    Upconversion nanocrystal doped polymer fiber thermometer

    In recent years, lanthanide-doped nanothermometers have been mainly used in thin films or dispersed in organic solvents. However, both approaches have disadvantages such as the short interaction lengths of the active material with the pump beam or complicated handling, which can directly affect the achievable temperature resolution. We investigated the usability of a polymer fiber doped with upconversion nanocrystals as a thermometer. The fiber was excited with a wavelength stabilized diode laser at a wavelength of 976 nm. Emission spectra were recorded in a temperature range from 10 to 35◦C and the thermal emission changes were measured. Additionally, the pump power was varied to study the effect of self-induced heating on the thermometer specifications. Our fiber sensor shows a maximal thermal sensitivity of 1.45%/K and the minimal thermal resolution is below 20 mK. These results demonstrate that polymer fibers doped with nanocrystals constitute an attractive alternative to conventional fluorescence thermometers, as they add a long pump interaction length while also being insensitive to strong electrical fields or inert to bio-chemical environments. © 2020 by the authors.     Towards highly efficient polymer fiber laser sources for integrated photonic sensors

    Lab-on-a-Chip (LoC) devices combining microfluidic analyte provision with integrated optical analysis are highly desirable for several applications in biological or medical sciences. While the microfluidic approach is already broadly addressed, some work needs to be done regarding the integrated optics, especially provision of highly integrable laser sources. Polymer optical fiber (POF) lasers represent an alignment-free, rugged, and flexible technology platform. Additionally, POFs are intrinsically compatible to polymer microfluidic devices. Home-made Rhodamine B (RB)-doped POFs were characterized with experimental and numerical parameter studies on their lasing potential. High output energies of 1.65 mJ, high slope efficiencies of 56%, and 50%-lifetimes of ≥900 k shots were extracted from RB:POFs. Furthermore, RB:POFs show broad spectral tunability over several tens of nanometers. A route to optimize polymer fiber lasers is revealed, providing functionality for a broad range of LoC devices. Spectral tunability, high efficiencies, and output energies enable a broad field of LoC applications. © 2020 by the authors. Licensee MDPI, Basel, Switzerland

    Profiling Microglia From Alzheimer's Disease Donors and Non-demented Elderly in Acute Human Postmortem Cortical Tissue

    Microglia are the tissue-resident macrophages of the central nervous system (CNS). Recent studies based on bulk and single-cell RNA sequencing in mice indicate high relevance of microglia with respect to risk genes and neuro-inflammation in Alzheimer's disease (AD). Here, we investigated microglia transcriptomes at bulk and single-cell levels in non-demented elderly and AD donors using acute human postmortem cortical brain samples. We identified seven human microglial subpopulations with heterogeneity in gene expression. Notably, gene expression profiles and subcluster composition of microglia did not differ between AD donors and non-demented elderly in bulk RNA sequencing nor in single-cell sequencing

    Limited Plasmodium sporozoite gliding motility in the absence of TRAP family adhesins

    Background!#!Plasmodium sporozoites are the highly motile forms of malaria-causing parasites that are transmitted by the mosquito to the vertebrate host. Sporozoites need to enter and cross several cellular and tissue barriers for which they employ a set of surface proteins. Three of these proteins are members of the thrombospondin related anonymous protein (TRAP) family. Here, potential additive, synergistic or antagonistic roles of these adhesion proteins were investigated.!##!Methods!#!Four transgenic Plasmodium berghei parasite lines that lacked two or all three of the TRAP family adhesins TRAP, TLP and TREP were generated using positive-negative selection. The parasite lines were investigated for their capacity to attach to and move on glass, their ability to egress from oocysts and their capacity to enter mosquito salivary glands. One strain was in addition interrogated for its capacity to infect mice.!##!Results!#!The major phenotype of the TRAP single gene deletion dominates additional gene deletion phenotypes. All parasite lines including the one lacking all three proteins were able to conduct some form of active, if unproductive movement.!##!Conclusions!#!The individual TRAP-family adhesins appear to play functionally distinct roles during motility and infection. Other proteins must contribute to substrate adhesion and gliding motility

    Information, possible worlds and the cooptation of scepticism

Its main goal is to articulate and defend the view that either informational scepticism is radical, but then it is epistemologically innocuous because redundant; or it is moderate, but then epistemologically beneficial because useful. In order to pursue this cooptation strategy, the article is divided into seven sections. Section 1 sets up the problem. Section 2 introduces Borel numbers as a convenient way to refer uniformly to (the data that individuate) different possible worlds. Section 3 adopts the Hamming distance between Borel numbers as a metric to calculate the distance between possible worlds. In Sects. 4 and 5, radical and moderate informational scepticism are analysed using Borel numbers and Hamming distances, and shown to be either harmless (extreme form) or actually fruitful (moderate form). Section 6 further clarifies the approach by replying to some potential objections. In the conclusion, the Peircean nature of the overall approach is briefly discussed.Peer reviewe
