    RATIONALE: Privileged episodic encoding of an aversive event often comes at a cost of neutral events flanking the aversive event, resulting in decreased episodic memory for these neutral events. This peri-emotional amnesia is amygdala-dependent and varies as a function of norepinephrine activity. However, less is known about the amnesiogenic potential of cortisol. OBJECTIVE: We used a strategy of pharmacologically potentiating cortisol and norepinephrine activity to probe the putative neurochemical substrates of peri-emotional amnesia. MATERIALS AND METHODS: Fifty-four healthy individuals participated in a randomized double-blind placebo-controlled study. Within the experimental context of an established peri-emotional amnesia paradigm, we tested the amnesiogenic potential of hydrocortisone (30 mg p.o.) in the presence or absence of the norepinephrine-reuptake inhibitor reboxetine (4 mg p.o.). RESULTS: Under dual challenge conditions, we observed a linear dose-response relationship in the magnitude and duration of emotion-induced retrograde amnesia. CONCLUSIONS: Our results are consistent with a phenotypic expression of retrograde amnesia varying as a function of norepinephrine and cortisol coactivation during episodic encoding of aversive events. Our study demonstrates that the adverse cognitive and behavioral sequelae of aversive emotion can be experimentally modeled by a pharmacological manipulation of its putative neurochemical substrates

    The effect of the spatial positioning of items on the reliability of questionnaires measuring affect

    Orientation: Extant research has shown that the relationship between spatial location and affect may have pervasive effects on evaluation. In particular, experimental findings on embodied cognition indicate that a person is spatially orientated to position what is positive at the top and what is negative at the bottom (vertical spatial orientation), and to a lesser extent, to position what is positive on the left and what is negative on the right (horizontal spatial orientation). It is therefore hypothesised, that when there is congruence between a respondent’s spatial orientation (related to affect) and the spatial positioning (layout) of a questionnaire, the reliability will be higher than in the case of incongruence. Research purpose: The principal objective of the two studies reported here was to ascertain the extent to which congruence between a respondent’s spatial orientation (related to affect) and the layout of the questionnaire (spatial positioning of questionnaire items) may impact on the reliability of a questionnaire measuring affect. Motivation for the study: The spatial position of items on a questionnaire measuring affect may indirectly impact on the reliability of the questionnaire. Research approach, design and method: In both studies, a controlled experimental research design was conducted using a sample of university students (n = 1825). Major findings: In both experiments, evidence was found to support the hypothesis that greater congruence between a respondent’s spatial orientation (related to affect) and the spatial positioning (layout) of a questionnaire leads to higher reliability on a questionnaire measuring affect. Practical implications: These findings may serve to create awareness of the influence of the spatial positioning of items as a confounding variable in questionnaire design. Contribution/value-add: Overall, this research complements previous studies by confirming the metaphorical representation of affect and enhances our understanding of embodiment-related conceptual processing and its subsequent influence on self-evaluations versus external evaluations on an unconscious level, specifically in relation to measuring affect

    New Generation of Instrumented Ranges: Enabling Automated Performance Analysis

    Military training conducted on physical ranges that match a unit’s future operational environment provides an invaluable experience. Today, to conduct a training exercise while ensuring a unit’s performance is closely observed, evaluated, and reported on in an After Action Review, the unit requires a number of instructors to accompany the different elements. Training organized on ranges for urban warfighting brings an additional level of complexity—the high level of occlusion typical for these environments multiplies the number of evaluators needed. While the units have great need for such training opportunities, they may not have the necessary human resources to conduct them successfully. In this paper we report on our US Navy/ONR-sponsored project aimed at a new generation of instrumented ranges, and the early results we have achieved. We suggest a radically different concept: instead of recording multiple video streams that need to be reviewed and evaluated by a number of instructors, our system will focus on capturing dynamic individual warfighter pose data and performing automated performance evaluation. We will use an in situ network of automatically-controlled pan-tilt-zoom video cameras and personal position and orientation sensing devices. Our system will record video, reconstruct dynamic 3D individual poses, analyze, recognize events, evaluate performances, generate reports, provide real-time free exploration of recorded data, and even allow the user to generate ‘what-if’ scenarios that were never recorded. The most direct benefit for an individual unit will be the ability to conduct training with fewer human resources, while having a more quantitative account of their performance (dispersion across the terrain, ‘weapon flagging’ incidents, number of patrols conducted). The instructors will have immediate feedback on some elements of the unit’s performance. Having data sets for multiple units will enable historical trend analysis, thus providing new insights and benefits for the entire service.Office of Naval Researc

    Keratin 16 (K16) is a cytoskeletal scaffolding protein highly expressed at pressure-bearing sites of the mammalian footpad. It can be induced in hyperproliferative states such as wound healing, inflammation and cancer. Here we show that the inactive rhomboid protease RHBDF2 (iRHOM2) regulates thickening of the footpad epidermis through its interaction with K16. K16 expression is absent in the thinned footpads of irhom2-/- mice compared with irhom2+/+mice, due to reduced keratinocyte proliferation. Gain-of-function mutations in iRHOM2 underlie Tylosis with oesophageal cancer (TOC), characterized by palmoplantar thickening, upregulate K16 with robust downregulation of its type II keratin binding partner, K6. By orchestrating the remodelling and turnover of K16, and uncoupling it from K6, iRHOM2 regulates the epithelial response to physical stress. These findings contribute to our understanding of the molecular mechanisms underlying hyperproliferation of the palmoplantar epidermis in both physiological and disease states, and how this 'stress' keratin is regulated

    Polymorphisms of Glutathione S-transferases Omega-1 among ethnic populations in China

    <p>Abstract</p> <p>Background</p> <p>Glutathione S-transferases (GSTs) is a genetic factor for many diseases and exhibits great diversities among various populations. We assessed association of the genotypes of Glutathione S-transferases Omega-1 (GSTO1) A140D with ethnicity in China.</p> <p>Results</p> <p>Peripheral blood samples were obtained from 1314 individuals from 14 ethnic groups. Polymorphisms of GSTO1 A140D were measured using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Logistic regression was employed to adjustment for regional factor. The frequency of GSTO1 140A allele was 15.49% in the total 14 ethnic populations. Compared to Han ethnic group, two ethnic populations were more likely to have AA or CA genotype [odds ratio (OR): 1.77, 95% confidence interval (95% CI): 1.05–2.98 for Uygur and OR: 1.78, 95% CI: 1.18–2.69 for Hui]. However, there were no statistically significant differences across 14 ethnic groups when region factor was adjusted. In Han ethnicity, region was significantly associated with AA or CA genotype. Han individuals who resided in North-west of China were more likely to have these genotypes than those in South of China (OR: 1.63, 95% CI: 1.21–2.20).</p> <p>Conclusion</p> <p>The prevalence of the GSTO1 140A varied significantly among different regional populations in China, which showed that geography played a more important role in the population differentiation for this allele than the ethnicity/race.</p

    Plasma 24S-hydroxycholesterol levels in elderly subjects with late onset Alzheimer's disease or vascular dementia: a case-control study

    <p>Abstract</p> <p>Background</p> <p>In central nervous system cholesterol cannot be degraded but is secreted into circulation predominantly in the form of its polar metabolite 24(<it>S</it>)-hydroxycholesterol (24S-OH-Chol). Some studies suggested an association between 24S-OH-Chol metabolism and different neurological diseases including dementia. A possible decrease in 24S-OH-Chol plasma levels has been reported late onset Alzheimer's disease (LOAD) and vascular dementia (VD), but results of previous studies are partially contradictory.</p> <p>Methods</p> <p>By high-speed liquid chromatography/tandem mass spectrometry we evaluated the plasma levels of 24S-OH-Chol in a sample of 160 older individuals: 60 patients with LOAD, 35 patients with VD, 25 subjects affected by cognitive impairment no-dementia (CIND), and 40 (144 for genetics study) cognitively normal Controls. We also investigated the possible association between PPARgamma Pro12Ala polymorphism and dementia or 24S-OH-Chol levels.</p> <p>Results</p> <p>Compared with Controls, plasma 24S-OH-Chol levels were higher in LOAD and lower in VD; a slight not-significant increase in CIND was observed (ANOVA p: 0.001). A positive correlation between 24S-OH-Chol/TC ratio and plasma C reactive protein (CRP) levels was found in the whole sample, independent of possible confounders (multiple regression p: 0.04; r<sup>2</sup>: 0.10). This correlation was strong in LOAD (r: 0.39), still present in CIND (r: 0.20), but was absent in VD patients (r: 0.08). The PPARgamma Pro12Ala polymorphism was not associated with the diagnosis of LOAD, VD, or CIND; no correlation emerged between the Ala allele and 24S-OH-Chol plasma levels.</p> <p>Conclusions</p> <p>Our results suggest that plasma 24S-OH-Chol levels might be increased in the first stages of LOAD, and this phenomenon might be related with systemic inflammation. The finding of lower 24S-OH-Chol concentrations in VD might be related with a more advanced stage of VD compared with LOAD in our sample, and/or to different pathogenetic mechanisms and evolution of these two forms of dementia.</p

    Image Feature Extraction Acceleration

    Image feature extraction is instrumental for most of the best-performing algorithms in computer vision. However, it is also expensive in terms of computational and memory resources for embedded systems due to the need of dealing with individual pixels at the earliest processing levels. In this regard, conventional system architectures do not take advantage of potential exploitation of parallelism and distributed memory from the very beginning of the processing chain. Raw pixel values provided by the front-end image sensor are squeezed into a high-speed interface with the rest of system components. Only then, after deserializing this massive dataflow, parallelism, if any, is exploited. This chapter introduces a rather different approach from an architectural point of view. We present two Application-Specific Integrated Circuits (ASICs) where the 2-D array of photo-sensitive devices featured by regular imagers is combined with distributed memory supporting concurrent processing. Custom circuitry is added per pixel in order to accelerate image feature extraction right at the focal plane. Specifically, the proposed sensing-processing chips aim at the acceleration of two flagships algorithms within the computer vision community: the Viola-Jones face detection algorithm and the Scale Invariant Feature Transform (SIFT).
