    Nowadays, theoretical and applied research is devoted to various problems education. The issues of challenging education are highlighted at regional and national levels. The Government of Russian Federation focuses on raising the general quality of training. Design-education is a new pedagogical area that attracts attention by means of national promotion in direction of system modernization of socio-cultural and economic areas. Theoretical and methodological basis of research combines proceedings of national researchers that cover professional activity of designers and reveal the specific features of design-education. The author refers to the modern research and legal acts that regulate professional activity of designers. The article focuses on such methods of scientific knowledge as analysis, generalization, comparative analysis and forecasting. The research is aimed at investigation of possibilities to raise the level of expertise and responsibility of designer by means of specific legal knowledge. In order to achieve this goal, the author has analyzed the main problems of design-training and revealed the outlooks of educational process and conditions that contribute to students’ achievement of high professional level. The paper outlines the necessity to develop legal culture by means of introducing special curriculum and development of specialized textbook. These measures conform to the modern requirements; they are directed to training of highly-qualified designers who obtain sufficient legal knowledge and able to compete in professional environment. . to modern requirements and aims to prepare highly specialist designers with a sufficient level of legal knowledge, the ability to successfully exist in a professional environment.Сегодня множество теоретических и прикладных исследований посвящено самым различным образовательным проблемам. Вопросы перспективного образования поднимаются на самом высоком уровне. Правительство Российской Федерации ставит во главу угла повышение общего качества подготовки специалистов. Дизайн-образование – относительно новая педагогическая область, внимание к которой усиливается по мере продвижения страны в направлении системной модернизации социокультурной и экономической сфер. Теоретико-методологическую основу исследования составили многочисленные труды отечественных авторов, всесторонне охватывающие профессиональную деятельность дизайнеров, раскрывающие специфику дизайн-образования. Автор также ссылается на современные исследования, правовые документы и законодательные акты, регламентирующие профессиональную деятельность дизайнеров. Из применяемых методов научного познания стоит выделить: анализ, обобщение, сравнение данных и прогнозирование. Целью исследования явилось изучение возможностей повышения уровня компетентности и ответственности профессионального дизайнера за счет получения специфических правовых знаний. Для достижения цели проведен анализ основных проблем дизайн-образования, выявлены перспективы совершенствования образовательного процесса, условия достижения студентами-дизайнерами высокого профессионального уровня. Подтверждена необходимость повышения уровня знаний в сфере законодательной базы за счет внедрения специальной учебной программы и создания специализированного учебного пособия. Предложенный комплекс мер отвечает современным требованиям и направлен на  подготовку высокоэффективных специалистов- дизайнеров с достаточным уровнем правовых знаний, способных успешно существовать в профессиональной среде

    Human superoxide dismutase 1 (hSOD1) maturation through interaction with human copper chaperone for SOD1 (hCCS).

    Copper chaperone for superoxide dismutase 1 (SOD1), CCS, is the physiological partner for the complex mechanism of SOD1 maturation. We report an in vitro model for human CCS-dependent SOD1 maturation based on the study of the interactions of human SOD1 (hSOD1) with full-length WT human CCS (hCCS), as well as with hCCS mutants and various truncated constructs comprising one or two of the protein’s three domains. The synergy between electrospray ionization mass spectrometry (ESI-MS) and NMR is fully exploited. This is an in vitro study of this process at the molecular level. Domain 1 of hCCS is necessary to load hSOD1 with Cu(I), requiring the heterodimeric complex formation with hSOD1 fostered by the interaction with domain 2. Domain 3 is responsible for the catalytic formation of the hSOD1 Cys-57–Cys-146 disulfide bond, which involves both hCCS Cys-244 and Cys-246 via disulfide transfer

    Thermonuclear explosions as Type II supernovae

    We consider a binary stellar system, in which a low-mass, of 0.6 Msun, carbon-oxygen white dwarf (WD) mergers with a degenerate helium core of 0.4 Msun of a red giant. We analyse the outcome of a merger within a common envelope (CE). We predict the observational properties of the resulting transient. We find that the double detonation of the WD, being a pure thermonuclear explosion and embedded into the hydrogen-rich CE, has a light curve with the distinct plateau shape, i.e. looks like a supernova (SN) Type IIP, with a duration of about 40 days. We find five observed SNe IIP: SN 2004dy, SN 2005af, SN 2005hd, SN 2007aa, and SN 2008bu, that match the V-band light curve of our models. Hence, we show that a thermonuclear explosion within a CE might be mistakenly identified as a SN IIP, which are believed to be an outcome of a core-collapse neutrino-driven explosion of a massive star.     Influence of VEGF deprivation upon vascular formation by endothelium in the presence of macrophages

    Development of angiogenesis depends on the functional state of endothelial cells, as well as on the balanced secretion of cytokines, growth factors and chemokines by endothelial cells and cells of microenvironment. Macrophages represent an essential component of the microenvironment and take part in the formation of blood vessels both due to the production of cytokines and due to contact interactions with endothelial cells. VEGF is among the most important cytokines that control angiogenesis at all its stages. Currently, the role of VEGF in the intercellular interactions of endothelial cells and macrophages is not well described. The aim of our study was to investigate the effect of VEGF deprivation using monoclonal antibodies on angiogenesis under conditions of co-cultivation of endothelium and macrophages. Materials and methods: monoclonal antibodies to VEGF-A were used for VEGF deprivation in monoculture of endothelial cells and in co-culture of endothelial cells with macrophages. The IL-1β, IL-6 and TNFα cytokines were used as inducers. When VEGF-A was removed from the medium, endothelial cells show plasticity and form longer vessels, they modify the expression of VEGF receptors. Macrophages regulate endothelial cell activity through the secretion of cytokines, including VEGF, and through contact interactions with endothelial cells. THP-1 cells increase the sensitivity of endothelial cells to VEGF by stimulating the VEGFR1 and VEGFR3 expression, this effect is VEGF-A-independent. The IL-1β, IL-6, TNFa cytokines independently stimulate non-branching angiogenesis, increasing the length of the vessels. At the same time, IL-ip increases the VEGFR1 expression on the surface of endothelial cells. In contrast, IL-6 and TNFα decrease it, thereby regulating the sensitivity of endothelial cells to VEGF. The effects of these cytokines are not dependent on VEGF-A. The IL-1β, IL-6, TNFα cytokines promote acquisition of anti-angiogenic properties by THP-1 cells that is independent on VEGF-A, as well as on expression of its receptors by endothelial cells. Thus, VEGF is an important, but not the sole factor controlling angiogenesis. Under conditions of VEGF-A deficiency, either endothelial cells or microenvironment cells are able to compensate for its functional load due to the production of other growth factors

    Unprecedented early flux excess in the hybrid 02es-like type Ia supernova 2022ywc indicates interaction with circumstellar material

    We present optical photometric and spectroscopic observations of the 02es-like type Ia supernova (SN) 2022ywc. The transient occurred in the outskirts of an elliptical host galaxy and showed a striking double-peaked light curve with an early excess feature detected in the ATLAS orange and cyan bands. The early excess is remarkably luminous with an absolute magnitude 19\sim -19, comparable in luminosity to the subsequent radioactively-driven second peak. The spectra resemble the hybrid 02es-like SN 2016jhr, that is considered to be a helium shell detonation candidate. We investigate different physical mechanisms that could power such a prominent early excess and rule out massive helium shell detonation, surface 56^{56}Ni distribution and ejecta-companion interaction. We conclude that SN ejecta interacting with circumstellar material (CSM) is the most viable scenario. Semi-analytical modelling with MOSFiT indicates that SN ejecta interacting with 0.05\sim 0.05\,M_{\odot} of CSM at a distance of 1014\sim 10^{14} cm can explain the extraordinary light curve. A double-degenerate scenario may explain the origin of the CSM, either by tidally-stripped material from the secondary white dwarf, or disk-originated matter launched along polar axes following the disruption and accretion of the secondary white dwarf. A non-spherical CSM configuration could suggest that a small fraction of 02es-like events viewed along a favourable line of sight may be expected to display a very conspicuous early excess like SN 2022ywc.Comment: Accepted to ApJL after minor revisio

    PHENOTYPIC AND FUNCTIONAL CHARACTERISTICS OF MICROVESICLES PRODUCED BY NATURAL KILLER CELLS

    Natural killer (NK) cells are of special interest among a multitude of microvesicle (MV) source cells. NK cells are a lymphocyte subpopulation performing contact cytolysis of virus-infected cells and tumor cells. Each of the NK cell populations has a unique receptor repertoire on its surface and, thus, unique functions. During their contact with a target cell, the most common mechanism of cytolysis is an exocytosis of lytic granules. However, some indirect evidence suggests that MV with CD56 phenotype and leukocyte-derived MV with various phenotypes are present in the peripheral blood plasma.This research is aimed to study the phenotype, composition and cytotoxic activity of microvesicles produced by NK cells. The analysis of receptor expression showed that MV, as well as source cells of the NK-92 cell line, had a similar CD56 molecule expression profile. The expression profile in MV differs from the same in source cells by higher CD119 and CD11b expression and by lower CD18 expression. Culturing of NK-92 cells in the presence of PMA, IL-1β, TNFα, IFNγ resulted in alterations of cell phenotypes and MV. Immunoblots revealed a change of perforin and granzyme B (GrB) in MV. The analysis of the cytotoxic activity of NK-92 cells in a natural killer in vitro assay employing K562 target cells demonstrated that MV obtained from TNFα-activated cells of the NK-92 cell line increased the cytotoxicity of the same TNFα-activated NK-92 cells regarding cytotoxicity levels. This coincides with the previously revealed increased content of GrB in MV obtained from TNFα-activated cells of the NK-92 cell line. To sum up depending on the cytokine NK-92 cells produce MV that differ in their phenotype, composition and activity. Any changes in MV composition can result in changes in their functional activity: in particular, changes can increase the cytotoxic activity of NK cells of the NK-92 cell line. Thus, besides a well-known and proved way for GrB delivery to a target cell, we can suggest an additional way – the transportation of GrB within MV

While a sharp luminosity threshold has been suggested, there remains an increasing number of transitional objects that reach this threshold without the spectroscopic signatures common to SLSNe. In this work, we present data and analysis on four SNe transitional between SE-SNe and SLSNe; the He-poor SNe 2019dwa and 2019cri, and the He-rich SNe 2019hge and 2019unb. Each object displays long-lived and variable photometric evolution with luminosities around the SLSN threshold of Mr < -19.8 mag.
