    Deregulation of manganese superoxide dismutase (SOD2) expression and lymph node metastasis in tongue squamous cell carcinoma

    <p>Abstract</p> <p>Background</p> <p>Lymph node metastasis is a critical event in the progression of tongue squamous cell carcinoma (TSCC). The identification of biomarkers associated with the metastatic process would provide critical prognostic information to facilitate clinical decision making. Previous studies showed that deregulation of manganese superoxide dismutase (SOD2) expression is a frequent event in TSCC and may be associated with enhanced cell invasion. The purpose of this study is to further evaluate whether the expression level of SOD2 is correlated with the metastatic status in TSCC patients.</p> <p>Methods</p> <p>We first examined the SOD2 expression at mRNA level on 53 TSCC and 22 normal control samples based on pooled-analysis of existing microarray datasets. To confirm our observations, we examined the expression of SOD2 at protein level on an additional TSCC patient cohort (n = 100), as well as 31 premalignant dysplasias, 15 normal tongue mucosa, and 32 lymph node metastatic diseases by immunohistochemistry (IHC).</p> <p>Results</p> <p>The SOD2 mRNA level in primary TSCC tissue is reversely correlated with lymph node metastasis in the first TSCC patient cohort. The SOD2 protein level in primary TSCC tissue is also reversely correlated with lymph node metastasis in the second TSCC patient cohort. Deregulation of SOD2 expression is a common event in TSCC and appears to be associated with disease progression. Statistical analysis revealed that the reduced SOD2 expression in primary tumor tissue is associated with lymph node metastasis in both TSCC patient cohorts examined.</p> <p>Conclusions</p> <p>Our study suggested that the deregulation of SOD2 in TSCC has potential predictive values for lymph node metastasis, and may serve as a therapeutic target for patients at risk of metastasis.</p

    Incidence of transient interruption of contrast (TIC) – A retrospective single-centre analysis in CT pulmonary angiography exams acquired during inspiratory breath-hold with the breathing command: “Please inspire gently!”

    Objectives To assess the occurrence of transient interruption of contrast (TIC) phenomenon in pulmonary computed tomography angiography (CTPA) exams performed in inspiratory breath-hold after patients were told to inspire gently. Methods In this retrospective single-centre study, CTPA exams of 225 consecutive patients scanned on a 16-slice CT scanner system were analysed. A-priori to measurements, exams were screened for inadequate pulmonary artery contrast due to incorrect bolus tracking or failure of i.v. contrast administration. Those exams were excluded. Attenuation values in the thoracic aorta and in the pulmonary trunk were assessed in duplicate measurements (M1 and M2) and the aorto-pulmonary density ratio was calculated. An aorto-pulmonary ratio > 1 with still contrast inflow being visible within the superior vena cava was defined as TIC. Results 3 patients were excluded due to incorrect bolus tracking. Final analysis was performed in 222 patients (mean age 65 ± 19 years, range 18 to 99 years). Mean density in the pulmonary trunk was 275±17 HU, in the aorta 208 ± 15 HU. Mean aorto-pulmonary ratio was 0.81± 0.29. 48 patients (21.6%) had an aorto-pulmonary ratio >1. Correlation of mean aorto-pulmonary ratio and age was: -0.213 (p = 0.001). Age was not significantly different for an aorto-pulmonary ratio >1 vs. ≤1 (p = 0.122). Both in M1 and M2, 33/222 patients presented with absolute HU values of < 200 HU within the pulmonary artery. In M1 measurements, 24 of these 33 patients (72%) fulfilled TIC criteria (M2: 25/33 patients (75%)). Conclusions TIC is a common phenomenon in CTPA studies with inspiratory breath-hold commands after patients were told to inspire gently with an incidence of 22% in our retrospective cohort. Occurrence of TIC shows a significant negative correlation with increasing age and disproportionately often occurs in patients with lower absolute contrast density values within their pulmonary arteries.ISSN:1932-620

    A coupled subsample displacement estimation method for ultrasound-based strain elastography

    Obtaining accurate displacement estimates along both axial (parallel to the acoustic beam) and lateral (perpendicular to the beam) directions is an important task for several clinical applications such as shear strain imaging, modulus reconstruction and temperature imaging, where a full description of the two or three dimensional (2D/3D) deformation field is required. In this study we propose an improved speckle tracking algorithm where axial and lateral motion estimations are simultaneously performed to enhance motion tracking accuracy. More specifically, using conventional ultrasound echo data, this algorithm first finds an iso-contour in the vicinity of the peak correlation between two segments of the pre- and post-deformation ultrasound radiofrequency echo data. The algorithm then attempts to find the center of the iso-contour of the correlation function that corresponds to the unknown (sub-sample) motion vector between these two segments of echo data. This algorithm has been tested using computer-simulated data, studies with a tissue-mimicking phantom, and in vivo breast lesion data. Computer simulation results show that the method improves the accuracy of both lateral and axial tracking. Such improvements are more significant when the deformation is small or along the lateral direction. Results from the tissue-mimicking phantom study are consistent with findings observed in computer simulations. Using in vivo breast lesion data we found that, compared to the 2D quadratic subsample displacement estimation methods, higher quality axial strain and shear strain images (e.g. 18.6% improvement in contrast-to-noise ratio for shear strain images) can be obtained for large deformations (up to 5% frame-to-frame and 15% local strains) in a multi-compression technique. Our initial results demonstrated that this conceptually and computationally simple method could improve the image quality of ultrasound-based strain elastography (SE) with current clinical equipment
