąży. Z powodu wysokiego stopnia złożoności połączenia płodów operacja rozdzielnia bliźniąt nie była możliwa i po interdyscyplinarnej konsultacji ciąża została na prośbę pacjentki zakończona terminacją w 16 tygodniu

Prenatal diagnosis of Down syndrome in dizygotic twin pregnancy

We present a case of a 33-year-old pregnant woman who had a transvaginal ultrasound performed at week 9 of gestation. A dichorionic diamniotic twin pregnancy, with symmetrically developing fetuses, was confirmed. Chromosomal defect markers (NT, NB, DV, TV) were analyzed in the first genetic test, performed according to the Fetal Medicine Foundation (FMF) criteria, and the double marker test was performed (PAPP-A protein and free beta-hCG concentrations in patient serum were determined). In the subsequent diagnostic procedures, the patient was offered and consented to amniopuncture after week 15 of gestation. The material obtained in the course of that invasive procedure allowed to identify a normal male karyotype – 46, XY in the first fetus (Fetus I). Cytogenetic analysis of the material from the second fetus (Fetus II) resulted in the diagnosis of an abnormal female karyotype – 47, XX, +21. Based on the analyzed clinical case, we present the difficulties of performing prenatal diagnosis in a dizygotic twin pregnancy. The results prove the applicability and efficacy of prenatal diagnostics tests based on the FMF criteria also in twin pregnancies

The Application of Real-Time PCR Technique to Detect Rare Cell Clones with Primary T790M Substitution of EGFR Gene in Metastases of Non-small Cell Lung Cancer to Central Nervous System in Chemotherapy Naive Patients

The time-limited efficacy of reversible EGFR-TKIs in patients with advanced non-small cell lung cancer (NSCLC) with EGFR gene activating mutations is associated with development of treatment resistance after some period of therapy. This resistance predominantly results from secondary mutations located in EGFR gene, especially T790M substitution. There is limited information available concerning the prevalence of primary T790M mutations in patients with metastatic NSCLC tumors before treatment with EGFR-TKIs. The aim of work was to assess the prevalence of de novo T790M mutations in EGFR gene in tissue samples from NSCLC metastatases in central nervous system (CNS) in both chemotherapy and EGFR-TKI naive NSCLC patients. We analyzed DNA samples isolated from paraffin-embedded tissue from CNS metastases for T790M mutations using real-time PCR and TaqMan probe against the T790M mutant sequence. The tissue samples were taken during palliative neurosurgery in 143 NSCLC patients. Amplification of the T790M-specific sequence was detected in 25 patients (17.5 %). The quantity of mutated DNA was less than 1 % in all samples with amplification, and in vast majority (20 patients, 14 % of all samples) it was even less that 0.1 %. In 5 patients (3.5 %) quantity of mutated DNA ranged from 0.1 to 1 % and true positive results of T790M mutation presence in these patients were most possible. Amplification of this sequence was not concurrent with common EGFR mutations and was not associated with sex, smoking status and pathological type of cancer. There is a possibility to detect the primary T790M mutation in brain metastases of NSCLC in EGFR-TKIs naïve patients
